A new cyanogenic glucoside, isolated from pods of Acacia sieberiana var. woodii, was shown to be (2R)-2- (β-d-glucopyranosyloxy)-3-hydroxy-3-methylbutanenitrile by spectroscopic and chemical methods. The absolute configuration of this glucoside was correlated with that of proacacipetalin by oxymercuration of the latter, followed by borohydride reduction of the product. 相似文献

Biosynthesis of the C-terminal carboxamide group of peptide hormones was studied using comparatively pGlu-His-Pro-Gly and Glu-His-Pro-Gly-Lys-Arg as putative precursors of the tripeptide, thyroliberin (TRH). Rat hypothalamus granules were found to contain an amide group forming activity which converts both peptide substrates into TRH. Comparison of the rate of conversion of the two substrates indicated that the C-terminal dibasic extension favored a 10-fold increase in the production of amidated peptide. It is suggested that this type of structure may be present in the putative biosynthetic precursor of TRH and that it may provide a better substrate for the enzyme(s) involved in C-terminal amidation. 相似文献

The fine structure of the organic matrix of the shore crab cuticle (Carcinus maenas L.), observed in transmission electron microscopy, reveals three different levels of organization of the chitin—protein complex. The highest level corresponds to the ‘twisted plywood’ organization described by Bouligand (1972). Horizontal microfibrils, parallel to the cuticle plane, rotate progressively from one level to another. When viewed in oblique section this structure gives superimposed series of nested arcs, visible in light microscopy or at the lowest magnifications of the electron microscope, in all the chitin-protein layers. At the highest magnifications of the electron microscope and with the best resolution, when the ultrathin sections are exactly transverse to the microfibril, a constant pattern can be observed which consists of rods transparent to electrons, which are embedded in an electron-opaque matrix. In cross-section, these rods often form more or less hexagonal arrays. We call a microfibril one rod and the adjacent opaque material, and question the usual interpretation of the microfibril molecular structure. Between these two levels of organization, there is an intermediate level, which corresponds to the grouping of microfibrils. Microfibrils form a dense structure, with few free spaces in the membranous layer, the deepest and non-calcified layer of the cuticle. In other parts of the cuticle, microfibrils are grouped into fibrils of various diameters or form a reticulate structure, the free spaces of the organic matrix being occupied by the mineral. 相似文献

A vinculin-like protein was identified in chicken as well as in bovine platelets by ELISA competitive binding assay using antibodies against vinculin from chicken gizzard. By a modified procedure (J. Biol. Chem. (1980) 255, 1194–1199) we succeeded in isolating bovine platelet vinculin to apparent homogeneity. The structural identity of platelet and chicken gizzard vinculin was demonstrated by circular dichroism analysis. It was also shown that platelet vinculin induces a significant decrease in the low shear viscosity of F-actin. Vinculin, in all probability, plays an important role in the organization of actin filaments in platelets, especially in the linkages of microfilaments to the membrane. 相似文献

All-trans retinoic acid increased the incorporation of D-[3H]galactose into particulate and soluble glycoproteins in the epidermis of cultured pig skin slices nearly two-fold. Increased incorporation of D-[3H]galactose was not blocked by tunicamycin. This effect was specific for D-[3H]galactose since the incorporation of D-[3H]glucosamine and L-[14C]leucine into epidermal glycoproteins was unaffected by all-trans retinoic acid. All-trans retinoic acid and 13-cis retinoic acid had quantitatively similar effects on D-[3H]galactose incorporation. All-trans retinyl acetate and an aromatic retinoic acid analogue (‘Etretinate’) were less effective. SDS polyacrylamide gel electrophoresis and fluorography showed increased incorporation of D-[3H]galactose into all epidermal glycoproteins in the presence of all-trans retinoic acid. Subtypes of H1 histone vary in primary structure, and the higher organisms that have been studied each seem to have about a half-dozen subtypes. The proportions of these subtypes vary with the progress of differentiation as seen in embryonic development, hormonally induced changes, spermatogenesis, and terminal differentiation. The H1 subtypes differ among themselves in their ability to condense DNA and small chromatin fragments. The gel to liquid crystal phase transition of dipalmitoylphosphatidylcholine (DPPC) has been followed by the change in absorbance at 400 nm; this change is due to the change in lipid light scattering properties during the transition. The effect of sucrose on the change in absorbance during the transition of DPPC has been investigated. It has been shown that the presence of sucrose or glycerol in the multilamellar liposome suspension increases the change in absorbance due to the main transition, decreases the total absorbance, and decreases the change in absorbance due to the pretransition. This effect of sucrose and glycerol is shown to be an optical effect which is correlated with solvent index of refraction. 相似文献

The aim of this study was to determine the effect of consuming alcoholic vs. nonalcoholic beverages on performance of psycho-technical tasks (attentional and general nonverbal intelligence tasks) and social behavior at different times of day. Both alcoholic and nonalcoholic consumption took place in a largely festive situation. The experiment was conducted on 184 degree-level and postgraduate students (94 female and 90 male) divided into eight independent groups for study at different times: 8:00 to 11:00, 11:00 to 14:00, 14:00 to 17:00, 17:00 to 20:00 h. The main result obtained, by analysis of variance (ANOVA), showed that time of day had no effect on the performance of psycho-technical tasks nor on social communication, except for the retest situation in the attentional task. Alcohol (equal to ?0.5 g/L of blood) facilitated communication, but basically it had no effect on any of the psycho-technical performance tests. For the latter, an interaction was observed between when the test was done and type of beverage consumed. Alcohol appears to alter the expected change in performance in the retest situation. The results suggest that the body's sensitivity to a measured quantity of alcohol differs according to the cognitive processes involved. 相似文献

The rate of translocation of the hydrophobic ion dipicrylamine across planar lipid membranes formed from dipalmitoyllecithin in n-decane was determined by voltage jump relaxation experiments. The activation energy of the rate constant shows a change from a positive to a negative value at about 42°C near the main phase transition temperature of this lipid. Below this temperature, the rate constant was found to increase with decreasing temperature. This anomalous behaviour extends over a temperature range of at least 10 K and may be formally interpreted as an enhanced mobility of dipicrylamine in the solid state of the membrane. 相似文献
