The binding of [3H]GABA and [3H]flunitrazepam was performed with synaptic membranes and post-synaptic densities (PSDs) isolated from canine cerebral cortex and cerebellum. Two GABA binding sites were found with cerebral cortex membranes but only one with cerebellar membranes. PSDs isolated from these showed only single binding sites, with cerebellar PSDs exhibiting lower KD values and a larger concentration of sites than did cerebral cortex PSDs. In the case of flunitrazepam, only one binding site was found for all four preparations, with cerebellar PSDs having twice the concentration of sites of cerebral PSDs. Photoaffinity labeling of the flunitrazepam receptor in PSDs resulted in the binding to a 51,000 Mr protein in both cases, with cerebellar PSDs again showing an increased concentration over that found in cerebral cortex PSDs. Based on this work, and on earlier work of ourselves and of others, we conclude that both populations of isolated PSDs contain inhibitory sites, but that the intact PSDs in both preparations are derived from Gray type I, probably excitatory, synapses, and that the inhibitory sites are found in the broken-up material in the PSD fractions which are derived from Gray type II, probably inhibitory, synapses.  相似文献

目的:探讨灰阶超声和多普勒超声对甲状腺乳头状癌的诊断价值。方法:选取2013年9月~2014年9月在我院入院接受治疗的90例甲状腺乳头状癌。患者入院后,分别采用灰阶超声诊断和多普勒超声诊断对患者检查诊断,并对两种超声诊断方法的诊断率和对淋巴转移的诊断准确率进行比较分析,并对两种诊断方法的检查图像表现进行比较。结果:灰阶超声诊断诊断出82例,确诊率为91.1%,而多普勒超声诊断出85例,确诊率为94.4%。多普勒超声确诊率高于灰阶超声的,但无显著性差异(P0.05)。而灰阶超声诊断出有38例患者存在淋巴转移现象,诊断率为61.3%,多普勒超声诊断出有58例患者存在淋巴转移现象,诊断率为93.5%,多普勒超声的诊断率明显高于灰阶超声的,差异显著,具有统计学意义(P0.05)。结论:灰阶超声和多普勒超声对甲状腺乳头状癌都有较高的准确率,但多普勒超声在诊断患者是否有淋巴转移方面诊断率更高,结节内的血流信号更准确,其确诊率高达94.4%(P0.05)。通过检测有淋巴转移的患者的淋巴结内部的血管会变粗或增多等信息可提高淋巴转移的确诊率,具有可观的临床疗效。  相似文献

BackgroundIn Chile, the peony is the most important ornamental flower exported from the country. Gray mould is a phytopathological problem of this crop. This disease is caused by Botrytis cinerea and Botrytis paeoniae.AimsWe carried out the first survey of Botrytis species associated with peony gray mould in Southern Chile to estimate the diversity of these pathogens.MethodsDiseased peony leaves were collected from seven locations in Southern Chile covering a distance of 300 km. The Botrytis isolates obtained were studied by morphological and molecular methods. Finally, a PCR assay using primers based on the necrosis and ethylene-inducing protein gene (nep1) was used to specifically identify B. paeoniae.ResultsSeventeen isolates belonging to Botrytis genus were obtained, and all of them were pathogenic to peonies when inoculated in plants grown in a greenhouse. Morphological analyses showed that four isolates shared common characteristics, which distinguish them from the rest. Homology and phylogenetic analysis of G3PDH, as well as determination of the Bc-hch allele, allowed us to identify 12 isolates as B. cinerea, 4 as B. paeoniae and one isolate as Botrytis pseudocinerea. The PCR assay was found to be specific to B. paeoniae, amplifying a single band of 470 bp.ConclusionsThree Botrytis species involved in peony gray mould disease are present in Chile. This is the first time that both B. paeoniae and B. pseudocinerea have been reported to be present in the country and also that they affect peonies. Finally, to our knowledge, the PCR based method herein described is the first of its kind to be used to identify B. paeoniae.  相似文献

