Indirect effects from climate‐driven changes in ecosystems that are remote from direct human activity pose challenges for ecological restoration. Significant and often indirect impacts on alpine ecosystems, the primary ecosystem under consideration in this article, threaten historical‐reference conditions and the viability of some species. The impetus for restoration is similar to projects involving more direct and proximate impacts, but the issues are more complicated in remote ecosystems. Restoration efforts in remote ecosystems might do more harm than good, and the effort required for effective restoration might be greater than easily justified given the shortfall of resources for restoring more heavily impacted ecosystems. The long duration and integration of impacts on remote landscapes pose a distinct set of challenges to restorationists. Intervening in remote ecosystems makes them less remote by definition (they are now affected by human agency). In this article, we examine scientific, technical, and moral issues and offer an initial model for assessing the appropriateness of restoring remote landscapes.  相似文献

It was shown that H1 type histones possess anticancer activity and could be utilized in therapy of acute myeloid leukemia. We developed an experimental pilot-scale technology of the recombinant histone H1.3 variant production. The downstream process includes acidic extraction of the target protein from the culture broth, ion-exchange, reversed-phase and size-exclusion chromatography, and freeze-drying. The accent was made on the reduction of bacterial endotoxin contamination of the active pharmaceutical ingredient. Highly efficient downstream strategy of the target protein depyrogenation is discussed in the paper. The developed technology allows the production of the protein with high yield (approx. 110 g per 1000 L of the culture broth) and of high purity (estimated productivity is 75–100 g/month). The activity and purity of the active pharmaceutical ingredients produced for clinical trials were confirmed by different tests including ultra performance liquid chromatography, size exclusion chromatography, ELISA, SDS–PAGE, gel-clot LAL-test. Clinical trials were started in the Russian Federation.  相似文献

Aim Despite the increasing pace of urbanization, little is known about how this process affects biodiversity globally. We investigate macroecological patterns of bird assemblages in urbanized areas relative to semi‐natural ecosystems. Location World‐wide. Methods We use a database of quantitative bird surveys to compare key assemblage structure parameters for plots in urbanized and semi‐natural ecosystems controlling for spatial autocorrelation and survey methodology. We use the term ‘urbanized’ instead of ‘urban’ ecosystems as many of the plots were not located in the centre of towns but in remnant habitat patches within conurbations. Results Some macroecological relationships were conserved in urbanized landscapes. Species–area, species–abundance and species–biomass relationships did not differ significantly between urbanized and non‐urbanized environments. However, there were differences in the relationships between productivity and assemblage structure. In forests, species richness increased with productivity; in both forests and open habitats, the evenness of species abundances declined as productivity increased. Among urbanized plots, instead, both species richness and the evenness of species abundances were independent of variation in productivity. Main conclusions Remnant habitats within urbanized areas are subject to many ecological alterations, yet key macroecological patterns differ remarkably little in urbanized versus non‐urbanized plots. Our results support the need for increased conservation activities in urbanized landscapes, particularly given the additional benefits of local experiences of biodiversity for the human population. With increasing urbanization world‐wide, broad‐scale efforts are needed to understand and manage the effects of this driver of change on biodiversity.  相似文献

A new concept for the improvement of the downstream processing and purification is the so‐called magnetic separation. By using surface functionalized magnetic substrate particles, which selectively adsorb the target product, it can be directly separated out of a crude bioprocess stream. These methods are already used for analytical purposes, where only small amounts of functionalized particles are necessary. To apply the same concept at a larger scale, effective and economical procedures have to be provided. First, suitable process equipment has to be developed. Second, the magnetic particles have to be manufactured with a stable surface functionalization and long‐term stability for their reuse. Up to now mainly high‐gradient magnetic separation filter devices are applied for selective magnetic separation. They consist of a magnetic matrix in which the magnetic particles are trapped. In this work, a new magnetic filter is introduced that overcomes the capacity limitations of the current high‐gradient magnetic separation technology. The principle is demonstrated by selective recovery of lysozyme from hen egg white. Prior to the separation experiments magnetic beads with a strong acid cation‐exchange surface functionalization are synthesized. The separation procedure is implemented in only one unit operation. With the implementation of the displacement elution sequence lysozyme can be separated out of a hen egg white solution with a purification factor of PF=36 and a purity P=0.83.  相似文献

