Salicylic acid (SA) is one of the key signal molecules in regulating plant resistance to diverse pathogens. In Arabidopsis thaliana, it is predominantly associated with resistance against biotrophic and hemibiotrophic pathogens, and triggering systemic acquired resistance. In contrast, the effect of SA on the defence efficiency of wheat against fusarium head blight (FHB) and its causal agent, Fusarium graminearum, is still poorly understood. Here we show that the F. graminearum mycelial growth and conidia germination were significantly inhibited, and eventually halted in the presence of increasing concentration of SA in both liquid and solid media. Addition of SA also significantly reduced the production of the mycotoxin deoxynivalenol (DON). However the inhibitory effect of SA required acidic growth conditions to be observed while basic conditions allowed F. graminearum to use SA as a carbon source. High performance liquid chromatography (HPLC) analysis confirmed the capacity of F. graminearum to metabolize SA. To better understand the effect of SA on F. graminearum mycelial growth, we have compared the expression profiles of SA-treated and untreated F. graminearum liquid cultures after 8 and 24 h of treatment, using an F. graminearum custom-commercial microarray. The microarray analysis suggested that F. graminearum can metabolize SA through either the catechol or gentisate pathways that are present in some fungal species. Inoculation of F. graminearum conidia in a SA-containing solution has led to reduced FHB symptoms in the very susceptible Triticum aestivum cv. Roblin. In contrast, no inhibition was observed when SA and conidia were inoculated sequentially. The expression patterns for the wheat PR1, NPR1, Pdf1.2, and PR4 genes, a group of indicator genes for the defence response, suggested that SA-induced resistance contributed little to the reduction of symptoms in our assay conditions. Our results demonstrate that, although F. graminearum has the capacity to metabolize SA, SA has a significant and direct impact on F. graminearum through a reduction in efficiency of germination and growth at higher concentrations.  相似文献

Phytoliths (silica bodies) occur in Poaceae species in large numbers and have been shown to have antiherbivory roles. However, phytoliths occur also in many other taxa in much smaller numbers, which raises the question of the extent of both their potential and actual antiherbivory role in these taxa. In order to address the question of their potential antiherbivory role, we sampled 20 wild-growing southwest Asian species of the family Asteraceae, species of which have a much lower phytolith concentrations than Poaceae taxa. We studied the potential positive effect of grazing on phytolith formation and the possible tendency of plants to have higher concentrations of such defence structures in their reproductive organs. We sampled plants from populations of 12 non-spiny and eight spiny species growing in un-grazed and grazed plots in seven sites along a large rainfall gradient (80–900 mm mean annual) in Israel, a region known for its long and intensive grazing history. The study included 21 pairs of un-grazed and grazed plants from 16 of these 20 species. In addition, ten populations of eight species were sampled in order to examine whether phytolith concentrations in the reproductive organs are higher than in vegetative organs. We did not find consistently higher phytolith concentrations in grazed plants compared to un-grazed plants of the same species and habitat (15 species), and in 12 out of 21 pairs of un-grazed and grazed plants (from 15 species) we even found higher phytolith concentrations in un-grazed plants, a phenomenon which was more common in the more arid sites. Phytolith concentrations in inflorescences are commonly (6 out of the 8 species) lower than in the rest of the shoot. We conclude that the antiherbivory potential of phytoliths in the southwest Asian Asteraceae as a group is much smaller than in the Poaceae.  相似文献

The microtubule (MT) network is highly dynamic and undergoes dramatic reorganizations during the cell cycle. Dimers of α- and β-tubulins rapidly polymerize to and depolymerize from the end of MT fibrils in an intrinsic GTP-dependent manner. MT severing by ATP-driven enzymes such as katanin and spastin contributes significantly to microtubule dynamics, and it has been shown that katanin p60, a AAA+ family protein, has ATPase and MT-severing activities. The mechanism of MT severing by katanin p60 is poorly understood, and the residues in katanin p60 and tubulins important for severing activity were therefore explored in this study. MT-severing activity, but not ATPase activity, was inhibited by mutations of the conserved aromatic residue and the flanking basic residues in the pore region of the katanin p60 hexameric ring. When the acidic residue-rich C-terminal unstructured segment of either α- or β-tubulin was removed, polymerized MTs were resistant to katanin p60 treatment. Interactions between katanin p60 and the mutant MTs, on the other hand, were unaffected. Taken together, these findings led us to propose that the interactions between the positively charged residues of katanin p60 and the acidic tails of both tubulins are essential for efficient severing of MTs.  相似文献