The role of the sesquiterpene botrydial in the interaction of the phytopathogenic fungus Botrytis cinerea and plant-associated bacteria was analyzed. From a collection of soil and phyllospheric bacteria, nine strains sensitive to growth-inhibition by B. cinerea were identified. B. cinerea mutants unable to produce botrydial caused no bacterial inhibition, thus demonstrating the inhibitory role of botrydial. A taxonomic analysis showed that these bacteria corresponded to different Bacillus species (six strains), Pseudomonas yamanorum (two strains) and Erwinia aphidicola (one strain). Inoculation of WT and botrydial non-producing mutants of B. cinerea along with Bacillus amyloliquefaciens strain MEP218 in soil demonstrated that both microorganisms exert reciprocal inhibitory effects; the inhibition caused by B. cinerea being dependent on botrydial production. Moreover, botrydial production was modulated by the presence of B. amyloliquefaciens MEP218 in confrontation assays in vitro. Purified botrydial in turn, inhibited growth of Bacillus strains in vitro and cyclic lipopeptide (surfactin) production by B. amyloliquefaciens MEP218. As a whole, results demonstrate that botrydial confers B. cinerea the ability to inhibit potential biocontrol bacteria of the genus Bacillus. We propose that resistance to botrydial could be used as an additional criterion for the selection of biocontrol agents of plant diseases caused by B. cinerea.  相似文献

The physical interactions of fetal antigens (tumor-associated fetal antigens; TAFA-I, TAFA-II, and TAFA-III) with rat histocompatibility antigens were studied. TAFA-I and TAFA-III are present on syngeneic (NBR) and allogeneic (Fisher F344, Wistar Furth, and White Buffalo) rat embryo fibroblasts and on tumor cells. TAFA-II was found only on NBR (syngeneic) rat embryo fibroblasts and on NBR tumor cells. Antibody-blocking experiments were used to examine the fetal and histocompatibility antigen topography on cell membranes of tumor cells transformed by chemical and viral carcinogens. Precoating the tumor cells with alloantisera inhibited the subsequent adsorption of anti-NBR embryo, anti-TAFA-I, and anti-TAFA-III sera, but not anti-TAFA-II serum. Immunofluorescent cocapping experiments indicated that TAFA-I and TAFA-III, as well as other fetal antigens found on cells from 14-day gestation NBR embryos cocap with histocompatibility antigens when tested on syngeneic embryo fibroblasts and on sarcoma cells. TAFA-I cocapped with White Buffalo (Buf) strain rat histocompatibility antigens on herpes simplex Type II virus-transformed cells. The specificity of the TAFA-histocompatibility interactions was confirmed by demonstrating that the different anti-TAFA sera did not have contaminating antiviral antigen specificity; and also that these interactions did not occur on normal adult fibroblasts or spleen cells.  相似文献

Infection of mice with live influenza A virus induces cytolytic T lymphocytes (CTL) as well as B cells capable of reacting with target cells infected with the appropriate virus subtypes. In Balb/c mice CTL reveal a broad cross-reactivity against all influenza A substrains known. In contrast B-cell responses are restricted to virus subtypes which are identical in regard to the hemagglutinin (HA) of the sensitizing virus. Reinfection with homologous live influenza virus within 6–7 months results in no or in a drastically diminished B-cell response as compared to a priming situation and fails to induce CTL. Inability to induce secondary immunity to homologous influenza virus was correlated with the presence of circulating antibodies specific for the sensitizing virus subtype. Cross-boosting with heterologous live influenza A virus induces homotypic and heterotypic CTL and B-cell immunity with characteristics of secondary responses. Preparations of inactivated intact influenza virus are unable to reactivate CTL memory in vivo but induce B-cell activity. B-cell responses stimulated by this procedure are restricted to the boosting virus. Attenuated viruses, which are produced by recombination of wild strains with cold-adapted strains, are also efficient in stimulating in vivo CTL memory if used for cross-boosting.  相似文献

