Kinetics of spermatogonia as well as localization in niches have been described in rodents, but rarely in large animals or in species of economical interest. In this regard, and envisioning the possibility of spermatogonial transplantation from donkeys (Equus asinus) to mules (Equus mulus mulus), many variables that may contribute for an enhanced understanding of the spermatogonial biology in donkeys were investigated. Testes from five adult donkeys were routinely processed for high-resolution light microscopy. Donkey seminiferous epithelium can be divided in XII stages based on the development of the acrosomal system. In addition, spermatogonial morphology and morphometric analysis were performed allowing the characterization of two groups of spermatogonia: undifferentiated (Aund) and differentiating (A1, A2, A3, B1 and B2). Aund spermatogonia were present along all XII stages of the seminiferous epithelium cycle of this species, whereas differentiating spermatogonia were only at specific stages. Number of differentiating spermatogonia gradually increased as the cycle progressed, despite the apparent rigid regulation of the balance between mitosis and apoptosis throughout the spermatogenic process. Understanding of spermatogonial biology and kinetics in donkeys, revealed that type Aund spermatogonia are located in specific microenvironments, the spermatogonial niches. The present results enhance understanding of spermatogonial biology in donkeys providing information about subtypes, morphology, number and mitosis/apoptosis along the seminiferous epithelium cycle.  相似文献

The aim of this work was to study the effects of dilution and centrifugation (i.e., two methods of reducing the influence of the seminal plasma) on the survival of spermatozoa and the structure of motile sperm cell subpopulations in refrigerated Catalonian donkey (Equus asinus) semen. Fifty ejaculates from nine Catalonian jackasses were collected. Gel-free semen was diluted 1:1, 1:5 or 1:10 with Kenney extender. Another sample of semen was diluted 1:5, centrifuged, and then resuspended with Kenney extender until a final dilution of 25 × 106 sperm/ml was achieved (C). After 24 h, 48 h or 72 h of refrigerated storage at 5 °C, aliquots of these semen samples were incubated at 37 °C for 5 min. The percentage of viable sperm was determined by staining with eosin-nigrosin. The motility characteristics of the spermatozoa were examined using the CASA system (Microptic, Barcelona, Spain). At 24 h, more surviving spermatozoa were seen in the more diluted and in the centrifuged semen samples (1:1 48.71%; 1:5 56.58%, 1:10 62.65%; C 72.40%). These differences were maintained at 48 h (1:1 34.31%, 1:5 40.56%, 1:10 48.52%, C 66.30%). After 72 h, only the C samples showed a survival rate of above 25%. The four known donkey motile sperm subpopulations were maintained by refrigeration. However, the percentage of motile sperms in each subpopulation changed with dilution. Only the centrifuged samples, and only at 24 h, showed exactly the same motile sperm subpopulation proportions as recorded for fresh sperm. However, the 1:10 dilutions at 24 and 48 h, and the centrifuged semen at 48 h, showed few variations compared to fresh sperm. These results show that the elimination of seminal plasma increases the survival of spermatozoa and the maintenance of motility patterns.The initial sperm concentration had a significant (P < 0.05) influence on centrifugation efficacy, but did not influence the number of spermatozoa damaged by centrifugation. In contrast, the percentage of live spermatozoa in the fresh semen significantly influenced the number of spermatozoa damaged by centrifugation, but not centrifugation efficacy.  相似文献

The results of a 2-year survey on ticks infesting livestock on farms in Western Sicily, Italy, are reported. Several dogs living in the same area were also examined for ticks. Moreover, free-living ticks from one cattle farm were caught by CO2 traps and flagging. A total of 6208 specimens was collected belonging to 9 species: Rhipicephalus bursa (32.4%), Rhipicephalus turanicus (22.7%), Rhipicephalus sanguineus (19.3%), Hyalomma lusitanicum (12.0%), Haemaphysalis punctata (7.8%), Hyalomma marginatum (3.5%), Dermacentor marginatus (1.4%), Ixodes ricinus (0.8%) and Ixodes hexagonus (0.1%). The species belonging to the genus Rhipicephalus were present on all hosts and habitats monitored, R. bursa was found to be abundant on cattle (46.3%) and sheep (62.0%), R. sanguineus was prevalent on dogs (50.4%), and R. turanicus on donkeys (41.0%) and as free-living species (48.5%).  相似文献

