 Mature ovules of Dianthus (Caryophyllaceae) were histologically observed by clearing and serial sectioning to characterize the cells of the embryo sac. The results show that the mature embryo sac was located deep inside the hemitropous ovule due to thick nucellar tissue at the micropylar region. For the isolation of the embryo sacs, ovules were collected from ovaries of flowers 1 day after anthesis, and treated with an enzyme solution for digesting cell walls on a gyratory shaker. After 12 h of enzyme treatment, these ovules were dissected using a glass needle under an inverted microscope to release the embryo sacs. The embryo sacs, characterized by their specific size, were successfully released by these successive treatments. The viability of the embryo sacs was more than 80% as assessed with fluorescein diacetate staining. Fluorescent staining with 4,6-diamidino-2-phenylindole revealed the nuclei of the egg apparatus in the isolated embryo sacs. The procedure for isolating embryo sacs established in this study will offer a new approach to further in vitro studies on fertilization in Dianthus. 描述了在位于新疆昌吉州境内的南山马鹿(Cervus elaphus)保护区发现的石竹科(Caryophyllaceae)石竹属(Dianthus)植物一新种—南山石竹(Dianthus nanshanicus C.Y.Yang &; L.X.Dong),并描绘了形态图。该新种与瞿麦(Dianthus superbus L.)相近,区别在于: 本新种花冠白色,喉部无丝毛状鳞片,茎单生,瓣片深裂为细条状。雄蕊伸出至花瓣的1/2。花粉圆球形, 外壁具穴状和刺状纹饰。种皮细胞较小近光滑。报道了该种的采集过程、生境以及该种在新疆南天山居群的大小和保护状况,并简单介绍了Dianthus在世界的分布情况。  相似文献

Summary Plant survival ex vitro requires functioning stomata, adequate cuticular wax composition and deposition, and normal morphological development. Light intensity, CO2 and relative humidity were altered inside an acoustic window mist reactor to study their effects on carnation (Dianthus caryophyllus) growth, stomata development, hyperhydration and epicuticular wax content. Increasing the light intensity from 65 to 145 μmol m−2 s−1 and enrichment of the gas phase with CO2 (1350 ppm) reduced the number of hyperhydrated plants from 75 to 25% and increased the percentage dry weight of normal (healthy) plants from 17 to 25%. Lowering the relative humidity (≈70% RH) surrounding the plants during the mist-off phase for the last 2 wk of culture reduced the number of hyperhydrated plants from 70 to 9% and also increased the percentage of dry weight of normal plants from 16 to 25%. The stomata on plants grown in conditions of high light or low humidity had smaller apertures and appeared sunken when compared to stomata from plants grown in low light and high relative humidity. The epicuticular wax profiles of plants from the greenhouse or Magenta boxes showed a distinct shift in wax compounds with developmental age, plant type (hyperhydrated or normal), and type of box that was used (vented or not). In addition, very different wax profiles were observed from plants grown in reactors with altered CO2 and light intensities.  相似文献

以香石竹叶片为外植体,用农杆菌(Agrobacteriumtumefaciens)介导法,在选择分化培养基中培养后,将香石竹重复结构的ACC氧化酶(ACO)基因核DNA导入香石竹‘Maber品种.经Southera杂交检测,证明外源基因已整合到香石竹基因组,共获得3株转化植株.转基因植株在隔离条件下栽培时正常开花,转基因T257株系切花衰老过程中乙烯释放量较对照低95%,没有乙烯跃变峰出现.在25℃条件下比较瓶插期,有2个转化株系瓶插期显著延长,最长比对照长了5 d以上.  相似文献

