Dopamine Metabolism in Hypoxanthine-Guanine Phosphoribosyltransferase-Deficient Variants of PC12 Cells 总被引:2,自引:1,他引:1
Lesch-Nyhan syndrome results from a deficiency of hypoxanthine-guanine phosphoribosyltransferase (HPRT). It is manifest by behavioral abnormalities, including self-mutilation, and evidence of abnormal 3,4-dihydroxyphenylethylamine (dopamine) metabolism. To assess whether an HPRT deficiency in a dopaminergic cell can adversely affect dopamine metabolism in that cell, dopamine metabolism was examined in HPRT-deficient variants of PC12 pheochromocytoma cells and in cells that had regained HPRT activity by virtue of transformation with a recombinant retrovirus containing the human gene for HPRT. There was no correlation between HPRT activity and endogenous dopamine levels, dopamine uptake, dopamine release, or monoamine oxidase activity. Transformation with the HPRT retrovirus did not adversely affect dopamine metabolism. 相似文献

Regional amino acid concentrations were measured in rat brain fixed by microwave irradiation at three levels of elevated atmospheric pressure corresponding to different phases of the high-pressure neurological syndrome [20 atmospheres absolute (ATA), no clinical signs; 60 ATA, tremor; 85 ATA, severe tremor and myoclonic jerks]. No changes in amino acid content occurred at 20 or 60 ATA. At 85 ATA glutamine content increased in hippocampus, striatum, cerebellum, and substantia nigra, and gamma-aminobutyric acid content increased in hippocampus. It is suggested that enhanced glutamate release in various subcortical structures contributes to the myoclonic activity observed at 85 ATA. 相似文献

Using HeLa S-3 cells synchronized by selective detachment, in this paper we report a parallel study of nuclear morphology
and autoradiography grain patterns between middle G1 and middle S phases: Our results show two distinct [3H]-thymidine labeling patterns. The first “peripheral” labeling pattern has a characteristic nuclear size distribution, in
contrast to the heterogeneous and varying size distributions of Feulgen-stained nuclei, and apparently is characteristic of
very early S phase. The sizes of the second labeling pattern—homogeneous or inhomogeneous grain distribution throughout the
nucleus—are equal or larger than the first and vary with S phase progression. Together, the corresponding nuclear sizes of
the labeled nuclei represent the larger extreme of nuclear areas, and the labeling index closely parallels the fraction of
nuclei with areas larger than the minimum size of the labeled nuclei. These results suggest a characteristic nuclear size
(reflecting unique intranuclear DNA distribution) as a necessary, if not sufficient, requirement for S phase initiation. Parallel
experimentation with rat liver cells—synchronized in vivo by partial hepatectomy and analyzed by thin section autoradiography—confirms
the existence of a peripheral labeling pattern in both the very early part and the very late part of S phase, which reconciles
our data with previous results and points to the fact that both initiation and termination sites for DNA replication are near

Summary Current procedures for isolating intestinal epithelial cell surface and intracellular membranes are based on the assumption that each organelle is marked by some unique constitutent. This assumption seemed inconsistent with the dynamic picture of subcellular organization emerging from studies of membrane turnover and recycling. Therefore, we have designed an alternative fractionation which is independent ofa priori marker assignments. We subjected mucosal homogenates to a sequence of separations based on sedimentation coefficient, equilibrium density, and partitioning in aqueous polymer twophase systems. The resulting distributions of protein and enzymatic markers define a total of 17 physically and biochemically distinct membrane populations. Among these are: basal-lateral membranes, with Na,K-ATPase enriched 21-fold; brush-border membranes, with alkaline phosphatase enriched as much as 38-fold; two populations apparently derived from the endoplasmic reticulum; a series of five populations believed to have been derived from the Golgi complex; and a series of five acid phosphatase-rich populations which we cannot identify unequivocally. Each of the five enzymatic markers we have followed is associated with a multiplicity of membrane populations. Basallateral, endoplasmic reticulum, and Golgi membranes contain alkaline phosphatase at the same specific activity as the initial homogenate. Similarly, Na,K-ATPase appears to be associated branes at specific activities two-to seven-fold that of the initial homogenate. 相似文献

Considerable inherent variations in the relation between macropterous and brachypterous wing forms, and nymphal density were found in field populations of the brown planthopper, Nilaparvata lugens Stål (Homoptera: Delphacidae), collected from various locations in Japan. When compared under uniform laboratory rearing conditions, most of the female populations exhibited higher ratios of macropters with increasing nymphal density, but some showed extremely high proportion of brachypters and the others were highly macropterous, over broad ranges of density. These results indicate the possibility that the planthoppers in Japan, which are known not to persist in winter, are derived from different migration sources.About ten generations of successive selection for brachyptery from a population showing usual density-dependent wing morphism generated populations similar to highly brachypterous ones mentioned above. Genetic analysis of the inheritance of wing morphism revealed that brachyptery in the females was controlled by a single pair of dominant alleles. However, in the males wing forms did not segregate so clearly in the crossing experiments. This suggests that wing morphism in N. lugens in under sex-limited inheritance.

