应用光镜和透射电镜技术研究了日本沼虾输精管的结构及其在精荚形成中的作用。结果表明,日本沼虾输精管从形态结构上可分为近端输精管、卷曲输精管、远端输精管和膨大的远端输精管四部分。各部分的管壁皆由分泌上皮、基膜、肌肉层和结缔组织构成,其中分泌上皮包括高度明显不同的低柱状上皮和高拄状上皮两部分。输精管各部分管腔内含有处于不同形成阶段的精荚。进入近端输精管内的精子被支撑在一种嗜酸性基质中。近端输精管的分泌物主要帮助形成精子团,同时形成精荚壁的极小部分。卷曲和远端输精管分泌形成精荚壁的绝大部分,其分泌物由细胞顶端通过外排作用和顶泌机制分泌产生。膨大的远端输精管具有贮存精荚的作用,其分泌上皮也通过外排作用和顶泌机制产生分泌物包裹在已基本形成的精荚外侧,管壁肌肉层在雌雄交配时将管腔内的精荚切割成适宜长度并排出体外。

In this study, we attempted to identify the interactions and mechanisms between veratrine and paeoniflorin on isolated mouse vas deferens. Paeoniflorin had no effect on isolated mouse vas deferens. Veratrine (1 x 10(-5) approximately 1 x 10(-3) g/ml) could directly induce contraction of isolated rat and mouse vas deferens. The concentration induced by veratrine (1 x 10(-5) g/ml) was completely inhibited by Ca2+-free solution and verapamil (1 x 10(-5) M), in both the epididymal and the prostatic portions of isolated mouse vas deferens. Naloxone (1 x 10(-5) M) did not alter the contraction induced by veratrine (1 x 10(-5) g/ml) in either the epididymal or the prostatic portions of isolated mouse vas deferens. Paeoniflorin (4.8 x 10(-5) g/ml) inhibited the contraction induced by veratrine (1 x 10(-5) g/ml) in both the epididymal and the prostatic portions of isolated mouse vas deferens. Paeoniflorin (4.8 x 10(-5) g/ml) potentiated norepinephrine (1 x 10(-5) M)-induced phasic contraction in the epididymal portion, but decreased contractions in the prostatic portion. Paeoniflorin (4.8 x 10(-5) g/ml) increased KCI (56 mM)-induced phasic contraction in the epididymal portion, but decreased the tonic contraction in either the epididymal or the prostatic portion. Veratrine (1 x 10(-5) g/ml)-induced contractions could be decreased by pretreatment with ryanodine (1 x 10(-5) M) in both the epididymal and the prostatic portions. Pretreatment with the combination of paeoniflorin (4.8 x 10(-5) g/ml) and ryanodine (1 x 10(-5) M) did not potentiate the inhibition of paeoniflorin in the veratrine-induced contraction in both the epididymal and the prostatic portions of isolated mouse vas deferens. 相似文献

本文旨在探讨大鼠新鲜离体输精管平滑肌细胞中乙酰胆碱(acetylcholine,ACh)引起超极化反应的机制,采用细胞内微电极记录技术和细胞内荧光标记技术研究ACh对大鼠输精管不同走行方向平滑肌细胞的作用。用尖端含0.1%碘化吡啶(propidium iodide,PI)的记录电极标记电生理记录后的平滑肌细胞,其中37个为外层纵行细胞,17个为内层环行细胞。它们的平均静息膜电位分别为(-53.56±3.88)mV和(-51.62±4.27)mV,膜输入阻抗分别为(2245.60±372.50)MQ和(2101.50±513.50)MQ。ACh引起的膜超极化反应是浓度依赖性的,EC50为36 μmol/L。ACh引起的超极化反应可被非选择性的毒草碱(muscarinic receptor,M)受体阻断剂阿托品(atropine,1 μmol/L)和选择性的M3受体阻断剂diphenylacetoxy-N-methylpiperidine-methiodide(DAMP,100nmol/L)阻断。ACh引起的超极化还能被一氧化氮合酶抑制剂L-硝基-精氨酸甲酯(N-nitro-L-arginine methylester,L.NAME,300μmol/L)阻断,并可被ATP敏感的钾通道阻断剂glipizide(5μmol/L)或内向整流钾通道阻断剂钡离子(50μmol/L)部分阻断。Glipizide和钡离子联合使用可完全阻断ACh引起的超极化反应。上述结果表明:ACh通过作用于大鼠输精管平滑肌细胞膜上的M3受体引起超极化反应,一氧化氮、ATP敏感性钾通道和内向整流钾通道参与了ACh引起的超极化反应。 相似文献

