Films of methylcellulose (MC), poly(ethylene glycol)400 (PEG400) plasticized MC, and MC gels (MC crosslinked with glutaraldehyde (GA)) were prepared by casting from aqueous solutions. The swelling test has shown that the MC gels were insoluble in water and that their crosslinking density increased with increasing GA and HCl concentrations. The effect of the addition of PEG400 or GA to MC was investigated through dynamic mechanical analysis (DMA). The DMA analysis of PEG400/MC blends has shown that PEG400 was compatible with MC and was an effective plasticizer since the curves of tan δ against temperature exhibited single peaks (corresponding to a single glass transition temperature), which were displaced to lower values with increasing PEG400 content. The thermogravimetric analysis (TGA) indicated that the thermal stability of MC was not affected by the chemical crosslinking. The tensile strength was slightly increased through crosslinking while the elongation was slightly decreased. The presence of moisture in MC hydrogels decreased the tensile strength and enhanced the elongation while the addition of PEG400 decreased the tensile strength but sharply increased the elongation. 相似文献

Summary Next‐generation sequencing technologies are poised to revolutionize the field of biomedical research. The increased resolution of these data promise to provide a greater understanding of the molecular processes that control the morphology and behavior of a cell. However, the increased amounts of data require innovative statistical procedures that are powerful while still being computationally feasible. In this article, we present a method for identifying small RNA molecules, called miRNAs, which regulate genes by targeting their mRNAs for degradation or translational repression. In the first step of our modeling procedure, we apply an innovative dynamic linear model that identifies candidate miRNA genes in high‐throughput sequencing data. The model is flexible and can accurately identify interesting biological features while accounting for both the read count, read spacing, and sequencing depth. Additionally, miRNA candidates are also processed using a modified Smith–Waterman sequence alignment that scores the regions for potential RNA hairpins, one of the defining features of miRNAs. We illustrate our method on simulated datasets as well as on a small RNA Caenorhabditis elegans dataset from the Illumina sequencing platform. These examples show that our method is highly sensitive for identifying known and novel miRNA genes. 相似文献

The objective of this study was to investigate the influence of dynamic compressive loading on chondrogenesis of mesenchymal stem cells (MSCs) in the presence of TGF-β3. Isolated porcine MSCs were suspended in 2% agarose and subjected to intermittent dynamic compression (10% strain) for a period of 42 days in a dynamic compression bioreactor. After 42 days in culture, the free-swelling specimens exhibited more intense alcian blue staining for proteoglycans, while immunohistochemical analysis revealed increased collagen type II immunoreactivity. Glycosaminoglycan (GAG) content increased with time for both free-swelling and dynamically loaded constructs, and by day 42 it was significantly higher in both the core (2.5 ± 0.21%w/w vs. 0.94 ± 0.03%w/w) and annulus (1.09 ± 0.09%w/w vs. 0.59 ± 0.08%w/w) of free-swelling constructs compared to dynamically loaded constructs. This result suggests that further optimization is required in controlling the biomechanical and/or the biochemical environment if such stimuli are to have beneficial effects in generating functional cartilaginous tissue. 相似文献

In order to predict species response to climate and land-use change, numerically fast and easily applicable assessment tools for species survival are required. We present a set of formulae to calculate the mean lifetime of a metapopulation in a spatially heterogeneous and dynamic landscape subject to habitat patch diminution, loss and/or spatial shift of the habitat network. The formulae require as inputs (i) information about the number, location and size of the habitat patches for several time steps to quantify landscape dynamics in terms of patch destruction, diminution or shifting rates and (ii) data on species traits such as their vulnerability to environmental variation and their dispersal ability to quantify local colonisation and extinction rates. We validate the formulae with a spatially explicit simulation. The analysis is complemented by a protocol for the easy use of the approach and practical application examples. Using data from the English Longitudinal Study of Ageing over the period 2004–2017, this paper explores the effects of the Great Recession and its aftermath upon health-compromising behaviours in adults aged 50 and over. We introduce new techniques into this area of research, namely dynamic random-effects logit estimators which control for initial conditions and correlated individual effects. We observe a lack of crisis effect upon the probabilities of smoking and being physically inactive, as well as of transitioning in and out of these behaviours. In line with other recent literature, this suggests that the relationship between economic recessions and smoking and physical inactivity may have broken down. Alternatively, the over 50s may have been protected from the crisis and subsequent austerity measures. Nonetheless, both the crisis and post-crisis period were associated with a lower probability of drinking frequently. 相似文献

We investigated relationships between fecal androgen concentrations, facial coloration and behaviour in semi-free-ranging male mandrills. We found that fecal androgen levels were significantly positively related to dominance rank, independent of rank stability and the mating period, suggesting that male mandrills live in a permanently aggressive context in which they must actively maintain their dominance status. Facial red coloration was also significantly related to both fecal androgen levels and rank, with high ranking males having both higher androgen levels and redder faces, although dominant males did not always have the highest androgen levels or the reddest faces. Predictive relationships between androgen levels, coloration and rank were short-term. Androgen concentrations and facial redness both increased in the presence of receptive females, as did the former during periods of rank instability. We conclude that male facial redness is likely to represent an honest signal (to other males) of current androgen status, competitive ability and willingness to engage in fights and that females may also use this to assess male condition. 目的:对比锁定加压钢板与动力加压钢板治疗肱骨中下段骨折患者的临床疗效。方法:抽取我院2010年8月~2013年12月收治的肱骨中下段骨折患者76例,按照随机数字表法分为锁定组(锁定加压钢板治疗)与动力组(动力加压钢板治疗),每组38例,随访1年。对比两组患者临床指标,治疗效果及并发症发生率。结果:锁定组患者手术时间、骨折愈合时间及住院时间均小于动力组,差异均有统计学意义(P0.05);锁定组患者优良率为89.47%,显著高于动力组的71.05%,差异有统计学意义(P0.05);锁定组的并发症发生率为10.53%,显著低于动力组的31.58%,差异有统计学意义(P0.05)。结论:锁定加压钢板应用于治疗肱骨中下段骨折患者疗效优于动力加压钢板治疗,它具有创伤小、恢复快的优点,且并发症发生率较低,值得推广。 相似文献

The auditory systems of humans and many other species use the difference in the time of arrival of acoustic signals at the two ears to compute the lateral position of sound sources. This computation is assumed to initially occur in an assembly of neurons organized along a frequency-by-delay surface. Mathematically, the computations are equivalent to a two-dimensional cross-correlation of the input signals at the two ears, with the position of the peak activity along this surface designating the position of the source in space. In this study, partially correlated signals to the two ears are used to probe the mechanisms for encoding spatial cues in stationary or dynamic (moving) signals. It is demonstrated that a cross-correlation model of the auditory periphery coupled with statistical decision theory can predict the patterns of performance by human subjects for both stationary and motion stimuli as a function of stimulus decorrelation. Implications of these findings for the existence of a unique cortical motion system are discussed. 相似文献
