A cellular lineage analysis of the chick limb bud 总被引:2,自引:1,他引:1
The chick limb bud has been used as a model system for studying pattern formation and tissue development for more than 50 years. However, the lineal relationships among the different cell types and the migrational boundaries of individual cells within the limb mesenchyme have not been explored. We have used a retroviral lineage analysis system to track the fate of single limb bud mesenchymal cells at different times in early limb development. We find that progenitor cells labeled at stage 19-22 can give rise to multiple cell types including clones containing cells of all five of the major lateral plate mesoderm-derived tissues (cartilage, perichondrium, tendon, muscle connective tissue, and dermis). There is a bias, however, such that clones are more likely to contain the cell types of spatially adjacent tissues such as cartilage/perichondrium and tendon/muscle connective tissue. It has been recently proposed that distinct proximodistal segments are established early in limb development; however our analysis suggests that there is not a strict barrier to cellular migration along the proximodistal axis in the early stage 19-22 limb buds. Finally, our data indicate the presence of a dorsal/ventral boundary established by stage 16 that is inhibitory to cellular mixing. The tail of the earliest known articulated fully skeletonized vertebrate, the arandaspid Sacabambaspis from the Ordovician of Bolivia, is redescribed on the basis of further preparation of the only specimen in which it is most extensively preserved. The first, but soon discarded, reconstruction, which assumed the presence of a long horizontal notochordal lobe separating equal sized dorsal and ventral fin webs, appears to have considerable merit. Although the ventral web is significantly smaller than the dorsal one, the presence of a very long notochordal lobe bearing a small terminal web is confirmed. The discrepancy in the size of the ventral and dorsal webs rather suggests that the tail was hypocercal, a condition that would better accord with the caudal morphology of the living agnathans and the other jawless stem gnathostomes. 相似文献

摘要 目的:探讨纳布啡鞘内注射对糖尿病神经痛(diabetic neuropathic pain, DNP)模型大鼠行为能力及背根神经节瞬时受体电位V1(transient receptor potential V1,TRPV1)表达的影响。方法:将糖尿病神经痛模型大鼠(n=48)随机平方为三组-模型组、纳布啡1组与纳布啡2组,每组16只。纳布啡1组与纳布啡2组分别给予纳布啡鞘内注射0.5 μg/10 μL与1.0 μg/10 μL,模型组给予注射等剂量的0.9 %氯化钠溶液,每天1次。分别于治疗第7 d、第14 d,采用血糖仪测定与记录空腹血糖(fasting blood glucose,FBG)水平并进行机械痛阈检测;治疗第14 d、第28 d,采用酶联免疫法检测血清IL-6与TNF-α含量,采用免疫印迹法检测背根神经节TRPV1蛋白的相对表达。结果:模型组、纳布啡1组与纳布啡2组治疗第7 d、第14 d的空腹血糖水平都高于20.00 mmol/L,组内与组间对比差异不具有统计学意义(P>0.05)。纳布啡1组与纳布啡2组治疗第7 d、第14 d的机械痛阈高于模型组(P<0.05),也高于治疗前(P<0.05),纳布啡2组与纳布啡1组差异具有统计学意义(P<0.05)。纳布啡1组与纳布啡2组治疗第14 d、第28 d的血清白细胞介素(Interleukin,IL)-6与肿瘤坏死因子(Tumor necrosis factor,TNF)-α含量明显低于模型组(P<0.05),纳布啡2组明显低于纳布啡1组(P<0.05)。纳布啡1组与纳布啡2组治疗第14 d、第28 d的背根神经节TRPV1相对表达水平明显低于模型组(P<0.05),纳布啡2组明显低于纳布啡1组(P<0.05)。结论:纳布啡鞘内注射在糖尿病神经痛模型大鼠的应用能改善行为能力,抑制背根神经节TRPV1的表达,还可抑制血清IL-6与TNF-α的释放,从而发挥镇痛治疗效用。 相似文献

