Autophagy is an intracellular degradation process for recycling macromolecules and organelles. It plays important roles in plant development and in response to nutritional demand, stress, and senescence. Organisms from yeast to plants contain many autophagy-associated genes (ATG). In this study, we found that a total of 33 ATG homologues exist in the rice [Oryza sativa L. (Os)] genome, which were classified into 13 ATG subfamilies. Six of them are alternatively spliced genes. Evolutional analysis showed that expansion of 10 OsATG homologues occurred via segmental duplication events and that the occurrence of these OsATG homologues within each subfamily was asynchronous. The Ka/Ks ratios suggested purifying selection for four duplicated OsATG homologues and positive selection for two. Calculating the dates of the duplication events indicated that all duplication events might have occurred after the origin of the grasses, from 21.43 to 66.77 million years ago. Semi-quantitative RT–PCR analysis and mining the digital expression database of rice showed that all 33 OsATG homologues could be detected in at least one cell type of the various tissues under normal or stress growth conditions, but their expression was tightly regulated. The 10 duplicated genes showed expression divergence. The expression of most OsATG homologues was regulated by at least one treatment, including hormones, abiotic and biotic stresses, and nutrient limitation. The identification of OsATG homologues showing constitutive expression or responses to environmental stimuli provides new insights for in-depth characterization of selected genes of importance in rice.  相似文献

臭氧是氧气的同素异形体,在常温条件下它是一种有特殊臭味的淡蓝色气体。臭氧现已成为我国主要的一种环境污染物。臭氧的分布存在着明显的时间特异性和地区特异性,与气象条件、污染物的输送与扩散以及局地光化学反应强度等因素有着重要的关系。同时在人群流行病学和动物实验中发现了臭氧可以提高慢性阻塞性肺疾病、哮喘等呼吸系统疾病的发病率和病死率,同时提示了老年人、有呼吸系统疾病病史的人群和儿童都是易受臭氧影响的高危人群。国内外学者也对臭氧对机体损伤的机制进行了研究,主要集中在臭氧的致炎作用、氧化应激作用、造成机体气道高反应性和对细胞DNA的损伤等方面。本文主要就国内外对臭氧在时空分布、对机体呼吸系统影响和臭氧造成机体损伤的机制这3方面作一简要综述。  相似文献

Sarcobatus vermiculatus (Hook) Torrey is a leaf succulent, sodium-accumulating shrub usually found in saline substrates of the Great Basin desert, Utah, USA. Laboratory experiments were conducted to determine the effect of salinity (0, 200, 400, 600, 800 and 1000 mM NaCl) and temperature (day/night: 5/15, 10/20, 15/25, 20/30, and 25/35°C) on seed germination. S. vermiculatus showed 100% germination in non-saline controls, at all thermoperiods. Percentage and rate of germination decreased with increases in salinity and few seeds germinated at even 1000 mM NaCl. High salinity exposure caused the loss of viability at higher temperature regimes, while some recovery was recorded in low salinity treatments.  相似文献

AIMS: To study whether the exposure to cold (4 degrees C) and carbon dioxide which results in the elongation of Listeria cells, induces a viable but nonculturable (VBNC) state. METHODS AND RESULTS: When cold and CO2 stressed L. monocytogenes were observed under a fluorescence microscope, using the LIVE/DEAD BacLight bacteria viability kit (Molecular Probes, Eugene, OR, USA), the healthy, mildly injured, and the putative VBNC cells accounted for 31.0% of the stressed cell population. By using the selective plate count, 31.4% of the same stressed cell population was found to be healthy and mildly injured (putative VBNC cells not included). If there were VBNC state cells present, we should have observed a significant difference between the above two numbers. In fact, there was no significant difference between the results obtained from those two methods. CONCLUSIONS: There were no VBNC state cells observed in the stressed cell population. We conclude that cold and CO2 do not induce L. monocytogenes to enter a VBNC state. SIGNIFICANCE AND IMPACT OF THE STUDY: Cold and modified atmospheres are widely used in fresh muscle food and fruit preservation. Whether they would induce L. monocytogenes into a VBNC state is of a great concern for microbial food safety.  相似文献

