Genetic analysis of transpositions in the lac region of Escherichia coli

The lac region of Escherichia coli, carried on an F′ lacproB episome, was used as a target for the transposition of several transposable elements. Tn9 shows a preferential integration (by a factor of 50) into a region extending from the end of the Z gene through the Y gene. Throughout the remainder of the lacI, Z and Y genes one other short region, located in the middle of the I gene, is favored for integration. Within these favored regions many different integration points are evident. Inspection of the DNA sequence for the I and Y genes, and parts of the Z gene, shows a strong correlation between A + T richness and regions of preferential integration. Tn5 insertions follow a similar pattern, although with less preference; whereas Tn10 insertions (provided by T. J. Foster), also favor the Y gene and the end of Z, but are distributed among fewer integration points. Most of the Tn3 insertions into the episome are accompanied by a nearby or adjacent deletion.     

Phenice's visual sexing technique for the os pubis: a critique

Innominate bones from 362 California Indians were sexed with Phenice's three non-metrical features of the os pubis. The frequencies of marked, intermediate, and absent cases of these three morphological features were tabulated in males and females to see if unambiguous and reliable distinctions were consistently available. The results suggest that Phenice's technique offered extremely reliable sex evaluations in this material.     

Two different gamma-glutamyltransferases during development of liver and small intestine: a fetal (sialo-) and an adult (asialo-) glycoprotein.

Depending on the developmental stage, the gamma-glutamyltransferase (E.C. 2.3.2.2) exists in two different types in the liver and in the small intestine: a sialic acid-rich fetal type and a sialic acid-poor adult type. The fetal type could be detected in the undifferentiated cryptal cells, in the fetal small intestine and in the fetal liver, and the adult type in the differentiated villous cells and in the adult liver. The separation of both types was performed using ConA-sepharose, which does not bind the fetal type but the adult type. Binding was reached by neuraminidase treatment.     

Inhibitory effect of hydroxyurea on [3-H]leucine incorporation in human peripheral lymphocytes.

Hydroxyurea (HU), generally considered to be a specific inhibiter of DNA synthesis, has an inhibitory effect on the incorporation of TCA-precipitable [3-H]leucine in peripheral lymphocytes. This action is not secondary to the inhibition of DNA synthesis since incorporation of [3-H]leucine is unaffected when DNA synthesis is inhibited by 5-fluorodeoxyuridine (FUdR); it does not appear to be directly related to inhibition of RNA synthesis; and it is not mediated at the level of translation since HU has no effect on protein synthesis in rabbit reticulocytes. The relevance of these findings to the use of HU as a DNA inhibitor is discussed.
