We investigated the potential role of mitochondrial DNA (mtDNA) in colorectal carcinogenesis by constructing a eukaryotic expression vector of the mitochondrial D-loop gene from colorectal cancer cell SW480 and transfected NIH3T3 cells. The NIH3T3/SW480 cells exhibited a significantly increased growth rate and colony formation rate, and also had a decreased apoptotic rate. Polyploidy and aberrant chromosomes were detected in the NIH3T3/SW480 cells by chromosome karyotype analysis. Our results suggested that mtDNA from colorectal cancer cells promotes the malignant phenotype of NIH3T3 cells. Further study of the biological functions of NIH3T3/SW480 cells might be helpful in understanding the role of mtDNA in colorectal carcinogenesis.  相似文献

To evaluate inter- and intra-populational divergence of the endangered Siberian crane a comparative analysis of mtDNA D-loop region (490 bp) sequences was performed for 17 captive cranes including 14 originating from the eastern and 3 from the central nesting area. Thirteen variable sites form 9 haplotypes, 6 belonging to the eastern nesting site individuals, 2 belonging to the central nesting site and one being common for both nesting areas. The haplotype network analysis did not show significant separation of these two groups. The data obtained could provide genetic grounds for the programs of restoration of the central nesting area population by captive breeding and reintroduction to the wild using the eastern nesting site population gene pool. These results could also be a basis for further evaluation of Siberian crane populations genetic divergence needed for its adequate protection.  相似文献

洱海鲤属鱼类同域分化形成的分子遗传学证据   总被引:6,自引:0,他引:6  
对洱海的 3种鲤鱼———洱海鲤 (C .barbatus)、春鲤 (C .longipectoralis)和杞麓鲤 (C .carpiochila)线粒体DNA(mtDNA)进行了限制性片段长度多态 (RFLP)分析和mtDNA控制区 (D loop)序列测定。RFLP分析结果表明 ,16种酶在洱海鲤、春鲤和杞麓鲤 3个种共 14个个体上 ,仅DraⅠ在春鲤的一个个体上检出一个位点的限制性片段长度多态 ,独立为单倍型Ⅱ ,3个种的其余 13个个体共享单倍型Ⅰ ,种间遗传距离为 0 %~ 0 0 0 75 % ,此种间差异值比已报道的鱼类的种间差异甚至种内差异都低得多。D loop区的序列测定结果表明 ,长为 4 4 9bp的序列共显示有 7个变异位点 ,均为碱基替换 ,变异比例为 1 5 6 % ,在 14个鲤鱼个体中共检出 6种单倍型 ,种间的遗传距离为 0 0 82 %~ 0 171% ,与已研究的其他几种异域分布的鲤鱼相比 ,也至少小一个数量级。mtDNA的RFLP及D loop区的序列比较都揭示出洱海 3种鲤鱼之间的遗传差异远小于异域分布的鲤属鱼类物种之间及其他鱼类的同属种间的遗传差异。对洱海鲤属鱼类种间如此小的遗传差异唯一合理的解释就是 :它们确是由同域分化而形成的。  相似文献

中国部分家养山羊mtDNA D环区遗传多样性与进化   总被引:1,自引:0,他引:1  
对我国10个家养山羊品种140只个体的mtDNA D-loop区进行测序分析, 测得结果: 整个D-loop区为 1 211~1 213 bp, 共检测到84种单倍型, 171个多态位点。其中核苷酸多样性(Nucleotide diversity, Pi)为: 0.02063 ± 0.00225, 单倍型多样性(Haplotype gene diversity, Hd)为: 0.988 ± 0.003, 平均核苷酸差异数(Average number of nucleotide differences) k为: 24.896。表明我国家养山羊品种遗传多样性丰富。通过构建NJ网络进化树, 得出中国家养山羊主要分为两大支系, 并且其中一支与角骨羊(Capra aegagrus)聚在一起, 而旋角野山羊(Capra fal-coneri)单独聚为一支, 说明角骨羊对中国家养山羊贡献较大。  相似文献

We report rapid change of morphology and mitochondrial genes in white-footed mice (Peromyscus leucopus) in the Chicago (Illinois, USA) region. We sequenced mitochondrial DNA COX2 from 55 museum skins of white-footed mice caught in the Chicago area since 1855 and from 44 mice recently trapped in the same locations. We found consistent directional genotype replacement at five separate collection locations. We later focused on a single one of these locations (Volo Bog State Natural Area) and sequenced mitochondrial D-loop control region from 58 museum skins of mice collected in 1903-1976 and 32 mice recently trapped there. We found complete and more recent replacement of D-loop haplotypes, apparently occurring between 1976 and 2001. We tested whether these genetic changes were mirrored by changes in morphology by comparing 15 external and cranial traits. We found no significant morphological differences between mice collected in 1903-1976; however, mice collected in 2001-2003 showed 9 of 15 measurements to be significantly changed relative to the earlier samples. Recent mice were longer in total length, with broader, longer noses, and longer but shallower skulls(1). Discriminant function analysis allowed for 100% correct classification using these traits. Principal components analysis shows variance over time is well distributed across both external and cranial measures. The sequential replacements of haplotypes and the rapid change of morphology can best be explained by replacement of the regional population with immigrants from genetically distinct neighbouring populations, likely facilitated by the large environmental changes occurring over the time period. Replacement with genotypes from external populations may be a common mechanism of evolution of newly adaptive local forms in an increasingly human-impacted world.  相似文献