为探讨基于康复教育护理团队实施康复护理对颅脑外伤术后偏瘫患者生活能力及神经功能恢复的影响,本研究选取96例颅脑外伤术后偏瘫患者,随机分为对照组与观察组各48例,对照组采用常规护理,观察组应用基于康复教育护理团队的康复护理。通过比较两组干预前后日常生活能力评分(ADL)、Fugl-Meyer运动功能量表(FMA)评分及美国国立卫生院神经功能缺损评分(NIHSS)的变化,采用焦虑自评量表(SAS)及抑郁自评量表(SDS)评定干预前后患者心理状态的变化,并统计两组护理期间并发症发生率。研究发现,干预8周,两组轻度依赖、中度依赖所占比例上升,重度依赖、完全依赖所占比例下降,观察组轻度依赖所占比例高于对照组,完全依赖所占比例高于对照组(p<0.05);干预8周,两组FMA表感觉、运动功能、关节活动度、疼痛、平衡等维度评分均上升,观察组各维度评分上升幅度高于对照组(p<0.05),两组NIHSS评分、SAS评分、SDS评分均降低,观察组上述量表评分低于对照组(p<0.05)。观察组护理期间并发症发生率略低于对照组,但比较差异无统计学意义(p<0.05)。本研究证实,对颅脑外伤术后偏瘫患者采用基于康复教育护理团队的康复护理模式可改善患者日常生活能力、运动功能及神经功能,纠正患者不良情绪。  相似文献

The activities of the enzymes responsible for cell-wall strengthening and salicylic acid (SA) content in Norway spruce seedlings were investigated after inoculation with the ectomycorrhizal fungus Pisolithus tinctorius or the pathogen Heterobasidion annosum, and after treatment with elicitors from both of these fungi. Inoculation with both fungi increased guaiacol peroxidase (POD) activity in the roots of the pathogen-inoculated seedlings during the earliest phases of colonisation, and induced the activities of several POD isoforms. Two of these were only seen in pathogen-inoculated seedlings and corresponded with increased POD activity against ferulic acid. Colonisation with H. annosum triggered an increase in phenylalanine ammonia lyase (PAL) activity in the roots of the spruce seedlings, which was followed by an accumulation of free SA. One month after inoculation levels of free SA were increased also in the shoots of H. annosum-inoculated seedlings. In contrast increase in free SA content in the roots of P. tinctorius-inoculated seedlings was only transient. Similarly to inoculation, treatment with elicitors of H. annosum increased the PAL and POD activity, as well as SA content in the roots of spruce seedlings. A positive correlation between PAL activity and SA content in the H. annosum-inoculated seedlings and accumulation of SA precursors in the phenylpropanoid pathway indicate that the plant defence mechanisms, during which SA is synthesised through the PAL pathway, are exploited by H. annosum for facilitation of colonisation.  相似文献

Inverse dynamics based simulations on musculoskeletal models is a commonly used method for the analysis of human movement. Due to inaccuracies in the kinematic and force plate data, and a mismatch between the model and the subject, the equations of motion are violated when solving the inverse dynamics problem. As a result, dynamic inconsistency will exist and lead to residual forces and moments. In this study, we present and evaluate a computational method to perform inverse dynamics-based simulations without force plates, which both improves the dynamic consistency as well as removes the model?s dependency on measured external forces. Using the equations of motion and a scaled musculoskeletal model, the ground reaction forces and moments (GRF&Ms) are derived from three-dimensional full-body motion. The method entails a dynamic contact model and optimization techniques to solve the indeterminacy problem during a double contact phase and, in contrast to previously proposed techniques, does not require training or empirical data. The method was applied to nine healthy subjects performing several Activities of Daily Living (ADLs) and evaluated with simultaneously measured force plate data. Except for the transverse ground reaction moment, no significant differences (P>0.05) were found between the mean predicted and measured GRF&Ms for almost all ADLs. The mean residual forces and moments, however, were significantly reduced (P>0.05) in almost all ADLs using our method compared to conventional inverse dynamic simulations. Hence, the proposed method may be used instead of raw force plate data in human movement analysis using inverse dynamics.  相似文献
