Recently, Morchella importuna, M. sextelata, M. eximia, M. exuberans, Mel-13, and Mel-21 have been successfully cultivated in China and planting areas rapidly expanded because of their economic importance. Effective molecular markers are urgently needed for accurately identifying morel cultivars. Microsatellites are widely utilized for strain authentication in many fungal groups. To our knowledge, for the first time we characterized the distribution of microsatellites (simple sequence repeats, SSRs) in the M. importuna genome with 12902 SSRs and reported the first set of SSRs developed for Morchella species. Mono-nucleotides (66.2 %) were the most frequent motifs, followed by tri- (15.4 %), di- (12.1 %), tetra- (3.7 %), penta- (1.3 %) and hexa-nucleotides (1.3 %). We tested the cross-species amplification of 180 SSRs on 24 samples from the six species and high cross-species transferability of SSRs (87.7 %) was found. Among twenty-two microsatellites selected for genetic diversity analysis on 127 samples from the six species, fifteen to twenty polymorphic loci were identified in M. importuna, M. sextelata, M. eximia, M. exuberans, Mel-13 and Mel-21. Interspecific hybridization events were detected among morel species, indicating the potential application of morel crossbreeding. Ninety-one cultivated samples were characterized as new cultivars with different genotypes, but cultivar names used for these by farmers was confusing, with misnaming, synonyms and homonyms. Our results are not only helpful for cultivar identification and morel breeding programs in China, but also provide molecular tools for genetic studies in morels.  相似文献

The symbiotic performance of four pea ( Pisum sativum L.) cultivars in combination with each of four strains of Rhizobium leguminosarum was studied in growth chamber experiments in order to estimate the effects of cultivars, strains and cultivar × strain interaction on the variation in dry weight, N content and dry weight/N ratio. At harvest 63 days after planting, cultivars accounted for 75% of the variation in dry weight, while the Rhizobium strains accounted for 63% of the variation in N-content and 70% of the variation in dry weight/N ratio. Cultivar × strain interactions were statistically significant, but of minor quantitative importance, accounting for 5–15% of the total variation. Rhizobium strains also influenced the partitioning of N between reproductive and vegetative plant parts and between root and shoot biomass.  相似文献

Summary Three gram varieties varying in susceptibility to pre-emergence damping-off caused byRhizoctonia bataticola were compared for amounts of seed exudation. The results showed that the greatest seed exudation occurred with the variety G-130 which is the most susceptible to pre-emergence damping-off compared with the resistant varieties BG-203 and C-214. Germinating gram seeds exuded greater quantities of soluble carbohydrate and amino acids when incubated at 35°C than at 15 or 25°C. Exudates from seeds germinated at 35°C stimulated more mycelial growth ofR. bataticola than did exudates from seeds incubated at 15 or 25°C. Evidence is presented suggesting that increased seed exudation is a major factor contributing to increased pre-emergence damping-off of gram seedlings byR. bataticola at high temperatures.  相似文献

 Four zones of superoxide dismutase (SOD), activity were detected in apple. Genetic studies confirmed the presence of three genes Sod-1, Sod-3 and Sod-4 with three, two and two alleles respectively, including one null allele. One of these genes, Sod-1, was found to be loosely linked to Prx-2 and Prx-3. The Sod-1a allele predominated in all the three groups studied, cultivars, rootstocks and Malus species. The distinct achromatic bands produced by SOD after electrophoresis facilitate its important role with peroxidase (PRX), glutamate oxalacetate transaminase (GOT) and phosphogluconate dehydrogenase (PGD) in the discrimination of apple cultivars.  Inter-simple sequence repeat (ISSR) markers generated by 22 primers were tested for their ability to distinguish among samples from 94 trees of 68 citrus cultivars. Within each of the six cultivar groups studied, most of these cultivars are so closely related that they are difficult to distinguish by other molecular-marker techniques. ISSR markers involve PCR amplification of DNA using a single primer composed of a microsatellite sequence anchored at the 3′ or 5′ end by 2–4 arbitrary, often degenerate, nucleotides. The amplification products were separated on non-denaturing polyacrylamide gels and detected by silver staining. ISSR banding profiles were very repeatable on duplicate samples. Different citrus species had very different fingerprint patterns. Within Citrus sinensis (L.) Osbeck and C. paradisi Macf., in which all cultivars have originated by the selection of mutants, ISSR markers distinguished 14 of 33 sweet orange and 1 of 7 grapefruit cultivars. Five of six lemon cultivars were discriminated by ISSR markers. Many differences were found among mandarin cultivars; however, all five satsuma cultivars analyzed had identical ISSR fingerprints. Four of five citrange cultivars were distinguishable, but ‘Troyer’ and ‘Carrizo’ had identical ISSR fingerprints. ‘Kuharske Carrizo’ citrange, which has better citrus nematode resistance than other ‘Carrizo’ citrange accessions, had unique ISSR fingerprints. Three ISSR markers that differentiated certain sweet orange cultivars were hybridized to Southern blots of sweet orange DNA digested with different restriction endonucleases. The sweet orange cultivars tested could be distinguished by these ISSR-derived RFLP markers. Moreover, one ISSR marker unique to ‘Ruby’ blood orange was observed in its progeny trees. Isozyme polymorphism can assist in the identification of cherry cultivars. Ten isozymes, each showing variation at only one locus, provide 70 unique genotype profiles from leaf extracts of 78 different sweet cherry cultivars. Polymorphism in 6PGD, G6PD, GPI, IDH, PGM, FDP, SKDH and PER is demonstrated for the first time, while observations are extended for the previously described polymorphisms in MDH and GOT. Some cultivars with identical morphological characters and previously treated as one cultivar can be separated on the basis of isozyme genotype. Irradiated mutants and parent material could also be differentiated.  相似文献

