Tyrosinase plays a pivotal role in the synthesis of melanin pigment synthesis on skin utilizing tyrosine as a substrate. Melanin is responsible for the protection against harmful ultraviolet irradiation, which can cause significant pathological conditions, such as skin cancers. However, it can also create esthetic problems when accumulated as hyperpigmented spots. Various skin-whitening ingredients which inhibit tyrosinase activity have been identified. Some of them, especially ones with natural product origins, possess phenolic moiety and have been employed in cosmetic products. Semi-synthetic and synthetic inhibitors have also been developed under inspiration of the natural inhibitors yet some of which have no phenolic groups. In this review, tyrosinase inhibitors with natural, semi-synthetic and synthetic origins are listed up with their structures, activities and characteristics. Further, a recent report on the adverse effect of a natural melanin synthesis inhibitor which was included in skin-whitening cosmetics is also briefly discussed.  相似文献

Melanin pigments constituted 13.9% of the chemical composition of the sclerotial walls of Sclerotium rolfsii and was associated with proteins, reducing sugars and amino acids. The lipid and ash contents in the sclerotial walls were double those in the hyphal walls of the fungus. Increasing age of the culture and maturation of the sclerotia were always accompanied by elevation of lipid peroxides and melanin pigments. Such behaviour may indicate that lipid peroxidation and melanin formation are operating in parallel during sclerotial biogenesis and maturation. These two processes depend on the theory of oxidative stress, as affected by growth conditions. Both processes could be stopped or sharply retarded when subjected to some antioxidant growth factors such as vitamins (ascorbic acid), micro-elements (selenium) and sulfhydryl compounds (glutathione). A clear relation between oxidative stress, myceliogenic germination and lytic activity via melanin production was observed. This finding appears promising in applying a new control measure against diseases caused by sclerotia-producing fungi without using traditional toxic fungicides.  相似文献

Bacillus cereus 58 (Bc58) is a UV-resistant wild type strain that has an ability to produce a sorrel pigment induced by L-tyrosine. The Fourier-transform infrared (FT-IR) spectra and chemical tests of its pigment are similar to that of the standard melanin (Sigma). A bioassay shows that the LC50 of a Bacillus thuringiensis (Bt) formulation added with the melanin of Bc58 and exposed to UV for 5 h is 16.1 μg/ml, which is similar to that of the Bt formulation without UV treatment, however, it is almost double that of the Bt formulation exposed to UV without the melanin of Bc58. The result of SDS-PAGE indicates that the melanin of Bc58 can protect the insecticidal crystal proteins from degradation. Several studies have shown that melanin‐based traits play a crucial role in social contexts as they are associated with dominance, personality and social behaviour. However, most of these studies have focused on adults, and the role of these traits in juveniles has rarely been explored. Here, we explore the association between two melanin‐based traits and nestling personality in Common Kestrels Falco tinnunculus. Our results show that female nestlings with blacker plumages displayed bolder personalities, providing evidence of sex‐dependent phenotypic integration of these two traits in males and females. We consider that this differential integration may arise from different selection pressures acting on males and females on plumage coloration during adulthood and that nestling coloration can act as a status signal within the juvenile age‐class.  相似文献

Melanins are widely used in medicine, pharmacology, cosmetics and other fields. Although several technologies for the purification of water‐insoluble dioxyphenylalanine (DOPA) melanins have been described, a source of water‐soluble melanin is highly desirable. Here we describe an effective procedure for the isolation and purification of water‐soluble melanin using the culture medium of Bacillus thuringiensis subsp. galleriae strain K1. Water‐soluble melanin from this organism has an isoelectric point (pI = 3.0–3.2) and was purified optimally by adsorbtion using the IA‐1r resin and elution as a concentrated solution. The purified melanin obtained exhibited a similar infra‐red absorbtion spectrum to synthetic melanin and contained quinolic and phenolic structures and an amino acid content of around 20% after acid hydrolysis. The molecular weight of the purified melanin determined by SDS‐PAGE was 4 kDa and the electromagnetic spin resonance spectrum of the purified microbial melanin was a slightly asymmetric singlet without hyperfine structure with about 7 Gauss width of the line between points of the maximum incline and g = 2.006. The concentration of paramagnetic centers in melanin is 0.21 × 1018 spin/g. The results obtained provide a rapid, simple and inexpensive method for the large scale purification of water soluble melanin that may have widespread applications.  相似文献

Different camouflages work best with some background matching colour. Our understanding of the evolution of skin colour is based mainly on the genetics of pigmentation (“background matching”), with little known about the evolution of the neuroendocrine systems that facilitate “background adaptation” through colour phenotypic plasticity. To address the latter, we studied the evolution in vertebrates of three genes, pomc, pmch and pmchl, that code for α‐MSH and two melanin‐concentrating hormones (MCH and MCHL). These hormones induce either dispersion/aggregation or the synthesis of pigments. We find that α‐MSH is highly conserved during evolution, as is its role in dispersing/synthesizing pigments. Also conserved is the three‐exon pmch gene that encodes MCH, which participates in feeding behaviours. In contrast, pmchl (known previously as pmch), is a teleost‐specific intron‐less gene. Our data indicate that in zebrafish, pmchl‐expressing neurons extend axons to the pituitary, supportive of an MCHL hormonal role, whereas zebrafish and Xenopus pmch+ neurons send axons dorsally in the brain. The evolution of these genes and acquisition of hormonal status for MCHL explain different mechanisms used by vertebrates to background‐adapt.  相似文献