以对水质较为敏感的香石竹(Dianthus caryophyllus L.)为材料,用糖、苯甲酸钠等配制保鲜剂,观察保鲜剂对香石竹形态结构的影响和细胞内淀粉粒的变化。结果表明:用保鲜剂处理过的切花,花朵增大,花色鲜艳,茎、叶、花瓣、子房壁、胚珠等组织细胞壁出现皱缩的时间延迟,细胞内含淀粉粒的时间也较长。苯甲酸钠的保鲜效果优于硝酸银。  相似文献

利用扫描电镜对8种石竹属(Dianthus)植物的花粉形态进行了观察。结果表明:本属植物花粉为单粒花粉,圆球形,大小为30.5~44.0 μm,具多个萌发孔,孔数为12~16个,分散,孔圆形,孔径4.1~7.0 μm,具孔膜,孔膜上着生有若干棵小刺,花粉外壁具刺状和穴状纹饰,表面有或无穿孔。研究表明:花粉大小、表面纹饰、特别是萌发孔的形态特征存在较明显差异。可为石竹属植物的分类及野生资源利用等相关研究提供了可靠的孢粉学参考。  相似文献

New derivatives of aminooxyacetic acid were tested for their ability to inhibit ethylene formation in higher plants. Treatments with {[(isopropylidene)-aminojoxy}-acetic acid-2-(methoxy)-2-oxoethyl ester, {[(isopropylidene)-aminojoxy}-acetic acid-2-(hexyloxy)-2-oxoethyl ester or {[(cyclohexylidene)-amino]oxy}-acetic acid-2-(isopropyloxy)-2-oxoethyl ester reduced ethylene evolution by leaf discs of oilseed rape and drought-stressed barley leaves. The new compounds delayed senescence of cut carnation flowers. The endogenous levels of 1-aminocyclopropane-1-carboxylic acid (ACC) and its N-malonyl conjugate were also reduced in the leaf discs of oilseed rape. This suggests that a step in the biosynthesis of ethylene, prior to the formation of ACC, is inhibited by these new compounds. A lag phase in response suggests that these compounds have to be activated most likely by the production of metabolites with a free aminooxy group.  相似文献

Vitrified shoots regenerated from carnation petals (Dianthus caryophyllus L. cv. Scania) were recovered by culturing them in a medium containing 3.0 g/l Bacto-Peptone. Wax structures not found on vitrified shoots developed on the abaxial surface of leaves of recovered shoots and on those of normal leaves. Recovered shoots were rooted and successfully acclimatized while vitrified shoots could not survive the acclimatization process. The Bacto-Peptone solution was fractionated and the efficiency of each fraction for the recovery of vitrification was examined. Only basic, non high molecular fractions whose molecular weight was less than 10,000 were effective.  相似文献

Eighteen flavonoids were identified from an aqueous extract of the aerial parts of Dianthus versicolor, a plant used in traditional Mongolian medicine against liver diseases. The flavonoid C- and O-glycosides isoorientin-7-O-rutinoside, isoorientin-7-O-rhamnosyl-galactoside, isovitexin-7-O-rutinoside, isovitexin-7-O-rhamnosyl-galactoside, isoscoparin-7-O-rutinoside, isoscoparin-7-O-rhamnosyl-galactoside, isoscoparin-7-O-galactoside, and isoorientin-7-O-galactoside were isolated and structurally elucidated. Their structures were established on the basis of extensive spectroscopic techniques including LC–UV–DAD, LC–MSn, LC–HRMS, 1D and 2D NMR spectroscopy, and by GC–MS analysis after hydrolysis. Flavonoids with such a high glycosylation pattern are rare within the genus Dianthus. Furthermore, isovitexin-7-O-glucoside (saponarin), isovitexin-2″-O-rhamnoside, apigenin-6-glucoside (isovitexin), luteolin-7-O-glucoside, apigenin-7-O-glucoside, as well as the aglycons luteolin, apigenin, chrysoeriol, diosmetin, and acacetin were identified by TLC and LC–DAD–MSn in comparison to reference substances or literature data. The NMR data of seven structures have not been reported in the literature to date.  相似文献