This study provides basic data on the normal structure of the inferior olivary complex (IOC) of the donkey, Equus asinus, at the light microscopic level. In common with that of other mammals, the donkey IOC consisted of three major nuclei and four minor groups of cells. The former was comprised of the medial and dorsal accessory olives (MAO and DAO, respectively) and the principal olive (PO), and the latter was comprised of the dorsal cap, nucleus beta, ventrolateral outgrowth and dorsomedial cell column. The MAO had the longest rostral to caudal representation and formed the caudal pole of IOC. The DAO was located dorsally to the MAO in the caudal half of the IOC. In the rostral half, the DAO bended ventrally and merged with the dorsal lamella of PO. More rostrally, the DAO lost its connection with the dorsal lamella and then conversely connected with the ventral lamella of PO. The DAO formed the rostral pole of the IOC. The PO extended through the rostral half of the IOC. The dorsal cap was a small group of cells. Overall, the donkey IOC is similar to that of other mammals.  相似文献

Sixty-three embryos were recovered out of 83 estrous cycles (75.9%) and 98 ovulations (64.3%) of five Pantesca jennies, 2 to 5 yr old, naturally mated or artificially inseminated with fresh semen. Embryo recovery rate was influenced by number of ovulations per cycle (133% and 63% for double and single ovulations, respectively), by the day of embryo recovery attempt (12%, 83%, and 75% at Days 7, 8, and 9 after ovulation, respectively), and by the repetition of the embryo recovery attempt on successive cycles (60%, 79%, and 100% for cycles 1 to 7, 8 to 14, and 15 to 24, respectively). All recovered embryos but three were classified as good or excellent. Of 58 nonsurgical embryo transfers to Ragusana jenny recipients, 13 (22.4%), 10 (17.2%), and 9 (15.5%) resulted in a pregnancy at Days 14, 25, and 50, respectively. Recipients’ pregnancy rate was not influenced by the evaluated parameters: embryo quality and age, media employed to wash embryos, days after ovulation of the recipient, experience of the operator. Between 14 and 50 d of pregnancy, 4 of 13 (30.7%) embryos were lost with an influence of the days from ovulation of the recipient: recipients at Days 5 or 6 kept all pregnancies (N = 7), whereas recipients at Days 7 or 8 lost 3 of 4 pregnancies, as one of the two recipients at Day 3. More studies are needed before embryo transfer could be considered a reliable tool to preserve endangered donkey breeds.  相似文献

This paper investigated gestation length and estrus cycle characteristics in three different Spanish donkey breeds (Andalusian, Zamorano-Leones, and Catalonian) kept on farm conditions in southern Spain, using data for ten consecutive breeding seasons. Gestation length was measured in 58 pregnancies. Ovarian ultrasonography was used to detect the ovulation, in order to ascertain true gestation length (ovulation-parturition). Pregnancy was diagnosed approximately 14-18 d after ovulation and confirmed on approximately day 60. Average gestation length was 362 ±15.3 (SD) d, and no significant differences were observed between the three different breeds. Breeding season had a significant effect (P < 0.01), with longer gestation lengths when jennies were covered during the early period. Breed, age of jenny, year of birth, foal gender, month of breeding, and type of gestation had no significant effect on gestation length.After parturition, foal-heat was detected in 53.8% of the postpartum cycles studied (n = 78), and ovulation occurred on day 13.2 ± 2.7. The duration of foal-heat was 4.7 ±1.7 d, with a pregnancy rate of 40.5%.When subsequent estrus cycles were analyzed, the interovulatory interval (n = 68) and estrus duration (n = 258) were extended to a mean 23.8 ± 3.5 and 5.7 ± 2.2 d, respectively. Both variables were influenced by the year of study (P < 0.03 and P < 0.001), whereas month and season of ovulation (P < 0.005 and P < 0.009, respectively) affected only interovulatory intervals. Estrus duration was significantly longer than that observed at the foal-heat (P < 0.006), and the pregnancy rate was 65.8%.This study provides reference values for true gestation length and estrus cycle characteristics in Spanish jennies. Breeding season affected gestation length in farm conditions. Also, seasonal influence was observed on the length of the estrus cycle (i.e., interovulatory interval), although foal-heat was not affected by environmental factors.  相似文献

Donkey milk, similar to human milk in compositions, has been suggested as the best potential hypoallergenic replacement diet for babies suffering from cow milk protein allergens and a promising nutraceutical for aged people. In this study, label-free mass spectrometry analysis was conducted to quantitatively identify the whey proteins differentially expressed in high-milk-yield samples compared with low-milk-yield samples. A total of 216 whey proteins were identified, and 19 of them showed significant differences in high-milk-yield samples. Of these proteins, 16 were upregulated and 3 were downregulated. Differentially expressed proteins (DEPs) were subjected to intensive bioinformatic analysis. Results revealed that the majority of DEPs participated in protein processing in endoplasmic reticulum, estrogen signaling pathway, progesterone-mediated oocyte maturation, and PI3K-Akt signaling pathway. Functional protein analysis suggested that proteins functioned in binding, catalytic activity, molecular function regulation, structural molecule activity, and transporter activity. Our study was the first to analyze the whey protein profile of different samples of donkey milk and to identify candidate proteins that could be used to explore the molecular mechanism related to the yield traits of Dezhou donkey milk. This study provided the biomarkers for the selection of high-milk-yielding donkey and obtained valuable information for future studies.  相似文献

