Polar cell growth is regulated by auxin (Klämbt et al., 1992). It is essentially dependent upon exocytosis. Is exocytosis directly regulated by auxin? Aleurone cells secrete amylases, as well as other hydrolases, by exocytosis. In preliminary experiments, with 22 h of incubation, increasing concentration of N-1-naphthylphthalamic acid (NPA) parallels the inhibition of amylase secretion. A model system for secretion was designed. Barley aleurone layers, previously incubated in 1 μM GA3 for 16 h, then washed extensively in the presence of 10 μM NPA for 10 min, were incubated finally for 2 h in GA3, NPA and/or auxins. NPA inhibits amylase secretion. Auxins completely abolished secretion inhibition at the optimal concentration of 1 μM. Aleurone layers contain 1.5 to 3 μM endogenous indole-3-acetic acid (IAA) and have a high capacity to accumulate and retain IAA. A 7-fold accumulation occurred within 3 h after application of 20 μM radiolabeled IAA. A loss of about 10% could be measured within the following 4 h period in fresh media, independent of the presence of NPA. The inhibition of amylase secretion by NPA is not due to an inhibition of protein biosynthesis which was tested by the use of [35S]-methionine.  相似文献

Turkana tribespeople reside in a semi-arid savanna ecosystem in northwest Kenya. For over a decade, Ngisonyoka Turkana nomads have been studied within a multidisciplinary framework that embraces ecology, anthropology, and human population biology. Original research objectives of the South Turkana Ecosystem Project were to study nomads longitudinally and within the context of the dry savanna ecosystem. These objectives have been expanded to incorporate settled Turkana who were nomads in the recent past, but who, for a variety of reasons, have taken up a life of sedentary cultivation. The research described here focused on comparisons of growth patterns of nomadic and settled infants from birth to 24 months of age. Infants were measured (recumbent length, weight, head circumference, arm and calf circumference, and selected skinfolds) in 1989 and 1990. Settled infants were slightly longer, but nomadic infants were heavier and fatter. Head circumferences were the same. Some variation was observed by season and year of measurement (from comparative surveys). Both groups showed pronounced weight faltering after 6 months of age when compared with U. S. NCHS reference values. Head circumference faltering was moderate and recumbent length faltering was only slight after 12 months of age. Comparisons of Turkana infant growth in the two populations can contribute indirectly to a better understanding of infant nutritional status, probable morbidity, and other factors, some of which may be linked to female reproduction. © 1993 Wiley-Liss, Inc.  相似文献

North China and South Siberia, populated by Altaic- and Sino-Tibetan-speaking populations, possess extensive ethnolinguistic diversity and serve as the crossroads for the initial peopling of America and western–eastern transcontinental communication. However, the population genetic structure and admixture history of northern East Asians remain poorly understood due to a lack of genome-wide data, especially for Mongolic-speaking people in China. We genotyped genome-wide single nucleotide polymorphisms for 510 individuals from 38 Mongolic, Tungusic, and Sinitic-speaking populations. We first explored the shared alleles and haplotypes within the studied groups. We then merged with 3508 published modern and ancient Eurasian individuals to reconstruct the deep evolutionary and natural selection history of northern East Asians. We identified genetic substructures within Altaic-speaking populations: Western Turkic people harbored more western Eurasian-related ancestry; Northern Mongolic people in Siberia and eastern Tungusic people in Amur River Basin (ARB) possessed a majority of Neolithic ARB related ancestry; Southern Mongolic people in China possessed apparent genetic influence from Neolithic Yellow River Basin (YRB) farmers. Additionally, we found the differentiated admixture history between western and eastern Mongolians and geographically close Northeast Hans: the former received a genetic impact from western Eurasians, and the latter retained the primary Neolithic YRB and ARB ancestry. Moreover, we demonstrated that Kalmyk people from the northern Caucasus Mountains possessed a strong genetic affinity with Neolithic Mongolian Plateau (MP) people, supporting the hypothesis of their eastern Eurasian origin and long-distance migration history. We also illuminated that historical pastoral empires in the MP contributed considerably to the gene pool of northern Mongolic people but rarely to the southern ones. We finally found natural selection signatures in Mongolians associated with alcohol metabolism. Our results demonstrated that the Neolithic ancestral sources from the MP or ARB played an important role in spreading Altaic populations and languages. The observed multisources of genetic diversity contributed significantly to the extensive ethnolinguistic diversity in northern East Asia.  相似文献

The effect of gangliosides upon murine adult B cells at the single precursor cell level was examined using the splenic focus assay. Adult B cells were stimulated in in vitro organ fragment culture by a hapten-modified carrier protein in the presence of an excess of carrier-primed T cells. The addition of a potential tolerogen in the form of antigen coupled to a carrier not previously presented to the T cells resulted in a temporary unresponsiveness of the onset of antibody production in adult B cells, but not a permanent state of tolerance. This delay could be eliminated by the addition of purified murine gangliosides during the presentation of the hapten coupled to the unrecognized carrier. The ganglioside preparation was fractionated by ion-exchange chromatography and the active fraction was found to be a disialoganglioside. These results suggest that the ganglioside may interfere with membrane receptor-related events occurring during or after antigen binding or cross-linking to responding B cells.  相似文献

The DNA of the protozoan Theileria parva, the causal agent of the bovine East Coast Fever, has been prepared at least 99% pure from the intra-erythrocytic form of the parasite. Its buoyant density was found to be 1.696 g/cm3 and its calculated G + C content was 36.7%. Fragmentation of T. parva by the restriction enzyme EcoRI provides some evidence of the presence of repetitive DNA sequences.  相似文献