Zusammenfassung In Feldpopulationen von Nilaparvata lugens Stål., welche in verschiedenen Regionen Japans gesammelt wurden, bestand in der Beziehung zwischen makropteren bzw. brachypteren Flügelformen und der Larvendichte eine beträchtliche Variation. Unter einheitlichen Zuchtbedingungen im Laboratorium stieg der Makropterenanteil bei den meisten Weibchenpopulationen mit steigender Temperatur; bei einigen Populationen hingegen war entweder der Brachypterenanteil oder der Makropterenanteil extrem hoch und zwar über weite Dichtebereiche. Dies deutet auf die Möglichkeit hin, dass die Zikade in Japan, wo sie bekanntlich nicht überwintert, jeweils aus verschiedenen Quellen einwandert.Wenn eine Population mit der üblichen dichteabhängigen Flügelausbildung 10 Generationen lang auf Brachypterie selektioniert wurde, entstanden Populationen, die den erwähnten hochbrachypteren Populationen aus dem Feld glichen. Die genetische Analyse der Vererbung der Brachypterie ergab, dass bei Weibchen ein einzelnes dominantes Allel verantwortlich ist. Bei Männchen dagegen trennten sich bei Kreuzungsexperimenten die Flügelformen nicht so klar. Dies deuted auf Unterschiede zwischen den Geschlechtern bei der Vererbung der Flügelformen.相似文献

The softening of wet lipid bilayer membranes during their gel-to-fluid first-order phase transition is studied by computer simulation of a family of two-dimensional microscopic interaction models. The models include a variable number, q, of lipid chain conformational states, where 2q10. Results are presented as functions of q and temperature for a number of bulk properties, such as internal energy, specific heat, and lateral compressibility. A quantitative account is given of the statistics of the lipid clusters which are found to form in the neighborhood of the transition. The occurrence of these clusters is related to the softening and the strong thermal density fluctuations which dominate the specific heat and the lateral compressibility for the high-q models. The cluster distributions and the fluctuations behave in a manner reminiscent of critical phenomena and percolation. The findings of long-lived metastable states and extremely slow relaxational behavior in the transition region are shown to be caused by the presence of intermediate lipid chain conformational states which kinetically stabilize the cluster distribution and the effective phase coexistence. This has as its macroscopic consequence that the first-order transition apperas as a continuous transition, as invariably observed in all experiments on uncharged lecithin bilayer membranes. The results also suggest an explanation of the non-horizontal isotherms of lipid monolayers. Possible implications of lipid bilayer softening and enhanced passive permeability for the functioning of biological membranes are discussed.Abbreviations PC

Bolivian squirrel monkeys, unlike those of Brazilian origin, exhibit a marked fasting hyperbilirubinemia (FH) similar to that observed in Gilbert's syndrome in man. Since no delays in the hepatic clearance of sulfobromophthalein or indocyanine green are present, the Bolivian monkey appears to be similar to Gilbert's type I syndrome. FH can be significantly decreased by either phenobarbital or tin-protoporphyrin pretreatment. Nicotinic acid-induced hyperbilirubinemia and delayed tolbutamide clearance were not observed as in the human syndrome. 相似文献

The wake and sleep-onset times of a patient with a sleep-wake cycle longer than 24 hr were recorded by the patient for 4 years. During this time, the patient found himself unable to maintain a 24-hr sleep-wake schedule. When treated with 1-2 mg clonazepam, taken nightly, he was able to become entrained to a 24-hr day. Despite entrainment of his sleep-wake cycle, the patient reported depression, lack of motivation and fatigue and chose not to continue taking the drug. 相似文献

Addition of 15L-hydroperoxy-5,8,11,13-eicosatetraenoic acid (15-HPETE) to human leukocytes led to the formation of a novel series of compounds containing four conjugated double bonds. The yield of tetraenes was increased approx. 100-fold when ionophore A23187 (5 μM) was added simultaneously with 15-HPETE. Mice were injected for 1-2 months daily with 10 mg immunoglobulin G (IgG) from four patients with Lambert-Eaton myasthenic syndrome (LEMS); control mice were injected with pooled human IgG from normal donors. Gastrocnemius muscles were homogenised for the assay of acetylcholine (ACh), choline acetyltransferase (ChAT), and cholinesterase (ChE). The ACh, ChAT, and ChE contents of gastrocnemius muscles from "LEMS mice" were about the same as the control values, which were 180 pmol, 40 nmol X h-1 (37 degrees C), and 15 mumol X h-1 (37 degrees C), respectively. Hemidiaphragms were treated with an irreversible ChE inhibitor (Soman) and incubated at 20 degrees C for estimation of ACh release. Resting ACh release from experimental muscles was reduced by about 25% (P2 less than 0.05) and the release evoked by 3 s-1 nervous stimulation by 50% (P2 less than 0.05). On the other hand, 50 mM KCl-induced transmitter release was not abnormal in LEMS mice. The findings indicate that IgG antibody from patients with LEMS may bind to nerve terminal determinants that are involved in quantal and nonquantal ACh release. 相似文献