Synthetic soluble (—)-dopa melanin was prepared in deuteriated buffer, pH 8, by autooxidation of the precursor. At 6 mM of the precursor, the incorporation was over 90%. The changes in the line width measurements of N-CH3 protons of enantiomers of ephedrine in the soluble melanin were quantified by NMR spectroscopy. The dissociation constants of (—)-1R,2S-ephedrine, (+)-1S,2R-ephedrine, (—)-1R,2R-ψ-ephedrine, and (+)-1S,2S-ψ-ephedrine were 11.7, 4.20, 3.60, and 4.80 mM, respectively. Since the concentration of (—)-dopa was known and since the conversion of (—)-dopa to indole units of melanin was considered as 1:1, the stoichiometry of the interaction between the drug and the indole unit was calculated. Based on the dissociation constants of the enantiomers, it appears that up to four molecules of (—)-ephedrine can interact with one indole unit of the melanin, while such a ratio for other isomers appear to be 2:1. Increasing evidence of links between climate change, anthropogenic stress and coral disease underscores the importance of understanding the mechanisms by which reef-building corals resist infection and recover from injury. Cellular inflammation and melanin-producing signalling pathway are two mechanisms employed by invertebrates to remove foreign organisms such as pathogens, but they have not been recorded previously in scleractinian corals. This study demonstrates the presence of the phenoloxidase (PO) activating melanin pathway in two species of coral, Acropora millepora and a massive species of Porites, which both develop local pigmentation in response to interactions with a variety of organisms. L-DOPA (3-(3,4-dihydroxyphenyl)-L-alanine) substrate-based enzyme activation assays demonstrated PO activity in healthy tissues of both species and upregulation in pigmented tissues of A. millepora. Histological staining conclusively identified the presence of melanin in Porites tissues. These results demonstrate that the PO pathway is active in both coral species. Moreover, the upregulation of PO activity in areas of non-normal pigmentation in A. millepora and increased melanin production in pigmented Porites tissues suggest the presence of a generalized defence response to localized stress. Interspecific differences in the usage of pathways involved in innate immunity may underlie the comparative success of massive Porites sp. as long-lived stress tolerators.  相似文献

AIMS: To isolate a bacterium that produces high yield of melanin and to examine the effect of this bacterial pigment on the efficacy of a bioinsecticide. METHODS AND RESULTS: A novel melanin-producing bacterium, designated as strain WS, was isolated from the East Lake, Wuhan, China. Taxonomic studies of this strain indicate that it belongs to Aeromonas media. Physicochemical analysis of the pigment produced by strain WS (melanin WS) suggests that it is the authentic 3,4-dihydroxyphenylalanine (DOPA)-melanin. This melanin and that produced by Pseudomonas maltophilia P28 (melanin P28) share many biophysical properties, but the yield of the melanin WS is significantly higher than that of the melanin P28. In addition, the melanin WS appears to be more effective in the protection of a bioinsecticide against ultraviolet (UV) or solar radiation. At the concentration of 10 ppm, the melanin P28 exhibited no photoprotective effect on the bioinsecticide against UV radiation; in contrast, 5 ppm of melanin WS displayed an obvious protective effect. Similarly, the melanin WS displayed more protective effect on the bioinsecticide against solar radiation than the melanin P28 did over a 4-day period, with the effect being more dramatic for the last 2 days. CONCLUSIONS: We have isolated a novel bacterial strain of A. media that produces high levels of melanin. The melanin produced by this strain offers effective photoprotection of a commercial bioinsecticide BTI against UV and solar radiation. SIGNIFICANCE AND IMPACT OF THE STUDY: Our study suggests that the melanin produced by our newly isolated A. media strain has the potential to be used as a general photoprotective agent for bioinsecticides.  相似文献

Background

Tautomerization of dopachrome to 5,6-dihydroxyindole-2-carboxylic acid (DHICA) is a biologically crucial reaction relevant to melanin synthesis, cellular antioxidation, and cross-talk among epidermal cells. Since dopachrome spontaneously converts into 5,6-dihydroxyindole (DHI) via decarboxylation without any enzymes at physiologically usual pH, the mechanism of how tautomerization to DHICA occurs in physiological system is a subject of intense debate. A previous work has found that Cu(II) is an important factor to catalyze the tautomerization of dopachrome to DHICA. However, the effect of Cu(II) on the tautomerization has not been clarified at the atomic level.

Methods

We propose the reaction mechanism of the tautomerization to DHICA by Cu(II) from density functional theory-based calculation.

Results

We clarified that the activation barriers of α-deprotonation, β-deprotonation, and decarboxylation from dopachrome are significantly reduced by coordination of Cu(II) to quinonoid oxygens (5,6-oxygens) of dopachrome, with the lowest activation barrier of β-deprotonation among them. In contrast to our previous work, in which β-deprotonation and quinonoid protonation (O5/O6-protonation) were shown to be important to form DHI, our results show that the Cu(II) coordination to quinonoid oxygens inhibits the quinonoid protonation, leading to the preference of proton rearrangement from β-carbon to carboxylate group but not to the quinonoid oxygens.

Conclusion

Integrating these results, we conclude that dopachrome tautomerization first proceeds via proton rearrangement from β-carbon to carboxylate group and subsequently undergoes α-deprotonation to form DHICA.

General significance

This study would provide the biochemical basis of DHICA metabolism and the generalized view of dopachrome conversion which is important to understand melanogenesis.  相似文献
