Invertases of carnation petals. Partial purification, characterization and changes in activity during petal growth 总被引:1,自引:0,他引:1
Carnation ( Dianthus caryophyllus L. cv. White Sim) petals contained two distinct invertases (EC 3.2.1.26) based on chromatographic behavior on DEAE-cellulose. Both are soluble in 20 m M sodium phosphate buffer (pH 6.5) and exhibit acid pH optimum of 5.5. Extraction of a cell wall preparation from petals with 1 M NaCl released little additional activity. Furthermore, only traces of activity remained associated with the NaCl-extracted cell wall preparation. One of the soluble invertases, representing over 75% of the total activity, was partially purified by (NH4 )2 SO4 fractionation and sequential chromatography over diethylaminoethyl-cellulose, concanavalin-A sepharose and polyacrylamide P-200. The enzyme was purified 38-fold with a recovery of 12%. It had an apparent native molecular weight of 215 kDa. The partially purified invertase is a β-fructofuranosidase (EC 3.2.1.26) based on its specificity for sucrose. The Km for sucrose was 3.3 m M . Accumulation of reducing sugars and increased invertase activity during expansive petal growth indicates that sucrose is the major source of carbon for petal growth. 相似文献

Shoots which proliferated from shoot tip explants of Colorado White Simm carnation and Fantastic tomato on MS medium containing 5 mgl-1 benzyladenine were rooted and grown in vitro as microplants. Tomato microplants grown in medium with 5 gl-1 sucrose had less overall shoot and root growth than those with 10,20, or 30 gl-1 sucrose regardless of NAA level. Carnation shoot growth was reduced by 5 g l-1 sucrose but root growth was not affected except when no sucrose was supplied. Microplant height and rooting of carnation were maximal when grown in 20 gl-1 sucrose whereas tomato microplant growth was greatest with 30 gl-1 sucrose. Microplants of both species had reduced height and root growth when the MS nutrient salts were lowered to 25%, 50%, or 75% compared to full strength when sucrose was supplied at 5 gl-1. 相似文献

Pollination induced an immediate increase in ethylene production in Dianthus caryophyllus and Petunia hybrida. In Cymbidium, a lag of several hours was observed. In all three species, pollination induced premature flower senescence. Treatment of the stigmatic surface with aminoethoxyvinylglycine prior to pollination effectively blocked the increase in ethylene production and alleviated the detrimental effect of pollination on flower life.In all three tested species, octanoic and decanoic acids, when applied to the stigmatic surface, had no effect on ethylene production and flower life. In isolated Cymbidium lips placed with their cut base in solutions containing these fatty acids, no effects on red colouration, ethylene production, and ethylene forming enzyme activity were observed. In addition, ethylene sensitivity of isolated lips was not affected. The putative regulatory role of short-chain saturated fatty acids in (pollination-induced) flower senescence is discussed. 相似文献

Dianthus caryophyllus cv. Nelken was cultured in vitro under different ventilation rates (0.11, 0.21, 0.68 and 0.86 changes h−1). Ventilation modified the anatomical characteristics of shoots and leaves described for plants grown in non-ventilated vessels:
the cuticle became thicker, there was a decreased cell size and intracellular space size. Also, there were more photosynthetic
and supportive tissues, including thicker cell walls. Increased ventilation promoted in vitro hardening of micropropagated carnation shoots, and pushed the culture-induced phenotype closer to that of ex vitro acclimatized plants. Anatomical variability of in vitro-grown plants was demonstrated to be a consequence of ventilation.

Adventitious shoot formation was studied with leaf, stem and axillary bud explants of carnation (Dianthus caryophyllus L.). The shoot regeneration procedures were applicable for a wide range of cultivars and shoot regeneration percentages were high for all explant types. Using axillary bud explants, shoot regeneration efficiency was independent of the size of the bud and of its original position in the plant. In contrast, shoot regeneration from stem and leaf explants was strongly dependent on their original position on the plant. The most distal explants (just below the apex) showed the highest level of shoot regeneration. The adventitious shoot primordia developed at the periphery of the stem segment and at the base of leaf explants. In axillary bud, stem and leaf explants, shoot regeneration originated from node cells, located at the transition area between leaf and stem tissue. Moreover, a gradient in shoot regeneration response was observed, increasing towards the apical meristem.Abbreviations BA

Growth promotion of pistils in carnation buds ( Dianthus caryophyllus L., cv. White Sim) by 1-aminocyclopropane-1-carboxylic acid (ACC) can be reversed by administering silver thiosulphate via the stem or 2,5-norbornadiene in the ambient atmosphere.
