The influence of a varied sucrose supply on grain size and hormonal contents of detached wheat ears ( Triticwn aestivum L. cv. Schirokko) was investigated throughout grain development. In ears led limited amounts, or no sucrose, grain weights in both proximal and distal grain positions of the ear were reduced. Radioimmunoassay for abscisic acid, indoleacetic acid and zeatin/zeatin riboside showed that the changes in the levels of these hormones in grains and bracts were comparable to intact ears when detached ears were well supplied with sucrose. Under conditions of limited sucrose supply, higher abscisic acid levels in the distal and proximal grains of detached ears were found compared to ears supplied with adequate sucrose. Limiting sucrose supply to the ear did not alter the levels of indoleacetic acid or zeatin/zeatin riboside in either the grains or bracts of detached ears.  相似文献

The flowers of the hemiparasitic family Loranthaceae are always subtended by a rimmed structure known as the calyculus. The origin and identity of the calyculus have been disputed for more than a century. Various hypotheses have been proposed, for example, an outgrowth of the axis, a reduced calyx, and a bracteolar (prophyllar) origin, but controversies remain. To obtain a plausible explanation of the origin of the calyculus, we investigated the flowers of Loranthus tanakae using scanning electron microscopy and light microscopy to observe the entire developmental process of the floral parts. Our results show that bracts are not present in L. tanakae. The calyculus, which lacks serving vascular bundles, initiates as a semicircular primordium and then develops into a circular structure by an adnation at both sides. The flower primordium usually cleaves into six petals from its centre along a whorled pattern in two series with three petals each, before or after the calyculus closed. Isomerous stamen primordia probably follow the same initiation pattern as petals do. Several carpels of different sizes initiate simultaneously as a united primordium. We support the hypotheses that the calyculus is of bract or bracteole origin due to its independent initiation from the inflorescence rachis, its similar morphology and positioning as the bract or bracteole, and that having no developmental relationship with the petals. We suggest keeping the usage of the term “calyculus”. Loranthus flowers should be considered monochlamydeous with three whorls of floral parts, namely petal, androecium, and gynoecium.  相似文献

Summary The quantitative traits height and ear-emergence date were analyzed in the F2 progeny of a cross between a tall winter barley cultivar (Gerbel) and a short spring barley cultivar (Heriot). The trait distributions were found to be related to the genotypes at two biochemical loci, -amylase (Bmy1) and water-soluble protein (Wsp3), which are known to lie on the long arm of chromosome 4. Linkages between each trait and the markers were investigated using normal mixture models. The two parental phenotypes and the heterozygote phenotype of Bmy1 were distinguishable so the model could be used directly to estimate linkage between Bmy1 and a quantitative trait locus (QTL) for height (Height). The Gerbel homozygote and heterozygote phenotype of Wsp3 could not be distinguished and the model was adapted accordingly. The proportion of plants requiring vernalization was consistent with control by two independent genes acting epistatically, and a normal mixture model based on a two-gene hypothesis was fitted to the distribution of ear-emergence date to estimate linkage between the marker loci and a QTL for ear-emergence date (Vrn1). The parameters of each model were the recombination fraction between the marker locus and the QTL and the means and standard deviations associated with each QTL genotype; these were estimated by maximum likelihood. The fitted distributions correspond well to those observed and the order of the loci along the chromosome is inferred to be HeightVrn1Bmy1Wsp3, with Wsp3 being the most distal.  相似文献

The otic placode generates the auditory and vestibular sense organs and their afferent neurons; however, how auditory and vestibular fates are specified is unknown. We have generated a fate map of the otic placode and show that precursors for vestibular and auditory cells are regionally segregated in the otic epithelium. The anterior-lateral portion of the otic placode generates vestibular neurons, whereas the posterior-medial region gives rise to auditory neurons. Precursors for vestibular and auditory sense organs show the same distribution. Thus, different regions of the otic placode correspond to particular sense organs and their innervating neurons. Neurons from contiguous domains rarely intermingle suggesting that the regional organisation of the otic placode dictates positional cues to otic neurons. But, in addition, vestibular and cochlear neurogenesis also follows a stereotyped temporal pattern. Precursors from the anterior-lateral otic placode delaminate earlier than those from its medial-posterior portion. The expression of the proneural genes NeuroM and NeuroD reflects the sequence of neuroblast formation and differentiation. Both genes are transiently expressed in vestibular and then in cochlear neuroblasts, while differentiated neurons express Islet1, Tuj1 and TrkC, but not NeuroM or NeuroD. Together, our results indicate that the position of precursors within the otic placode confers identity to sensory organs and to the corresponding otic neurons. In addition, positional information is integrated with temporal cues that coordinate neurogenesis and sensory differentiation.  相似文献

