Administration of an aqueous extract of the calcinogenic plant (S.m.) to vitamin D-deficient or strontium fed chicks produces significant plasma 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3) activity within 6 hr. (via radioreceptor assay) and subsequently elicits the appearance of immunoreactive intestinal calcium binding protein. Studies of a purified aqueous extract of S.m. show that it does not compete effectively with radioactive 1,25-(OH)2D3 for binding to the sterol's intestinal receptor. However, treatment of the extract with β-glucosidase releases a biologically active substance which is soluble in organic solvents and efficiently competes with labeled sterol for the receptor. This factor migrates exactly with tritiated 1,25-(OH)2D3 on high resolution Celite liquid-liquid partition columns. Thus, S.m. contains a molecule very similar or identical to 1,25-(OH)2D3 which is combined with one or more carbohydrate moieties in the native plant. Apical ectodermal ridges (AERs) isolated from 3- to 4-day chick and quail embryos were prepared by means of trypsinization and microdissection and then were grafted to the dorsal or ventral side of a host chick wing bud. They induced supernumerary limb outgrowths from the host bud showing, respectively, a bidorsal or biventral organization, as determined by the patterns of feather germs. The grafted ridge cells persisted, as revealed by histological sections of supernumerary chick limb parts growing under the influence of quail AERs, whose cells are readily distinguished after application of the Feulgen reagent.These results show that the AER induces limb outgrowth regardless of whether it is associated with dorsal or ventral limb ectoderm and that its continued existence is not dependent on contributions of ectodermal cells from the opposed ectodermal faces of the limb bud. The AER is pictured as maintaining the subjacent mesoderm in a condition of developmental plasticity without specifying its differentiation with respect to the proximodistal axis. It remains uncertain whether the positional values of cells that develop under the influence of the AER arise within these cells themselves or appear in response to influences from proximal sources. 相似文献

Division of labour is of fundamental importance for the success of societies, yet little is known about how individual specialization affects the fitness of the group as a whole. While specialized workers may be more efficient in the tasks they perform than generalists, they may also lack the flexibility to respond to rapid shifts in task needs. Such rigidity could impose fitness costs when societies face dynamic and unpredictable events, such as an attack by socially parasitic slavemakers. Here, we experimentally assess the colony-level fitness consequences of behavioural specialization in Temnothorax longispinosus ants that are attacked by the slavemaker ant T. americanus. We manipulated the social organization of 102 T. longispinosus colonies, based on the behavioural responses of all 3842 workers. We find that strict specialization is disadvantageous for a colony''s annual reproduction and growth during slave raids. These fitness costs may favour generalist strategies in dynamic environments, as we also demonstrate that societies exposed to slavemakers in the field show a lower degree of specialization than those originating from slavemaker-free populations. Our findings provide an explanation for the ubiquity of generalists and highlight their importance for the flexibility and functional robustness of entire societies. 相似文献

The lethal mutagenesis hypothesis states that within-host populations of pathogens can be driven to extinction when the load of deleterious mutations is artificially increased with a mutagen, and becomes too high for the population to be maintained. Although chemical mutagens have been shown to lead to important reductions in viral titres for a wide variety of RNA viruses, the theoretical underpinnings of this process are still not clearly established. A few recent models sought to describe lethal mutagenesis but they often relied on restrictive assumptions. We extend this earlier work in two novel directions. First, we derive the dynamics of the genetic load in a multivariate Gaussian fitness landscape akin to classical quantitative genetics models. This fitness landscape yields a continuous distribution of mutation effects on fitness, ranging from deleterious to beneficial (i.e. compensatory) mutations. We also include an additional class of lethal mutations. Second, we couple this evolutionary model with an epidemiological model accounting for the within-host dynamics of the pathogen. We derive the epidemiological and evolutionary equilibrium of the system. At this equilibrium, the density of the pathogen is expected to decrease linearly with the genomic mutation rate U. We also provide a simple expression for the critical mutation rate leading to extinction. Stochastic simulations show that these predictions are accurate for a broad range of parameter values. As they depend on a small set of measurable epidemiological and evolutionary parameters, we used available information on several viruses to make quantitative and testable predictions on critical mutation rates. In the light of this model, we discuss the feasibility of lethal mutagenesis as an efficient therapeutic strategy. 相似文献

Hulecoeteomyia japonica japonica (= Aedes japonicus japonicus) (Diptera: Culicidae) (Theobald 1901), a container‐breeding invasive species in North America and Europe, is attracting particular attention for its high local abundances and possible roles in the transmission of human and animal pathogens. The preferential habitats of this species are forested and bushy areas, which renders control measures extremely inefficient. Use of the sterile insect technique (SIT) may contribute to the implementation of area‐wide integrated pest management strategies, as has been successfully proven with other aedine mosquito species. The present study investigates the effects of irradiation at a dose of 40 Gy on fitness parameters in H. j. japonica. Irradiation was performed on 16–24‐h‐old pupae from a colonized strain (PA) using a TrueBeam linear accelerator. Males from the PA strain were crossed with females of the same colony or with field‐collected females. Irradiation induced a slight increase in mortality in male pupae, but did not alter the survival and mating abilities of emerging adult males. Rates of blood feeding and fertility were lower when PA strain males were kept with field‐collected females rather than PA females. Irradiated males induced reductions in fertility (residual fertility: 2.6%) and fecundity in mated females. The data indicate that the SIT is a suitable technique to enhance the control of this species. 相似文献

Fitness traits of three Drosophila simulans strains infected by endocellular bacteria belonging to the genus Wolbachia have been compared with those of replicate stocks previously cured from the infection by an antibiotic treatment. The traits measured were development time, egg-to-adult viability, egg hatch, productivity, fecundity, and the number of functional ovarioles. Individuals of the first strain were bi-infected by two Wolbachia variants, wHa and wNo. The second strain was infected by wHa, the third one by wNo. The Wolbachia studied here cause cytoplasmic incompatibility (CI), a high embryonic mortality (70% to > 90%) when an infected male is crossed with an uninfected female. Three generations after antibiotic treatment, we observed in all strains a significant drop in productivity in the cured stocks. This drop was not due to antibiotic toxicity and was associated with the loss of the Wolbachia. However the effect had disappeared in two of the three strains five generations after treatment, and could not be found in the third strain in a third measurement carried out 14 generations after treatment. The temporary nature of the productivity difference indicates that Wolbachia do not enhance productivity in infected strains. On the other hand, in all traits measured, our results show the absence of any negative effects of the Wolbachia on their host. This could be explained when considering Wolbachia evolution, as maternally transmitted parasites bear a strong selective pressure not to harm their female host. However, CI would allow the bacteria to be maintained even when harming the female. The apparent absence of deleterious effects caused by these Wolbachia might result from a trade-off, where a relatively low bacteria density would advantage the Wolbachia by suppressing any deleterious effects on the female host, at the cost of a weaker maternal transmission rate of the infection. 相似文献