Sperm quality in donkeys (Equus asinus) after freezing thawing is still considered lower than that from other animals, including horses. The aim of this study was to test a new freezing extender supplemented with jenny colostrum on donkey sperm. After thawing, we evaluated sperm motility by means of computer-assisted analysis, viability by SYBR-14 and propidium iodide (PI), membrane functional integrity by HOS-test and acrosome integrity by isothiocyanate conjugated with peanut agglutinin (FITC-PNA) and PI. Ejaculates were collected from five fertile Donkeys. Sperm samples were pooled, diluted and cryopreserved into three experimental extender groups: BotuCrio®, lactose-extender supplemented with egg yolk (20%) and lactose-extender supplemented with jenny colostrum (20%). The results demonstrated that lactose-jenny colostrum samples displayed significantly higher values in almost all parameters evaluated (p < 0.05) compared with the other two extenders after thawing (BotuCrio® and lactose-egg yolk based extender, respectively) –Total Motility, Viability, HOS test, VCL, VSL and VAP. Acrosome status, LIN, STR and WOB despite showing lower values, none of them were statistically significant (p > 0.05). In conclusion, the extender containing jenny colostrum can be successfully used for donkey semen crypreservation and could effectively improve donkey sperm qualities after freezing -thawing.  相似文献

Embryo transfer (ET) in the donkey resulted in a very low recipient pregnancy rates. The aim of these studies was to investigate if nonsurgical transfer techniques or donkey embryo quality affect donkey recipient pregnancy failure. In Study 1, the impact of transfer technique was investigated by evaluating if cervical catheterization is associated with prostaglandin release and suppression of luteal function and if donkey recipients would become pregnant after nonsurgical transfer of horse embryos. Four jennies, from 5 to 8 d after ovulation, were submitted to a sham transcervical ET and to evaluation of PGFM and progesterone plasma concentrations. Five 8 d horse embryos were nonsurgically transferred into synchronized donkey recipients (HD). Cervical stimulation caused a transient PGF release in two of four jennies in the absence of a significant decrease in progesterone plasma concentration. All transferred horse embryos resulted in pregnancies in the jenny recipients. In Study 2, donkey embryo viability was investigated by 1.2 meters, 6-diamidino-2-phenylindole (DAPI) staining of 10 embryos and by the transfer of 6 and 12 donkey embryos in synchronized mare (DH) and donkey (DD) recipients, respectively, of known fertility. The estimated proportion of dead cells in DAPI stained embryos was 0.9% (range 0-3.9%) and below what is considered normal (20%) for horse embryos. Three of six and six of 12 of the DH and DD ETs, respectively resulted in pregnancies at 14 and 25 d (50%), a higher pregnancy rate than previously reported after DD ET. The overall results of this study suggest that the transcervical technique for ET and donkey embryo viability are not the reasons for the low pregnancy rates that have previously been described in donkey recipients, and that nonsurgical ET in donkeys can result in acceptable results.  相似文献

In mammals, the epididymis has numerous interrelated functions including absorptive and secretory activity that affect luminal environment and cell membrane, and the maturation and storage of sperm. Spermatozoa acquire their motility and fertilizing ability during their passage through the epididymis and the motility of epididymal spermatozoa should be a balance between the maturation of flagellum and the inhibition of the flagellar machinery. In this study maturational change in sperm characteristics were evaluated in the epididymis of donkey. Spermatozoa collected from four portions of the epididymis (head, cranial corpus, caudal corpus, tail) were compared before and after ejaculation for viability, mitochondrial activity, kinetic parameters, and morphology. A significant increase in the mitochondrial activity along the epididymis was reported, suggesting a possible involvement in the motion mechanism. This should be corroborated by the significant correlation between mitochondrial activity and the total and progressive motility and the increase in velocities of spermatozoa recorded by computer-assisted sperm analysis. The percentage of most of the abnormal spermatozoa were similar in all tracts, with a great variability between jackasses. Only the bent midpiece percentage decreased significantly along epididymis. A significant increase in the percentage of distal cytoplasmic droplets (DCD), and a simultaneous decrease in the proximal cytoplasmic droplets (PCD), was found. The DCD fell down after ejaculation suggesting the late loss of the cytoplasmic residual (DCD) in the donkey, as hypothesized in the stallion. Because the prevalence of PCD were similar in both tail epididymal and ejaculated spermatozoa, a defect of the maturative process in the PCD sperm should be speculated.  相似文献