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder resulting in cognitive decline and enhancement of oxidative loads in the brain. Flavonoids have been considered to exert human health benefits by anti-oxidant and anti-inflammatory properties. The present study is aimed to elucidate the neuroprotective effect of catechin hydrate (CH), a natural flavanoid with potential antioxidant and anti-inflammatory properties, on intracerebroventricular streptozotocin (ICV-STZ) induced neuronal loss and memory impairment. To test this hypothesis, male Wistar rats were pretreated with CH (10 and 20 mg/kg bwt) orally once daily for 21 days and then bilaterally injected with ICV-STZ (3 mg/kg bwt), while sham group rats receive the same volume of vehicle. After 2 weeks of ICV-STZ infusion, rats were tested for cognitive performance using Morris water maze (MWM) test and then sacrifice for biochemical and histopathological assays. CH was found to be successful in upregulating the antioxidant status and prevented the memory loss. The expression of choline acetyl transferase (ChAT) was decreased in ICV-STZ group and CH pretreatment increases the expression of ChAT. Moreover, inflammatory mediators like TNF-α, IL-1β levels and expression of iNOS were significantly attenuated by CH pretreatment. The study suggests that CH is effective in preventing memory loss, ameliorating the oxidative stress and might be beneficial for the treatment of sporadic dementia of Alzheimer’s type (SDAT).  相似文献

桉树幼苗对难溶性磷的吸收及其根系对低磷胁迫的响应   总被引:2,自引:0,他引:2  
以‘广林9号’桉树幼苗为试验材料,采用水培和土培试验方法研究了桉树幼苗对难溶性磷酸盐的吸收及其在低磷胁迫下的根构型和根系的生理反应,以揭示桉树高效吸收磷素的机制。结果显示:(1)桉树幼苗在含磷酸铝的缺磷培养液中吸收的磷达4.24mg/株,与供应水溶性磷和磷酸钙处理的相当。(2)土壤缺磷或仅在上土层(0~20cm)施磷肥处理均有利于桉树幼苗浅层根的分布,使根表面积及根数在上土层与下层(20~40cm)比值明显增高。(3)桉树幼苗根尖的H+-ATPase活性在缺磷处理15d后显著提高,其根尖周围的溴甲酚紫指示剂变黄,根基环境明显酸化;根尖分泌的酸性磷酸酶活性在低磷胁迫也显著提升,且随着处理时间(10、15、20d)的延长而进一步提高;铝和低磷胁迫能明显诱导桉树根系分泌草酸,其分泌量显著高于对照和缺磷处理。研究结果表明,桉树幼苗具有较强的难溶性磷吸收能力,而在缺磷及磷铝胁迫下根系的浅层化、根尖酸化及根分泌的酸性磷酸酶及草酸量增加可能是桉树幼苗适应酸性土壤铝毒和缺磷环境的重要机制。  相似文献

Foundation seaweed species are experiencing widespread declines and localized extinctions due to increased instability of sea surface temperature. Characterizing temperature thresholds are useful for predicting patterns of change and identifying species most vulnerable to extremes. Existing methods for characterizing seaweed thermal tolerance produce diverse metrics and are often time-consuming, making comparisons between species and techniques difficult, hindering insight into global patterns of change. Using three kelp species, we adapted a high-throughput method – previously used in terrestrial plant thermal biology – for use on kelps. This method employs temperature-dependent fluorescence (TF0) curves under heating or cooling regimes to determine the critical temperature (Tcrit) of photosystem II (PSII), i.e., the breakpoint between slow and fast rise fluorescence response to changing temperature, enabling rapid assays of photosynthetic thermal tolerance using a standardized metric. This method enables characterization of Tcrit for up to 48 samples per two-hour assay, demonstrating the capacity of TF0 curves for high-throughput assays of thermal tolerance. Temperature-dependent fluorescence curves and their derived metric, Tcrit, may offer a timely and powerful new method for the field of phycology, enabling characterization and comparison of photosynthetic thermal tolerance of seaweeds across many populations, species, and biomes.  相似文献