Covalent tobacco mosaic virus-enkephalin analogue conjugates containing between 5 and 450 agonist molecules attached through their C-terminal amino acids were prepared and assayed in pharmacological and binding assays that distinguish between mu- and delta-receptors. Increases in peptide- and receptor-specific potency and affinity were observed with a certain preference of mu- over delta-receptors. Seventeen analogues of dermorphin were synthesized and bio-assayed to determine the influence of side chains of the individual amino acid residues forming the sequence of dermorphin on the biological activity of this opioid peptide. Syntheses were carried out using solid-phase procedure, and the analogues obtained were purified by gel filtration on Sephadex G-10. Biological activities determined in guinea pig ileum (GPI) and mouse vas deferens (MVD) tests showed that the N-terminal tetrapeptide is responsible for the activity of dermorphin. Substitutions in the C-terminal fragment, particularly in position 5, for other amino acid residues results in substantial differentiation towards mu and delta receptors. 相似文献

The concentrations of dopamine (DA), 5-hydroxytryptamine (5-HT) and noradrenaline (NA) in the rat vas deferens divided in eight or four sections were determined by high performance liquid chromatography with electrochemical detection. Dopamine and NA had the same regional distribution; their concentrations were maximal near the prostatic end and decreased towards the epididymis. The concentration of 5-HT also decreased from the prostatic to the epididimal end, but 5-HT did not follow the same regional distribution as DA and NA. Reserpine (0.02 or 0.2 mg/kg, i.p., 24 hr) and 6-hydroxydopamine (2×80 mg/kg, i.v., 6 days) decreased the contents of DA and NA; the concentrations of both amines were modified to a similar extent. Reserpine also diminished the content of 5-HT. Pargyline (200 mg/kg, i.p., 2 hr) increased the concentration of 5-HT whilep-chlorophenylalanine (300 mg/kg, oral, 3 days) decreased the contents of the amine in some sections of the vas deferens. This study suggests that DA and NA co-exist in the same sympathetic neurons. Some of the 5-HT could be stored in mast cells as previously proposed, but the finding that tissue content of 5-HT changes after inhibiting the deamination or synthesis of the amine suggests that other source(s) of 5-HT distinct from mast cells exist in the rat vas deferens. 相似文献

We have compared the effects of vasoactive intestinal peptide (VIP) and of the VIP-related peptides pituitary adenylate cyclase activating peptide (PACAP) 1–27 and 1–38, helodermin, helospectin I and helospectin II, on the electrically evoked twitches in the isolated vas deferens of the rat. While VIP was virtually without effect, PACAP 1–38 suppressed the electrically evoked twitches effectively and in a concentration-dependent manner (pIC50 value 7.5). The naturally occurring N-terminal fragment PACAP 1–27 was less effective than PACAP 1–38 (Imax values 37.2% suppression compared to 76.5%) and less potent. The C-terminal fragment PACAP 16–38 was virtually inactive. Also helodermin and helospectin I+II suppressed the electrically evoked twitches effectively and in a concentration-dependent manner (pIC50 values 6.9; 7.2; 6.8, respectively). The three peptides produced similar maximum reduction of the twitches (74–80%).
The findings suggest that PACAP, helodermin and helospectin suppress the electrically evoked contractions in the rat vas deferens via receptors distinct from VIP receptors. 相似文献

We studied the male reproductive cycle in a population of Ophisops elegans from Mount Sannine, Lebanon, by histological analysis. Testicular histology showed active spermatogenesis in spring, followed by a testicular regression at the end of summer and a subsequent recrudescence in autumn. Monthly variations in the epididymis, the ductus deferens and the sexual segment of the kidney were in synchrony with the testicular cycle. They were hypertrophied as spermatogenetic activity increased and atrophied as spermatogenetic activity decreased. Males of O. elegans showed a vernal type of spermatogenesis with a close relationship between the evolution of the seminiferous tubules and the secondary sexual characters. 相似文献