牛鞭草品种EST-SSR指纹图谱构建及遗传多样性分析   总被引:1,自引:0,他引:1  
为探讨牛鞭草(Hemarthria spp.)品种间的遗传多样性,从86对EST-SSR引物中筛选出8对引物对牛鞭草属6个品种进行指纹图谱的构建及遗传多样性分析。结果表明,8对EST-SSR引物对牛鞭草属6个品种共扩增出193条清晰条带,多态性条带161条,多态性比例为83.4%。每条引物的多态信息含量(PIC)为0.480~0.695,平均为0.602。UPGMA聚类分析表明,牛鞭草属6个品种在相似系数为0.652处可分为两大类群。8对EST-SSR引物均能将6个品种完全区分开,以3对EST-SSR引物扩增的电泳图谱为基础,建立了牛鞭草属6个品种的指纹图谱标准模式图,每个品种都有唯一的指纹图谱。牛鞭草属6个品种的平均Nei’s基因多样性指数为0.333,平均Shannon信息指数为0.496,品种间的相似系数介于0.399~0.782之间。可见,牛鞭草属植物品种的遗传多样性较丰富,种间差异明显。  相似文献

To set up a rational collecting strategy for germplasm of the edible-seeded cucurbit Cucumeropsis mannii, a study was conducted using 24 morphological and seven putative enzyme markers to determine the intra-specific variability from 16 and 22 accessions (representing three cultivars), respectively. The analysis of variance, showed a significant difference between the three cultivars. Principal component analysis pointed out a variation among individuals, mainly on the basis of flower, fruit, and seed size. Dendrogram with UPGMA method allowed clustering of the cultivars. Genetic diversity indices estimated equalled: 9.96% for the proportion of polymorphic loci (P), 1.10 for the number of alleles (A) and 0.023 for observed heterozygosity (Ho). The level of the within accessions genetic diversity (HS = 0.078) was higher than among accessions (DST = 0.042). Nei's genetic distances between the three cultivars were also low (0.079–0.147), indicating a high degree of similarity of the analysed cultivars.  相似文献

从野生稻遗传资源在育种中的作用出发,概述了栽培稻与野生稻远缘杂交和多倍体化在水稻育种中的应用。对远缘杂交中的杂交不亲和性以及杂种不育等问题采用激素处理、胚挽救等途径解决,对所获杂种从形态学、细胞学、生物化学以及分子生物学等多方面鉴定,对栽野杂种存在的野生性状通过选择与改造相结合的方法而淘汰,对其有利基因则加以选择和聚合以获得稳定优良品种。这些从理论上和实践上证明远缘杂交和多倍体化相结合的异源多倍体化是获得高产优质水稻新品种的一条新途径。  相似文献

利用水培试验研究了不同Cd水平下大麦幼苗的Cd和几种矿质元素吸收、积累、生长和生物学产量的品种间差异 .结果表明 ,1μmol·L-1Cd处理显著降低麦苗株高、绿叶数、叶绿素计读数、地上部和根系干重 ,显著抑制植株对Zn、Mn、Cu的吸收和累积 ;品种之间存在着显著差异 ,无芒六棱受抑制最为严重 ,米麦 114和浙农 1号表现出相对较强的抗性 .麦苗Cd含量和累积量品种之间也有显著差异 ,浙农 1号的Cd含量最高 ,米麦 114最低 .相关分析表明 ,麦苗生物学产量与地上部Cd含量、累积量及根系Cd含量呈显著负相关 ,其中与地上部Cd含量的相关性最强 ,与根系Cd累积量无显著相关 .  相似文献
