Soil represents a very heterogeneous environment for its microbiota. Among the soil inhabitants, bacteria and fungi are important organisms as they are involved in key biogeochemical cycling processes. A main energy source driving the system is formed by plants through the provision of plant-fixed (reduced) carbon to the soil, whereas soil nitrogen and phosphorus may move from the soil back to the plant. The carbonaceous compounds released form the key energy and nutrient sources for the soil microbiota. In the grossly carbon-limited soil, the emergence of plant roots and the formation of their associated mycorrhizae thus create nutritional hot spots for soil-dwelling bacteria. As there is natural (fitness) selection on bacteria in the soil, those bacteria that are best able to benefit from the hot spots have probably been selected. The purpose of this review is to examine the interactions of bacteria with soil fungi in these hot spots and to highlight the key mechanisms involved in the selection of fungal-responsive bacteria. Salient bacterial mechanisms that are involved in these interactions have emerged from this examination. Thus, the efficient acquisition for specific released nutrients, the presence of type-III secretion systems and the capacity of flagellar movement and to form a biofilm are pinpointed as key aspects of bacterial life in the mycosphere. The possible involvement of functions present on plasmid-borne genes is also interrogated.  相似文献

Evaluation of three indices for biofilm accumulation on complete dentures Objectives: The objective of this study was to evaluate the accuracy and reproducibility of three complete denture biofilm indices (Prosthesis Hygiene Index; Jeganathan et al. Index; Budtz‐Jørgensen Index) by means of a computerised comparison method. Background: Clinical studies into denture hygiene have employed a large number of biofilm indices among their outcome variables. However, the knowledge about the validity of these indices is still scarce. Materials and methods: Sixty‐two complete denture wearers were selected. The internal surfaces of the upper complete dentures were stained (5% erythrosine) and photographed. The slides were projected on paper, and the biofilm indices were applied over the photos by means of a scoring method. For the computerised method, the areas (total and biofilm‐covered) were measured by dedicated software (Image Tool). In addition, to compare the results of the computerised method and Prosthetic Hygiene Index, a new scoring scale (including four and five graded) was introduced. For the Jeganathan et al. and Budtz‐Jørgensen indices, the original scales were used. Values for each index were compared with the computerised method by the Friedman test. Their reproducibility was measured by means of weighed κ. Significance for both tests was set at 0.05. Results: The indices tested provided similar mean measures but they tended to overestimate biofilm coverage when compared with the computerised method (p < 0.001). Agreement between the Prosthesis Hygiene Index and the computerised method was not significant, regardless of the scale used. Jeghanathan et al. Index showed weak agreement, and consistent results were found for Budtz‐Jorgensen Index (κ = 0.19 and 0.39 respectively). Conclusion: Assessment of accuracy for the biofilm indices showed instrument bias that was similar among the tested methods. Weak inter‐instrument reproducibility was found for the indices, except for the Budtz‐Jørgensen Index. This should be the method of choice for clinical studies when more sophisticated approaches are not possible.  相似文献

1. The efficacy of leaf‐litter decomposition, sediment respiration, biofilm biomass, growth, chlorophyll a concentration and the autotrophic index (biofilm ash‐free dry mass/chlorophyll a) and fungal biomass for detecting human‐induced change was evaluated using 24 references and 15 disturbed stream sites located in central Portugal. 2. Decomposition rates of alder (Alnus glutinosa) and oak (Quercus robur) leaves and sediment respiration rates were effective in discriminating impairment. Decomposition was negatively correlated with abiotic factors, such as ammonium and nitrite concentrations, connectivity and alterations in the hydrological regime, and positively correlated with nitrate concentration and oxygen concentration. Sediment respiration rates were correlated with organic contamination, land use and morphological changes. 3. Growth rates of biofilm, concentration of chlorophyll a and the autotrophic index, although 41–73% higher at disturbed compared to reference sites, were not significantly different. These three variables were significantly correlated with total organic carbon, oxygen concentration, pH, nitrite and the presence of dams. Fungal biomass on leaves and biofilm biomass on natural substrata did not differ between reference and disturbed sites. 4. Our findings lend support to the use of functional variables like decomposition and sediment respiration in monitoring and when used together with structural variables should give a more holistic measure of stream health.  相似文献