The incorporation of feed-use (FU) amino acids (AAs) in diets results in a reduced use of protein-rich ingredients such as soybean meal, recognized to have elevated contributions to environmental impacts. This study investigated whether the incorporation of L-lysine.HCl, L-threonine and FU-methionine reduces the environmental impacts of pig and broiler feeds using Life Cycle Assessment. The following impact categories were considered: climate change, eutrophication, acidification, terrestrial ecotoxicity, cumulative energy demand and land occupation. Several feeds were formulated either to minimize the cost of the formulation (with or without AA utilization), to maximize AA incorporation (i.e. the cost of AA was considered to be similar to that of soybean meal), or to minimize greenhouse gas emissions. For both pig and broiler feeds, calculations were made first using only cereals and soybean meal as main ingredients and then using cereals and several protein-rich ingredients (soybean meal, rapeseed meal and peas). In addition, these calculations were performed using two types of soybean meal (from Brazil, associated with recent deforestation or not). For broiler feeds, two types of maize (from France, irrigated, with mineral fertilization v. not irrigated, with animal manure fertilization) were also tested. Regarding the feeds formulated to minimize cost, incorporation of AA decreased the values for eutrophication, terrestrial ecotoxicity and cumulative energy demand of both pig and broiler feeds, regardless of the base ingredients. Reduction in climate change and acidification due to the incorporation of AA depended on the nature of the feed ingredients, with the effect of AA incorporation being greater when combined with ingredients with high impacts such as soybean meal associated with deforestation. Feeds formulated to maximize AA incorporation generally had a similar composition to those formulated to minimize cost, suggesting that the costs of AA were not the limiting factor in their incorporation. Feeds formulated to minimize greenhouse gas emissions had the lowest values for climate change and cumulative energy demand, but not for other impacts. Further research is needed to elucidate whether the incorporation of additional AA (tryptophan and valine) along with L-lysine, L-threonine and FU-methionine could decrease on the environmental impacts of pig and broiler feeds further.  相似文献

Recently the possibility of producing the capsular polysaccharide K4, a fructosylated chondroitin, in fed-batch experiments was assessed. In the present study, a novel downstream process to obtain chondroitin from Escherichia coli K4 fermentation broth was developed. The process is simple, scalable and economical. In particular, downstream procedures were optimized with a particular aim of purifying a product suitable for further chemical modifications, in an attempt to develop a biotechnological platform for chondroitin sulfate production. During process development, membrane devices (ultrafiltration/diafiltration) were exploited, selecting the right cassette cut-offs for different phases of purification. The operational conditions (cross-flow rate and transmembrane pressure) used for the process were determined on an ?KTA cross-flow instrument (GE Healthcare, USA), a lab-scale automatic tangential flow filtration system. In addition, parameters such as selectivity and throughput were calculated based on the analytical quantification of K4 and defructosylated K4, as well as the major contaminants. The complete downstream procedure yielded about 75% chondroitin with a purity higher than 90%.  相似文献