Characteristics of the intestinal proteins of a detritus-eating aquatic insect, Pteronarcys californica were assessed to determine their possible role as a nutrient source during metamorphosis. Total soluble intestinal protein was significantly correlated to larval weitht, averaging 3.9% of dry weight for both males and females. Proteinase activity in the gut was separated into several bands by electrophoresis. Electrophoresis patterns of total intestinal proteins of P. californica larvae of differing instar, sex, and diet were remarkably similar. Larvae incubated with tritium-labelled amino acids produced labelled intestinal proteins. When early instar larvae were removed from label, there was no significant loss of tritium from the intestine after 2 months. Penultimate instars labelled in the same manner however, showed depuration of tritium from the gut according to first order kinetics. This loss was correlated to the 6–8 wk non-feeding stage of metamorphosis, just prior to adult emergence. These results indicate that proteins are accumulated in the intestinal tract of immature P. californica, then are reabsorbed from the gut during metamorphosis.  相似文献

This paper presents a reassessment of the taxonomic and phylogenetic affinities of the fossil catarrhine primates from the important middle Miocene site of Fort Ternan in Kenya. Although the sample of specimens is rather small, the material can be attributed to at least five different species, identified here asKenyapithecus wickeri, Proconsul sp., a large species of oreopithecid,Simiolus sp., and a small species of catarrhine of indeterminate status.Kenyapithecus wickeri probably represents a conservative sister-taxon of the extant large hominoids. It is more derived than“Sivapithecus” africanus from Maboko Island, from which it can be distinguished at the generic level. A small species of catarrhine from Fort Ternan can be attributed toSimiolus. It is probably a different species fromSimiolus enjiessi from the early Miocene of East Africa, but additional material is needed to confirm its taxonomic distinctiveness. The occurrence of at least five species of catarrhine primates at Fort Ternan confirms that species diversity levels were as high during the middle Miocene as they had been during the early Miocene. However, the overall taxonomic and ecological composition of the middle Miocene catarrhine community was quite different, evidently due to a significant change in the local ecological setting. Taxonomic differences between the catarrhine faunas at Fort Ternan and Maboko Island can probably be explained as a consequence of a chronological separation between the two sites, and, to a lesser degree, to paleoecological differences.  相似文献

本文采用比较的方法,研究了黄海南部东海北部和东海中南部两个鲐鲹鱼索饵场浮游动物生态特征。结果表明,南部渔场生物量的极差、变异系数小于北部渔场。若扣除个别站位的影响,则南部渔场的平均值高于北部渔场。北部渔场群落多样度、均匀度、丰富度、单纯度较高,南部渔场则相反。北部渔场浮游动物优势种较少,且大多数种具有较高的聚集强度;而南部渔场优势种较多,优势度低,聚集性差。  相似文献

Usufruct rights to trees (Ekwar) in the Turkana silvo-pastoral system are an important aspect of natural resource management, particularly in the drier central parts of Kenya. Originating from a participatory forestry extension program, a survey was carried out that showed the extent and duration, often in excess of one generation, of occupancy of a person's Ekwar. Such rights center around the dry season fodder resources, especially of Acacia tortilis. However they are not definite and are linked to risk-spreading by flexibility in livestock management and the need that they be maintained through efficient usage and social linkages. Hitherto, such natural resource management systems have all but been ignored in the development process in favor of the tragedy of the commons paradigm. Likewise, pastoral development has tended to emphasize range and water, while trees are not given the attention they deserve. This endangers the resilience of the system, and it is therefore important that development works with, not against, such environmentally-sound practices to try to make them more sustainable in the long term.  相似文献

We have determined the dose-response of 1,25-dihydroxyvitamin D-3 (1,25-(OH)2D3) on the intracellular free calcium-ion concentration ([Ca2+1]i) in the osteoblastic osteosarcoma cells, ROS 17/2.8, using 19F-NMR and the intracellular divalent cation indicator, 1,2-bis(2-amino-5-fluorephenoxy)ethane-N, N, N′, N′-tetraacetic acid (5F-BAPTA). The dose-response demonstrated an inverted U-shaped relationship with maximal elevation of [Ca2+]i at doses of 1 to 10 nM 1,25-(OH)2D3. At 10 nM, 1,25-(OH)2D3 elevated the [Ca2+]i from a control level of 118±4 nM to a peak value of 237±8 nM within 40 min. 1,25-(OH)2D3 also increased the intial rate of Ca2+ influx into ROs 17/2.8 cells, measured by 45Ca uptake, with a dose-response relationship which paralleled its effects on [Ca2+]i. Treatment of ROS 17/2.8 cells with Pb2+ at 1 and 5 μM significantly increased [Ca2+]i but significantly reduced the 1,25-(OH)2D3-induced elevation of [Ca2+]i. Simultaneous treatment of naive cells with 1,25-(OH)2D3 and Pb2+ produce little reduction of 1,25-(OH)2D3 and Pb2+ produce little reduction of 1,25-(OH)2D3-induced 45Ca uptake while 40 min treatment with Pb2+ before addition of 1,25-(OH)2D3 significantly reduced the 1,25-(OH)2D3-induced increase in 45Ca influx. These findings suggest that Pb2+ acts by inhibiting 1,25-(OH)2D3-activation of Ca2+ channels and interferes with 1,25-(OH)2D3 regulation of Ca2+ metabolism in osteoblastic bone cells.  相似文献