This study describes the gel‐free phosphoproteomic analysis of the phytopathogenic fungi Alternaria brassicicola and Botrytis cinerea grown in vitro under nonlimiting conditions. Using a combination of strong cation exchange and IMAC prior to LC‐MS, we identified over 1350 phosphopeptides per fungus representing over 800 phosphoproteins. The preferred phosphorylation sites were found on serine (>80%) and threonine (>15%), whereas phosphorylated tyrosine residues were found at less than 1% in A. brassicicola and at a slightly higher ratio in B. cinerea (1.5%). Biological processes represented principally among the phoshoproteins were those involved in response and transduction of stimuli as well as in regulation of cellular and metabolic processes. Most known elements of signal transduction were found in the datasets of both fungi. This study also revealed unexpected phosphorylation sites in histidine kinases, a category overrepresented in filamentous ascomycetes compared to yeast. Animal studies have found that deficits in brain docosahexaenoic acid (DHA, 22:6n-3) accrual during perinatal development leads to transient and enduring abnormalities in brain development and function. Determining the relevance of this evidence to brain disorders in humans has been hampered by an inability to determine antimortem brain DHA levels and limitations associated with a postmortem approach. Accordingly, there is a need for alternate or complementary approaches to better understand the role of DHA in cortical function and pathology, and conventional magnetic resonance imaging (MRI) techniques may be ideally suited for this application. A major advantage of neuroimaging is that it permits prospective evaluation of the effects of manipulating DHA status on both clinical and neuroimaging variables. Emerging evidence from MRI studies suggest that greater DHA status is associated with cortical structural and functional integrity, and suggest that reduced DHA status and abnormalities in cortical function observed in psychiatric disorders may be interrelated phenomenon. Preliminary evidence from animal MRI studies support a critical role of DHA in normal brain development. Neuroimaging research in both human and animals therefore holds tremendous promise for developing a better understanding of the role of DHA status in cortical function, as well as for elucidating the impact of DHA deficiency on neuropathological processes implicated in the etiology and progression of neurodevelopmental and psychiatric disorders.  相似文献

Behavior of the laboratory gray short-tailed opossums (Monodelphis domestica), Warsaw Wild Captive Pisula Stryjek rats (WWCPS) and laboratory rats (Wistar) has been registered in the period of familiarization with a new environment and consecutive confrontation with a novel, innocuous object placed in that familiarized environment. In the new environment the sequence of anxiety, investigation, and habituation was shortest in the opossum, longer in the laboratory rat and longest in the WWCPS rat. When placed in it, gray short-tailed opossums investigated the new environment with the shortest delay and most intensity. In reaction to novel objects, opossums and laboratory rats prolonged the time spent in the proximity of the new object, while the WWCPS rat did not show that reaction. Both opossums and laboratory rats increased the number of contacts with the new object, whereas WWCPS rats reduced those contacts. Behavior of all three species and lines grouped in different clusters. Some other quantitative and qualitative differences in behavior of the investigated animals are also described, showing a higher level of anxiety in both lines of rats than in the opossum. Behavioral differences between species and lines of animals used in this study may be attributed to different ecological adaptations of rats and opossums and to the effect of domestication in the laboratory rats. These behavioral differences make comparisons of opossums vs rat, and wild rat vs laboratory rat interesting models for studying the brain mechanisms of anxiety and neotic motivations.  相似文献

2011 年11 月5 日,福建省平潭县白青乡青峰村渔民出海作业时发现一头误撞定置刺网死亡的灰鲸,为福建省的首次灰鲸误捕案例,也是本世纪以来我国海域的首次灰鲸误捕记录。本文描述了该灰鲸的外形特征和骨骼系统,并报道外形和骨骼的测量数据。该灰鲸系雌性,体长1 309 cm,体重约21 t,为目前本种在我国海域搁浅/ 误捕记录中的最大个体。头骨长281 cm,宽128 cm,重250 kg;脊椎式为C7 + T13 + L13 +Ca23 = 56;指式为Ⅰ1,Ⅱ3,Ⅲ7,Ⅳ5,Ⅴ3;肋骨14 对,V 形骨10 枚。脊椎式、指式、肋骨和V 形骨较之以前的报道存在差异,说明灰鲸的脊椎式、指式、肋骨和V 形骨可能存在个体差异。此外,本文还综述了西北太平洋灰鲸目前所面临的主要致危因素。  相似文献