The human cervical spine supports substantial compressive load in vivo. However, the traditional in vitro testing methods rarely include compressive loads, especially in investigations of multi-segment cervical spine constructs. Previously, a systematic comparison was performed between the standard pure moment with no compressive loading and published compressive loading techniques (follower load – FL, axial load – AL, and combined load – CL). The systematic comparison was structured a priori using a statistical design of experiments and the desirability function approach, which was chosen based on the goal of determining the optimal compressive loading parameters necessary to mimic the segmental contribution patterns exhibited in vivo. The optimized set of compressive loading parameters resulted in in vitro segmental rotations that were within one standard deviation and 10% of average percent error of the in vivo mean throughout the entire motion path. As hypothesized, the values for the optimized independent variables of FL and AL varied dynamically throughout the motion path. FL was not necessary at the extremes of the flexion–extension (FE) motion path but peaked through the neutral position, whereas, a large negative value of AL was necessary in extension and increased linearly to a large positive value in flexion. Although further validation is required, the long-term goal is to develop a “physiologic” in vitro testing method, which will be valuable for evaluating adjacent segment effect following spinal fusion surgery, disc arthroplasty instrumentation testing and design, as well as mechanobiology experiments where correct kinematics and arthrokinematics are critical. 相似文献

Understanding changes in lumbar spine (LS) angles and intervertebral disc (IVD) behavior in end-range positions in healthy subjects can provide a basis for developing more specific LS models and comparing people with spine pathology. The purposes of this study are to quantify 3D LS angles and changes in IVD characteristics with end-range positions in 3 planes of motion using upright MRI in healthy people, and to determine which intervertebral segments contribute most in each plane of movement. Thirteen people (average age = 24.4 years, range 18–51 years; 9 females; BMI = 22.4 ± 1.8 kg/m2) with no history of low back pain were scanned in an upright MRI in standing, sitting flexion, sitting axial rotation (left, right), prone on elbows, prone extension, and standing lateral bending (left, right). Global and local intervertebral LS angles were measured. Anterior-posterior length of the IVD and location of the nucleus pulposus was measured. For the sagittal plane, lower LS segments contribute most to change in position, and the location of the nucleus pulposus migrated from a more posterior position in sitting flexion to a more anterior position in end-range extension. For lateral bending, the upper LS contributes most to end-range positions. Small degrees of intervertebral rotation (1–2°) across all levels were observed for axial plane positions. This paper addresses the design of a biomimetic fish robot actuated by piezoeeramic actuators and the effect of artificial caudal fins on the fish robot's performance. The limited bending displacement produced by a lightweight piezocomposite actuator was amplified and transformed into a large tail beat motion by means of a linkage system. Caudal fins that mimic the shape of a mackerel fin were fabricated for the purpose of examining the effect of caudal fm characteristics on thrust production at an operating frequency range. The thickness distribution of a real mackerel's fin was measured and used to design artificial caudal fins. The thrust performance of the biomimetic fish robot propelled by fins of various thicknesses was examined in terms of the Strouhal number, the Froude number, the Reynolds number, and the power consumption. For the same fm area and aspect ratio, an artificial caudal fin with a distributed thickness shows the best forward speed and the least power consumption. 相似文献

The protocol described in this paper offers a simple and rapid method for PCR analysis of transgenes using a restricted amount of fin tissue from small-sized transgenic fish. A simple preparation of fin lysate using a buffer containing a low concentration of an ionic detergent, SDS (0.01%), followed by neutralization with a second buffer containing higher concentrations of non-ionic detergents NP40 (2%) and Tween 20 (2%) consistently provides a reliable quantity of high-quality DNA template for PCR amplification of transgenes. Based on this protocol, transgenic fish can be clearly distinguished from non-transgenic fish using PCR in a rapid and reproducible manner. WNTs are secreted signaling molecules which control cell differentiation and proliferation. They are known to play essential roles in various developmental processes. Wnt genes have been identified in a variety of animals, and it has been shown that their amino acid sequences are highly conserved throughout evolution. To investigate the role of wnt genes during fish development from the evolutionary viewpoint, six medaka wnt genes (wnt4, wnt5a, wnt6, wnt7b, wnt8b and wnt8-like) were isolated and their embryonic expression was examined. These wnt genes were expressed in various tissues during embryonic development, and most of their expression patterns were conserved or comparable to those of other vertebrates. Thus, these wnt genes may be useful as molecular markers to investigate development and organogenesis using the medaka. Focus was on wnt5a, which was expressed in the pectoral fin buds, because its expression pattern was particularly comparable to that in tetrapod limbs. Its detailed expression pattern was further examined during pectoral fin bud development. The conservation and diversification of Wnt5a expression through the evolutionary transition from fish fins to tetrapod limbs is discussed. 相似文献