Neanderthals have a distinctive suite of dental features, including large anterior crown and root dimensions and molars with enlarged pulp cavities. Yet, there is little known about variation in molar root morphology in Neanderthals and other recent and fossil members of Homo. Here, we provide the first comprehensive metric analysis of permanent mandibular molar root morphology in Middle and Late Pleistocene Homo neanderthalensis, and Late Pleistocene (Aterian) and recent Homo sapiens. We specifically address the question of whether root form can be used to distinguish between these groups and assess whether any variation in root form can be related to differences in tooth function. We apply a microtomographic imaging approach to visualise and quantify the external and internal dental morphologies of both isolated molars and molars embedded in the mandible (n = 127). Univariate and multivariate analyses reveal both similarities (root length and pulp volume) and differences (occurrence of pyramidal roots and dental tissue volume proportion) in molar root morphology among penecontemporaneous Neanderthals and Aterian H. sapiens. In contrast, the molars of recent H. sapiens are markedly smaller than both Pleistocene H. sapiens and Neanderthals, but share with the former the dentine volume reduction and a smaller root-to-crown volume compared with Neanderthals. Furthermore, we found the first molar to have the largest average root surface area in recent H. sapiens and Neanderthals, although in the latter the difference between M1 and M2 is small. In contrast, Aterian H. sapiens root surface areas peak at M2. Since root surface area is linked to masticatory function, this suggests a distinct occlusal loading regime in Neanderthals compared with both recent and Pleistocene H. sapiens.  相似文献

Several methods for determining the diversity of Lactobacillus spp were evaluated with the purpose of developing a realistic approach for further studies. The patient population was comprised of young children with an oral disease called severe early childhood caries. The ultimate goal of these studies was to ascertain the role of lactobacilli in the caries process. To accomplish that goal, we evaluated several methods and approaches for determining diversity including AP-PCR, chromosomal DNA fingerprinting, denaturing gradient gel electrophoresis, and 16S rRNA gene sequencing. Central to these methods was the gathering and screening of isolates from cultivation medium. Using various estimates of diversity, we addressed the question as to how many isolates represent the overall diversity and how cultivation compares to non-cultivation techniques. Finally, we proposed a working approach for achieving the goals outlined framed by both practical constraints in terms of time, effort and efficacy while yielding a reliable outcome.  相似文献

Phenol-soluble modulins (PSMs), such as α-PSMs, β-PSMs, and δ-toxin, are virulence peptides secreted by different Staphylococcus aureus strains. PSMs are able to form amyloid fibrils, which may strengthen the biofilm matrix that promotes bacterial colonization of and extended growth on surfaces (e.g., cell tissue) and increases antibiotic resistance. Many components contribute to biofilm formation, including the human-produced highly sulfated glycosaminoglycan heparin. Although heparin promotes S. aureus infection, the molecular basis for this is unclear. Given that heparin is known to induce fibrillation of a wide range of proteins, we hypothesized that heparin aids bacterial colonization by promoting PSM fibrillation. Here, we address this hypothesis using a combination of thioflavin T-fluorescence kinetic studies, CD, FTIR, electron microscopy, and peptide microarrays to investigate the mechanism of aggregation, the structure of the fibrils, and identify possible binding regions. We found that heparin accelerates fibrillation of all α-PSMs (except PSMα2) and δ-toxin but inhibits β-PSM fibrillation by blocking nucleation or reducing fibrillation levels. Given that S. aureus secretes higher levels of α-PSM than β-PSM peptides, heparin is therefore likely to promote fibrillation overall. Heparin binding is driven by multiple positively charged lysine residues in α-PSMs and δ-toxins, the removal of which strongly reduced binding affinity. Binding of heparin did not affect the structure of the resulting fibrils, that is, the outcome of the aggregation process. Rather, heparin provided a scaffold to catalyze or inhibit fibrillation. Based on our findings, we speculate that heparin may strengthen the bacterial biofilm and therefore enhance colonization via increased PSM fibrillation.  相似文献

aro mutants of Salmonella enterica are frequently used as live vaccines for the oral vaccination of domestic animals. Interestingly, besides their auxotrophy, they appear to be of reduced resistance to the components of innate immune response due to a defect in outer membrane and/or cell wall integrity. Because different extracellular structures associated with the cell wall or outer membrane are involved in biofilm formation, we were interested in the ability of aroA and aroD mutants of S . Enteritidis to adhere to solid surfaces. We found that aroA and aroD mutants did not adhere to solid surfaces although they bind Congo red and produced d -mannose and d -glucose capsular polysaccharides in the same amounts as the wild-type strain. However, the aro mutants exhibited a decreased production of cellulose, N -acetyl- d -glucosamine or N -acetylneuraminic acid containing capsular polysaccharide and fimbriae which explains their inability to form biofilms. aroA and aroD containing plasmids complemented all the defects of the aro mutants. Beside its attenuation for different hosts, the loss of ability to form biofilm is an additional interesting characteristic of aro mutants.  相似文献

Bromage and Dean originally outlined a nondestructive method for the study of enamel formation and concluded that early hominids resembled the extant apes more closely than they did modern humans in their rates of growth and maturation. The method used assumed that an enamel circadian rhythmicity was referable to a longer near-weekly period represented by perikymata (periodic surface growth features). This assumption became a matter of debate and discussion. In this study, developing teeth in Macaca nemestrina were labeled with polychrome fluorescent dyes. Examination of the distribution of these dyes in two sectioned teeth provides experimental confirmation of enamel circadian periodicity.  相似文献