Human transformations of the Wadden Sea ecosystem through time: a synthesis   总被引:3,自引:3,他引:0  
Todays Wadden Sea is a heavily human-altered ecosystem. Shaped by natural forces since its origin 7,500 years ago, humans gradually gained dominance in influencing ecosystem structure and functioning. Here, we reconstruct the timeline of human impacts and the history of ecological changes in the Wadden Sea. We then discuss the ecosystem and societal consequences of observed changes, and conclude with management implications. Human influences have intensified and multiplied over time. Large-scale habitat transformation over the last 1,000 years has eliminated diverse terrestrial, freshwater, brackish and marine habitats. Intensive exploitation of everything from oysters to whales has depleted most large predators and habitat-building species since medieval times. In the twentieth century, pollution, eutrophication, species invasions and, presumably, climate change have had marked impacts on the Wadden Sea flora and fauna. Yet habitat loss and overexploitation were the two main causes for the extinction or severe depletion of 144 species (~20% of total macrobiota). The loss of biodiversity, large predators, special habitats, filter and storage capacity, and degradation in water quality have led to a simplification and homogenisation of the food web structure and ecosystem functioning that has affected the Wadden Sea ecosystem and coastal societies alike. Recent conservation efforts have reversed some negative trends by enabling some birds and mammals to recover and by creating new economic options for society. The Wadden Sea history provides a unique long-term perspective on ecological change, new objectives for conservation, restoration and management, and an ecological baseline that allows us to envision a rich, productive and diverse Wadden Sea ecosystem and coastal society.  相似文献

Non-indigenous species (NIS) have been called biological pollutants, which implies that reducing their numbers should reduce negative impacts. To test this hypothesis, we used food web models, parameterized with data from field studies, to ask how reducing the number of NIS co-occurring with endangered salmon would affect salmon mortality. Our analyses indicate that predation on Upper Columbia River spring chinook salmon Oncorhynchus tshawytscha and steelhead O. mykiss juveniles was affected very little by NIS reduction. The effects of removing NIS were partly or totally offset by indirect food web interactions, and were subtle compared to effects of native predator management. We predict that the most effective way of reducing predation on salmon smolts will involve managing native predators and targeted removals of specific NIS. Minimizing impact of established NIS thus entails not only reducing NIS prevalence, but also considering background management practices and community context.  相似文献

This study addresses the impact of small-scale gold mining on the environment and communities of the Puyango River basin in the southwest of Ecuador between 1999 and 2001. Our primary objectives were to measure mercury, manganese, and lead in the river, to identify pathways of population exposure, and biological indicators of human exposure. A multi-method design at the catchment scale was applied to the analysis of water, sediments, and particulate matter for mercury, lead, and manganese over two different seasons. A household survey was undertaken, and individuals were assessed for lead (blood samples), and for mercury (urine and hair samples). River water samples had high concentrations of particulate matter (700 mg/L). Mercury and lead levels in water at the gold processing plants were very high in both seasons (250 ng Hg/L and 160 μg Pb/L). Mercury and lead, there was a downstream gradient with the areas adjacent to the gold processing plants having the highest levels. In Portovelo, the Upper Basin city, 14% of the population reported occupational exposure to inorganic mercury. Although no one in Portovelo consumes water from the river, 10% of the population consume local fish. This contrasts the Lower Basin where 98% of the population consume fish, and 100% use river water for drinking and cooking. Lead blood levels over 20 μg/dl were found in 39.4% of the study population and blood mercury over 10 μg/L only in 10.0%. Urinary mercury over 4 μg/L was found in 66.5% of the examined persons. Hair methylmercury did not exceed the safe limit of 2 μg/g, the mean concentration was 1.2 μg/g. Mining activity and erosion contributes to heavy-metal contamination (mercury, lead, and manganese) throughout the Puyango Basin. The relation between environmental contamination is complex and further research is being conducted to understand these relations.  相似文献

Commercial forests in many parts of the world are deficient in nitrogen and phosphorus. These nutrient-deficient forests often exist in close proximity to large animal feeding operations, meat processing and other food, textile, or other biomass-processing plants, and municipal waste treatment facilities. Many of these facilities produce large surpluses of nitrogen, phosphorus, and organic matter as gaseous ammonia, urea, uric acid, phosphorus compounds, bacterial sludges, and partially treated municipal wastewaters. These co-existing and substantial nutrient deficiencies and surpluses offer ready-made opportunities for discovery, demonstration, and commercial development of science-based, technology-facilitated, environmentally sound, economically viable, and socially acceptable "win-win alliances" among these major industries based on the principles of industrial ecology and sustainable development. The major challenge is to discover practical means to capture the surplus nutrients and put them to work in forest stands from which value-added products can be produced and sold at a profit.  相似文献
