Global contractile heart failure was induced in turkey poults by furazolidone feeding (700 ppm). Abnormal calcium regulation
appears to be a key factor in the pathophysiology of heart failure, but the cellular mechanisms contributing to changes in
calcium fluxes have not been clearly defined. Isolated ventricular myocytes from non-failing and failing hearts were therefore
used to determine whether the whole heart and ventricular muscle contractile dysfunctions were realized at the single cell
level. Whole cell current- and voltage-clamp techniques were used to evaluate action potential configurations and L-type calcium
currents, respectively. Intracellular calcium transients were evaluated in isolated myocytes with fura-2 and in isolated left
ventricular muscles using aequorin. Action potential durations were prolonged in failing myocytes, which correspond to slowed
cytosolic calcium clearing. Calcium current-voltage relationships were normal in failing myocytes; preliminary evidence suggests
that depressed transient outward potassium currents contribute to prolonged action potential durations. The number of calcium
channels (as measured by radioligand binding) were also similar in non-failing and failing hearts. Isolated ventricular muscles
from failing hearts had enhanced inotropic responses, in a dose-dependent fashion, to a calcium channel agonist (Bay K 8644).
These data suggest that changes in intracellular calcium mobilization kinetics and longer calcium-myofilament interaction
may be able to compensate for contractile failure. We conclude that the relationship between calcium current density and sarcoplasmic
reticulum calcium release is a dynamic process that may be altered in the setting of heart failure at higher contraction rates.

Trichomoniasis caused by the parasitic protozoan Trichomonas vaginalis is the most common sexually transmitted disease in the world. Dendritic cells are antigen presenting cells that initiate immune responses by directing the activation and differentiation of naïve T cells. In this study, we analyzed the effect of Trichomonas vaginalis-derived Secretory Products on the differentiation and function of dendritic cells. Differentiation of bone marrow-derived dendritic cells in the presence of T. vaginalis-derived Secretory Products resulted in inhibition of lipopolysaccharide-induced maturation of dendritic cells, down-regulation of IL-12, and up-regulation of IL-10. The protein components of T. vaginalis-derived Secretory Products were shown to be responsible for altered function of bone marrow-derived dendritic cells. Chromatin immunoprecipitation assay demonstrated that IL-12 expression was regulated at the chromatin level in T. vaginalis-derived Secretory Productstreated dendritic cells. Our results demonstrated that T. vaginalis-derived Secretory Products modulate the maturation and cytokine production of dendritic cells leading to immune tolerance. Dysregulation of cytokine expression causes inflammatory diseases or chronic
infection conditions. We have identified that Tat-activating regulatory
DNA-binding protein-43 (TDP-43) is involved in cytokine RNA processing in order
to promote an optimal immune response. The interaction of TDP-43 with
spliceosomal components from the Cajal body leads to the formation of a novel
sub-nuclear body called the Interleukin (IL)-6 and IL-10 Splicing Activating
Compartment (InSAC). TDP-43 binds to the IL-6 and IL-10 RNAs in a
sequence-dependent manner. In cell-based studies, we observed that
lipopoly-saccharide (LPS) stimulation induces the formation of the InSAC through
TDP-43 ubiquitination, thereby influencing the processing and expression levels
of IL-6 RNA. Moreover, TDP-43 knockdown in vivo results in a
decrease in IL-6 production and its RNA splicing and stability. Thus, these
findings demonstrate that the InSAC is linked to the activation and modulation

Many cell lines, despite the fact that they are easy to culture, tend to lose some of their in vivo characteristics in vitro, we therefore decided to investigate whether culturing HK-2 cells on kidney derived micro-scaffolds (KMS) could improve proximal tubule functionality to these cells. Kidney derived micro-scaffolds (KMS) have been prepared that, due to the fact that they are only 300 µm in depth, allow for transfer of gasses and nutrients via diffusion whilst maintaining the kidney's intricate microstructure. Culturing HK-2 on KMS shows significant increase in expression of AQP-1, ATP1B1, SLC23A1 and SLC5A2 after 1, 2 and 3 weeks compared with HK-2 grown under standard tissue culture conditions. Additionally, very high levels of expression of CCL-2 (15–30 fold increase) and LRP-2 (25–200 fold increase) were observed when the HK-2 were grown on KMS compared with HK-2 grown under standard tissue culture conditions. Furthermore, HK-2 cells grown under standard conditions released higher levels of Il-6 and Il-8 compared with primary tubule cells (Asterand AS-9-2) and secreted no MCP-1 or RANTES as opposed to primary cells that released MCP-1 and RANTES following stimulation. However, HK-2 grown on KMS showed both a marked decrease in Il-6/Il-8 secretion in line with the primary cells and secreted MCP-1 as well. Exposure to environmental endocrine-disrupting chemical Bisphenol-A (BPA) is often associated with dysregulated immune homeostasis, but the mechanisms remain unclear. In the present study, the effects of BPA on the cytokines responses of human macrophages were investigated. Treatment with BPA increased pro-inflammation cytokines tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) production, but decreased anti-inflammation cytokines interleukin-10 (IL-10) and transforming growth factor-β (TGF-β) production in THP1 macrophages, as well as in primary human macrophages. BPA effected cytokines expression through estrogen receptor α/β (ERα/β)-dependent mechanism with the evidence of ERα/β antagonist reversed the expression of cytokines. We also identified that activation of extracellular regulated protein kinases (ERK)/nuclear factor κB (NF-κB) signal cascade marked the effects of BPA on cytokines expression. Our results indicated that BPA effected inflammatory responses of macrophages via modulating of cytokines expression, and provided a new insight into the link between exposure to BPA and human health. 相似文献

Assessment of the potential risks posed by chlorinated solvents in groundwater is the key to establish the extent of the contamination and derive achievable remedial targets should remediation deems necessary. This article first presents the application of the American Society for Testing and Materials (ASTM) Risk Based Corrective Actions (RBCA) Guidance to quantitatively evaluate human health and environmental risk for a former chemical works in Shanghai, China. The observed maximum trichloroethylene (TCE) concentration in groundwater at the site reached 1220 mg/l that exceeded its solubility of 1070 mg/l at 10°C (Soil annual average temperature is 10°C in Shanghai). The maximum concentration for cis-1, 2-DCE (DCE) was also found to be elevated at 264 mg/l. A critical exposure pathway was considered to be indoor vapor intrusion of TCE into the buildings with excess lifetime cancer risk for children being 1.7 × 10?3. This cancer risk exceeded regulatory limits of 1 × 10?4 to 1 × 10?6 for The Netherlands, the United Kingdom, and the United States. The calculated groundwater remedial targets for TCE and DCE are 7 mg/l and 904 mg/l, respectively, in order to protect child residents from inhalation of indoor vapors within the close proximity of the source area. 相似文献