小麦光温敏雄性不育系BNS育性转换与内源激素的关系研究   总被引:1,自引:0,他引:1  
BNS是一种光温敏型小麦不育新类型,具有不育彻底、育性转换稳定等特性,对小麦杂种优势研究有重要价值。为研究内源激素在BNS育性调控中的作用,以秋播与春播BNS花粉发育的四分体期至三核期4个关键时期幼穗为实验材料,采用间接酶联免疫(ELISA)法分别测定了BNS不育和可育幼穗中生长素(IAA)、脱落酸(ABA)、赤霉素(GA3)和玉米素核苷(ZR)4种内源激素的含量变化。结果显示:(1)BNS在陕西关中地区正常秋播雄性不育、早春播雄性育性正常。(2)单核期和三核期是BNS温度敏感期,单核期均温高于15.35℃、三核期均温高于18.64℃时,BNS育性发生转换,雄性不育度随温度升高逐渐降低。(3)BNS在敏感期响应外界温度,4种内源激素的含量发生显著变化,调控育性表达;单核期不育幼穗中IAA、GA3和ZR含量比可育幼穗低21.9%、33.6%和30.2%,ABA含量反而增加23.4%;三核期不育幼穗ABA和ZR含量分别比可育幼穗高59.7%和31.4%,GA3则低44.0%。(4)与雄性可育幼穗相比,BNS雄性不育幼穗在单核期的IAA/GA3、ABA/GA3值偏高;二核期IAA/ABA值较低;三核期的IAA/GA3、ABA/GA3值较高,IAA/ABA值偏低。3个时期的IAA、ABA和GA3比例失调,阻碍了小孢子正常发育,导致BNS雄性败育。研究认为:BNS在花粉发育的单核期和三核期对外界温度变化敏感,通过内源激素代谢调控自身育性转换。  相似文献

Vertebrate inner ear development is initiated by the specification of the otic placode, an ectodermal structure induced by signals from neighboring tissue. Although several signaling molecules have been identified as candidate otic inducers, many details of the process of inner ear induction remain elusive. Here, we report that otic induction is responsive to the level of Hedgehog (Hh) signaling activity in Xenopus, making use of both gain- and loss-of-function approaches. Ectopic activation of Hedgehog signaling resulted in the development of ectopic vesicular structures expressing the otic marker genes XPax-2, Xdll-3, and Xwnt-3A, thus revealing otic identity. Induction of ectopic otic vesicles was also achieved by misexpression of two different inhibitors of Hh signaling: the putative Hh antagonist mHIP and XPtc1deltaLoop2, a dominant-negative form of the Hh receptor Patched. In addition, misexpression of XPtc1deltaLoop2 as well as treatment of Xenopus embryos with the specific Hh signaling antagonist cyclopamine resulted in the formation of enlarged otic vesicles. In summary, our observations suggest that a defined level of Hh signaling provides a restrictive environment for otic fate in Xenopus embryos.  相似文献

Myosin IIIa (Myo3A) transports cargo to the distal end of actin protrusions and contains a kinase domain that is thought to autoregulate its activity. Because Myo3A tends to cluster at the tips of actin protrusions, we investigated whether intermolecular phosphorylation could regulate Myo3A biochemical activity, cellular localization, and cellular function. Inactivation of Myo3A 2IQ kinase domain with the point mutation K50R did not alter maximal ATPase activity, whereas phosphorylation of Myo3A 2IQ resulted in reduced maximal ATPase activity and actin affinity. The rate and degree of Myo3A 2IQ autophosphorylation was unchanged by the presence of actin but was found to be dependent upon Myo3A 2IQ concentration within the range of 0.1 to 1.2 μm, indicating intermolecular autophosphorylation. In cultured cells, we observed that the filopodial tip localization of Myo3A lacking the kinase domain decreased when co-expressed with kinase-active, full-length Myo3A. The cellular consequence of reduced Myo3A tip localization was decreased filopodial density along the cell periphery, identifying a novel cellular function for Myo3A in mediating the formation and stability of actin-based protrusions. Our results suggest that Myo3A motor activity is regulated through a mechanism involving concentration-dependent autophosphorylation. We suggest that this regulatory mechanism plays an essential role in mediating the transport and actin bundle formation/stability functions of Myo3A.  相似文献

摘要 目的:探讨胶原生物膜在耳内镜下乳突根治术中的应用效果。方法:选取徐州医科大学附属医院2021年4月至2022年 2月收治的51例中耳胆脂瘤患者进行回顾性分析,其中研究组27例患者予以胶原生物膜修复皮肤缺损,对照组予以颞肌筋膜修复术腔皮肤缺损,观察两组患者术后临床症状,手术时长,术腔完全上皮化时间、干耳时间及术前术后听力改变。结果:研究组术后患者因外耳道进水,存在感染及肉芽生长者1例,予以清理后未再次生长;对照组术后发生1例外耳道口狭窄的情况,予以橡胶扩张管进行扩张并后并定期清理术腔肉芽、脱落痂皮,患者外耳道恢复良好。两组术前耳闷、耳痛、耳鸣及术后耳痛VAS评分无明显差异(P>0.05);研究组术后耳闷及耳鸣VAS评分较对照组降低(P<0.05)。研究组平均手术时长、术后术腔完全上皮化时间及平均干耳时间短于对照组(P<0.05)。两组术前术后气骨导差(ABG)、平均气导听阈(AC)比较差异均无统计学意义(P>0.05)。结论:作为术区移植物,胶原生物膜应用于耳内镜下中耳胆脂瘤乳突根治术可加快创面术腔的修复,减少局部创伤与操作步骤,改善临床症状,缩短手术时间、术后术腔完全上皮化时间及获得干耳时间,可作为临床上有效的修复材料。  相似文献