Molecular variation of mitochondrial DNA (mtDNA) was investigated for rhesus macaques (Macaca mulatta) of Bangladesh. A partial sequence (583–599 bp) of mtDNA containing the second variable region of the D-loop was compared for 39 individuals from five localities in the country. A total of seven haplotypes were detected with substitutional or insertion/deletion mutations. They contained a unique polymorphism of pentanucleotide STRs (short tandem repeats). There were at least four different length types, from two to five repeats of the unit nucleotide. One site of substitution and one site of single nucleotide insertion/deletion were also involved in the polymorphism. The mutation hot spots of the STR polymorphism were located between the first and second conserved sequence blocks (CSB1 and CSB2), as observed previously in some other mammals. The geographical distribution of the STR polymorphism revealed local differences; the northeastern population was polymorphic with three STR haplotypes, but other local populations were simply monomorphic with a single STR haplotype. Molecular phylogenetic analysis with reported sequences from outside Bangladesh indicated a low substitution diversity of mtDNA in Bangladesh. Clustering results suggested a close relationship to India and divergence from Laos and China.  相似文献

Mitochondrial DNA was long believed to be purely clonal and free from recombination. Major phylogenetic studies still depend on such assumptions. The peculiar genetic system of marine mussels Mytilus in which two divergent mitochondrial genomes exist provides a unique opportunity to study mtDNA recombination. Previous reports showed the existence of a few haplotypes having very strong recombination signal in the control region of mtDNA. Those recombinant variants have been found in a Baltic Sea population of Mytilus trossulus as well as in Mytilus galloprovincialis from the Black Sea. In both cases the mosaic genomes switched their transmission route and have been inherited paternally. In the present study rearranged mtDNA genomes found in all three European Mytilus species are described. The structure of their control region is a result of intra- and intermolecular recombination between mitochondrial genomes. Together with the phylogenetic reconstruction and geographic distribution, this suggests that two interlineage recombination events have occurred in the control region of mtDNA of European mussels Mytilus. Contrary to earlier observations, some of the mosaic genomes do not show any gender bias, which has important implications regarding the transmission and evolution of blue mussel mitochondrial genomes.  相似文献

系统评估地方鸡的遗传变异水平并追溯其母系起源, 可为保护利用优质家禽种质资源库提供科学依据。本研究测定了广东省和邻省共12个地方鸡品种的线粒体DNA D-loop序列, 分析品种间的遗传距离与系统关系, 并构建单倍型系统发生树和中介网络图。360份样品共检测到60个突变位点, 均为转换。定义了85种单倍型, 归属于单倍型类群A、B、C和E, 在12个鸡品种中均有分布, 其中B是优势单倍型类群(187个, 51.94%), E次之(76个, 21.11%)。B02 和C01是优势单倍型(85个, 23.61%; 48个, 13.33%), 为12个鸡品种共有; E03位居第三(35个, 9.72%), 杏花鸡、黄郎鸡和宁都三黄鸡未见此单倍型。杏花鸡集中分布在单倍型类群B, 惠阳胡须鸡和中山沙栏鸡则主要分布在单倍型类群E; 怀乡鸡的单倍型数量最多, 中山沙栏鸡的最少。广东地方鸡品种间遗传距离为0.012-0.015, 单倍型多样性0.805 ± 0.047至0.949 ± 0.026, 核苷酸多样性0.0102 ± 0.0017至0.0138 ± 0.0009。邻接树和中介网络图将85种单倍型划分为进化枝A、B、C和E, 广东省与邻省地方鸡单倍型的地理分布模式相似。中性检验显示广东地方鸡未经历明显的群体历史扩张。结果表明广东地方鸡处于较好的保护状态, 遗传多样性水平较高, 品种的形成受到邻省和北方家鸡的影响, 东南亚红原鸡对广东地方鸡也有重要的遗传贡献。  相似文献

The Azorean bat Nyctalus azoreum is the only endemic mammal native to the remote archipelago of the Azores. It evolved from a continental ancestor related to the Leisler's bat Nyctalus leisleri and is considered threatened because of its restricted and highly fragmented distribution. We studied the genetic variability in 159 individuals from 14 colonies sampled throughout the archipelago. Sequences of the D-loop region revealed moderate but highly structured genetic variability. Half of the 15 distinct haplotypes were restricted to a single island, but the most common was found throughout the archipelago, suggesting a single colonization event followed by limited interisland female gene flow. All N. azoreum haplotypes were closely related and formed a star-like structure typical of expanded populations. The inferred age of demographic expansions was consistent with the arrival of founder animals during the Holocene, well before the first humans inhabited the Azores. Comparisons with a population of N. leisleri from continental Portugal confirmed not only that all N. azoreum lineages were unique to the archipelago, but also that the current levels of genetic diversity were surprisingly high for an insular species. Our data imply that the Azorean bat has a high conservation value. We argue that geographical patterns of genetic structuring indicate the existence of two management units.  相似文献

Genetic variation was surveyed at nine microsatellite loci and the mitochondrial control region (868 bp) to test for the presence of genetic stock structure in young-of-the-year Atlantic bluefin tuna (Thunnus thynnus thynnus) from the Mediterranean Sea. Bluefin tuna were sampled over a period of 5 years from the Balearic and Tyrrhenian seas in the western basin of the Mediterranean Sea, and from the southern Ionian Sea in the eastern basin of the Mediterranean Sea. Analyses of multilocus microsatellite genotypes and mitochondrial control region sequences revealed no significant heterogeneity among collections taken from the same location in different years; however, significant spatial genetic heterogeneity was observed across all samples for both microsatellite markers and mitochondrial control region sequences (FST=0.0023, P=0.038 and PhiST=0.0233, P=0.000, respectively). Significant genetic differentiation between the Tyrrhenian and Ionian collections was found for both microsatellite and mitochondrial markers (FST=0.0087, P=0.015 and PhiST=0.0367, P=0.030, respectively). These results suggest the possibility of a genetically discrete population in the eastern basin of the Mediterranean Sea.  相似文献
