Mutants altered in their response to auxins and cytokinins have been isolated in the moss Physcomitrella patens either by screening clones from mutagenized spores for growth on high concentrations of cytokinin or auxin, in which case mutants showing altered sensitivities can be recognized 3–4 weeks later, or by non-selective isolation of morphologically abnormal mutants, some of which are found to have altered sensitivities. Most of the mutants obtained selectively are also morphologically abnormal. The mutants are heterogeneous in their responses to auxin and cytokinin, and the behaviour of some is consistent with their being unable to make auxin, while that of others may be due to their being unable to synthesize cytokinin. Physiological analysis of the mutants has shown that both endogenous auxin and cytokinin are likely to play important and interdependent roles in several steps of gametophytic development. Although their morphological abnormalities lead to sterility, genetic analysis of some of the mutants has been possible by polyethyleneglycol induced protoplast fusion.Abbreviations NTG

Effect of benzyladenine on the development of plastids and microbodies in excised watermelon cotyledons 总被引:3,自引:0,他引:3
Excised watermelon (Citrullus vulgaris Schrad.) cotyledons were grown in the dark in the presence of 0.1 mM benzyladenine (BA). Under these conditions reserve breakdown and organelle differentiation progress very slowly. Treatment with BA accelerates, breakdown of reserves and stimulates development of organelles. Electron micrographs of cells from treated cotyledons show a larger number of plastids with a more developed inner membrane system. The levels of plastid pigments and enzymes are increased while starch content is reduced. Glyoxysomal enzyme levels are increased by BA during the first three days of development and their decline is accelerated thereafter. Also the activity of hydroxypyruvate reductase (EC 1.1.1.81.), a peroxisomal enzyme, is increased, but this increase is not followed by a decay phase. In water controls, hydroxypyruvate reductase bands together with glyoxysomal enzymes after equilibrium centrifugation in a sucrose gradient. In treated cotyledons the equilibrium position of glyoxysomal enzymes is uchanged while that of hydroxypyruvate reductase is shifted to a lower density.Abbreviations BA

At least two types of cytokinin-binding sites are present in a particulate fraction of tobacco (Nicotiana tabacum L.) cells that sediments at 80,000 x g. The major binding component has a low affinity towards cytokinins, is resistant to heating at 100°C, and is not specific for biologically active cytokinin analogues. The second site occurs in much lower frequency, is heat labile, shows high affinity towards cytokinins, and is specific for biologically active analogs of the hormone. The testing for binding specificity was mainly performed with a series of halogenated benzyladenine derivatives having a wide range of biological activities. The low-affinity binding site shows some of the same features as talcum powder, a non-biological material which binds cytokinins in a non-specific fashion. The properties of the high-affinity binding site are consistent with the expected characteristics of a cytokinin receptor. However, the role of the observed high-affinity binding site with regard to the biological action of cytokinins is not yet known.Abbreviations BA

Radish seedlings (Raphanus sativus L. Saxa Treib) were grown in the dark with or without added kinetin (2 mg/l=9.29 M). Low-temperature (77°K) fluorescence emission and absorption spectra of etiolated cotyledons were registered at increasing seedling age before and immediately, 30 s and 30 min after one 1-ms flash. Kinetin was found to induce a higher accumulation of the phototransformable protochlorophyll(ide) P657–650 in the etiolated cotyledons, especially from day 6 to day 10 after germination. The amount of the P657–650 protochlorophyll(ide) resynthesized during a 30-min dark period after a 1-ms flash decreased with seedling age. It was smaller in cotyledons from kinetin-treated seedlings at day 6 after germination and at that age only. The ability to perform the Shibata shift decreased with increasing seedling age. In cotyledons from 10- and 13-day-old seedlings, the shift was accomplished to a greater extent when the plants were grown in the presence of kinetin. 相似文献

A cytokinin was isolated from the culture medium of callus cells of the moss hybridFunaria hygrometrica (L.) Sibth xPhyscomitrium piriforme Brid. The purification procedure included ethyl-acetate extraction, silver-salt precipitation, crystallization as picrate, and ion exchange chromatography. The structure of the cytokinin was confirmed as N6–(2-isopentenyl)adenine by means of gas chromatography and mass spectrometry. A cytokinin photoaffinity reagent, 8-azido-N
6-benzyladenine (8N3BA), was synthesized from 8-bromoadenosine via azide replacement, benzylation at N–1, rearrangement to the N-6-benzyl derivative and acid hydrolysis. The compound thus obtained was found to have full cytokinin activity in the moss and tobacco cell-suspension bioassays. Photolysis of 8N3BA was accomplished with long and short-wavelength ultraviolet light and produced compounds which had very little or no biological activity in the two bioassays. In-vivo photolysis of 8N3BA caused loss of the cytokinin activity of this compound in moss protonemata. This result was similar to earlier ones where the biological response of moss protonemata to benzyladenine was reversed following removal of the hormone by a short rinse with water.Abbreviations BA

Cytokinin bases (zeatin and dihydrozeatin) and ribosides (zeatin riboside and dihydrozeatin riboside) were identified as major cytokinins in tobacco xylem sap by radioimmunoassay. When 3 H-labelled zeatin riboside or dihydrozeatin riboside were supplied to tobacco plants via the xylem, leaves of differing maturity did not differ appreciably in level of radioactivity or in metabolism of the cytokinin. The major metabolites of zeatin riboside in leaves were adenine, adenosine and adenine nucleotides, whereas that of dihydrozeatin riboside was dihydrozeatin 7-glucoside. Incorporation of [14 C]adenine into zeatin was evident in upper green leaves. indicating that young leaves have the capacity to synthesize cytokinins in situ. In contrast, fully expanded green leaves and senescing tobacco leaves exhibited little or no incorporation of [14 C]adenine into cytokinins. This difference in cytokinin biosynthetic capacity may contribute to the differing cytokinin levels in leaves of different matirity, and may participate in control of sequential leaf senescence in tobacco. 相似文献

Liliurn davidii Duch. pollen was germinated in PEG-400. A rapid immunoaffinity chromatograpy system with high efficiency and specificity was adopted to isolate and purify trans-zeatin riboside (t-ZR) and isopentenyladenosine (ipA). Cytokinin determination was carried out by a highly sensitive enzyme linked immunosolvent assay (ELISA) for t-ZR and iPA respectively, t-ZR and iPA were present at the level of 10-8 g/g fr. wt in Lilium davidii pollen. After hydration, t-ZR content of pollen decreased slightly, while iPA increased remarkably, and the total amount of t-ZR and iPA underwent almost no change. Pollen tubes grew fast in the first 3 h after germination, and the amount of t-ZR and iPA also increased greatly both in pollen tubes and in germination medium. The expression characteristics of a class of pathogenesis-related protein (PR) genes, namely basic chitinase, β-1, 3-glucanase, osmotin and extensin, were studied in tobacco (Nicotiana tabacum cv. Wisconsin 38) plants. RNA blot hybridization showed that these four genes were regulated in a developmental and organ-specific manner in tobacco. In the transgenic fascicular shoots which contained the active cytokinin biosynthetic gene (ipt gene) from Agrobacterium tumefaciens, the expressions of these four genes were co-regulated by overproduction of endogenous cytokinins and vector effect. Cytokinins reduced the expressions while vector effect induced the expressions of these four genes. Heat shock also de creased the steady-state levels of the four RNAs. These data suggest a complex regulation of PR genes. 相似文献
