Regulated movements of the nucleus are essential during zygote formation, cell migrations, and differentiation of neurons. The nucleus moves along microtubules (MTs) and is repositioned on F-actin at the cellular cortex. Two families of nuclear envelope proteins, SUN and KASH, link the nucleus to the actin and MT cytoskeletons during nuclear movements. However, the role of actin nucleators in nuclear migration and positioning is poorly understood. We show that the branched actin nucleator, Arp2/3, affects nuclear movements throughout embryonic and larval development in C. elegans, including nuclear migrations in epidermal cells and neuronal precursors. In one-cell embryos the migration of the male pronucleus to meet the female pronucleus after fertilization requires Arp2/3. Loss of Arp2/3 or its activators changes the dynamics of non-muscle myosin, NMY-2, and alters the cortical accumulation of posterior PAR proteins. Reduced establishment of the posterior microtubule cytoskeleton in Arp2/3 mutants correlates with reduced male pronuclear migration. The UNC-84/SUN nuclear envelope protein that links the nucleus to the MT and actin cytoskeleton is known to regulate later nuclear migrations. We show here it also positions the male pronucleus. These studies demonstrate a global role for Arp2/3 in nuclear migrations. In the C. elegans one-cell embryo Arp2/3 promotes the establishment of anterior/posterior polarity and promotes MT growth that propels the anterior migration of the male pronucleus. In contrast with previous studies emphasizing pulling forces on the male pronucleus, we propose that robust MT nucleation pushes the male pronucleus anteriorly to join the female pronucleus. 相似文献

Rice has emerged as a model monocot for studies in agriculture and biotechnology due to its relatively small genome and a
ready accessibility to plant material. Tissue culture is one of the tools required for genetic transformation and some breeding
programs, and the selection of high-frequency regenerator types is essential for success in these technologies. Thirty-three
rice entries with agricultural and biotechnological characteristics of interest were screened with the aim to identify the
best regenerators. Entries that exhibited between 50% and 90% regeneration frequencies include ‘Taipei-309,’ ‘Super Dwarf,’
‘Norin’ (japonica types), PI 312777, ‘Ali Combo’ (indica types), ‘STG-S,’ and ‘LA3’ (red rice types). One third of the entries
tested were at least two times better at regeneration than the often-cited regenerator ‘Nipponbare.’ Those entries showing
at least 85% frequency of greening or somatic embryo formation at 15 or 30 d on regeneration medium ultimately produced whole
plants after 45 d on regeneration medium at high frequency (at least 40%); those entries not reaching the 85% threshold of
greening by Days 15 or 30 exhibited moderate (15–40%) to low (less than 10%) frequency of whole plant regeneration. This greening
response suggests the means for an early prediction system for identification of useful rice regenerator lines, which would

Summary Theoretical studies on the necessary numbers of components in mixtures (for example multiclonal varieties or mixtures of lines) have been performed according to the relations between the juvenile-mature correlations of mixtures and their number of components. For the juvenile-mature correlation rE based upon the values of the single components (= component means at juvenile and mature ages) and the juvenile-mature correlation rM based upon the means of mixtures of different components we usually will have rM>rE. Furthermore, rM will increase with an increasing number of components in the mixtures. The effectiveness of an early selection will be mainly determined by the magnitude of the juvenile-mature correlation. If we have rM>rE an improvement of early testing can be realized by using mixtures instead of single components. But, what are the necessary numbers of components so that rM will be sufficiently high to enable an effective early selection of mixtures? Some relations between rE and rM can be obtained and conclusions have been derived.The statistical approach significant difference between rE and rM for a given numerical value of rM leads to estimates for the necessary number n of components dependent on rM, , rE and N where: N = total number of components, which are available for the composition of mixtures and = error probability. For different tree species rE can be estimated by an appropriate formula which depends on T with T = time (in years) from planting date until the mature age.Lambeth's formula, for example, has been developed for height growth in pines. For this situation numerical calculations are performed using rM=0.90 and =0.05. The necessary numbers n for T=5, T=10, T=20 and T=50 are: 6, 9, 10 and 12 (for N=50); 13, 17, 20 and 23 (for N=100); 26, 34, 40 and 46 (for N=200); 38, 51, 60 and 69 (for N=300); 64, 85, 100 and 114 (for N=500) and 128, 171, 199 and 228 (for N=1,000). Dmanisi is the oldest site outside of Africa that records unquestioned hominin occupations as well as the dispersal of hominins in Europe and Asia. The site has yielded large numbers of artefacts from several periods of hominin occupation. This analysis of Dmanisi stone tool technology includes a review of all the pieces recovered during the last 15 years of excavations. This lithic assemblage gives insights into the hominin behaviour at 1.7-1.8 Ma in Eurasia. Dmanisi hominins exploited local rocks derived from either nearby riverbeds or outcrops, and petrographic study provides data on patterns of stone procurement. Recent geological surveys and technological studies of the artefacts illustrate the roles of hominins in composing the assemblage. Dmanisi hominins selected two types of blanks, including cobbles and angular blocks, of basalt, andesite, and tuffs. Many complete cobbles, pebbles, and rolled blocks in basalt were unmodified, and geological analyses and surveys indicate that hominins brought manuports back to the site, suggesting a complex procurement strategy. Cores, flakes and debris show that all stages of flaking activity took place at the site. Numerous unifacial cores suggest that knapping was not very elaborate. Centripetal knapping is observed on some flake-cores. Knapping was influenced by the blank shape and natural angles. Most flaked objects were either cores or chopper-cores. Flakes predominate while flake tools are rare. The Dmanisi lithic assemblage is comparable to Oldowan sites in Africa in terms of reduction sequence, organisation of the removals, platform types, and the lack of retouched flakes. Tumour necrosis factor alpha (TNF-alpha), a pro-inflammatory cytokine may play an active role in stimulating inflammatory reactions during pregnancy. However, the expression of endometrial TNF-alpha has not been investigated especially during early pregnancy, a phenomenon invariably accompanied by inflammatory reaction. In the present study, the endometrial expressions of TNF-alpha and its receptors (TNFR1 and TNFR2) during early pregnancy, when the embryo lies free in the zona hatched state in the uterine lumen, were analyzed by immunohistochemistry. The endometrial expressions of TNF-alpha, TNFR1 and TNFR2 were found to be significantly up-regulated (p < 0.05) in the glandular epithelium on day 6 post-ovulation in pregnant animals. The alteration in the expression of these molecules may contribute to the induction of local inflammatory reactions during implantation. 相似文献
