Summary This paper describes the use of oligonucleotide probes to reveal highly polymorphic DNA regions in pomato. With a (GATA)4 probe the level of polymorphism detected is high enough to identify all 15 tomato cultivars used in this study. Individual plants of one cultivar all showed the same cultivar-specific DNA-finger-print. In an F2-population of self-fertilized cv. Sonatine, GATA-containing loci segregated in a Mendelian (31) fashion. Experiments with in-vitro propagated plant material showed that the DNA-fingerprints are not affected by tissue-culture procedures. This indicates that changes in the genetic integrity, which often accompany in-vitro propagation (somaclonal variation), are not extended to the DNA detected with the (GATA)4 probe. The relative high stability and the Mendelian segregation of (GATA)4-derived DNA-fingerprints make them ideally suited for identification of tomato cultivars.  相似文献

本文对黄瓜的四个不同品种(“津一号”、“津二号”、“津四号”和“一串连”)进行了核型及倍性变化的研究,染色体数目为2n=14,其染色体组成为2n=2x=14=14m(2SAT)均为中部着丝点染色体,每一品种只有一对随体染色体,“津一号”和“津二号”还具有一个次级缢痕。黄瓜体细胞倍性的变化有3n、4n和6n,随着细胞倍性增高,其多倍性细胞频率依次下降,不同品种倍性的变化以“津四号”为最高,“一串连”品种为最低。  相似文献

Twenty-five accessions of mango were examined for random amplified polymorphic DNA (RAPD) genetic markers with 80 10-mer random primers. Of the 80 primers screened, 33 did not amplify, 19 were monomorphic, and 28 gave reproducible, polymorphic DNA amplification patterns. Eleven primers were selected from the 28 for the study. The number of bands generated was primer- and genotype-dependent, and ranged from 1 to 10. No primer gave unique banding patterns for each of the 25 accessions; however, ten different combinations of 2 primer banding patterns produced unique fingerprints for each accession. A maternal half-sib (MHS) family was included among the 25 accessions to see if genetic relationships could be detected. RAPD data were used to generate simple matching coefficients, which were analyzed phenetically and by means of principal coordinate analysis (PCA). The MHS clustered together in both the phenetic and the PCA while the randomly selected accessions were scattered with no apparent pattern. The uses of RAPD analysis for Mangifera germ plasm classification and clonal identification are discussed.  相似文献

Summary Isozyme analysis of wild and domesticated accessions indicated that domestication of the cultivated carrot Daucus carota ssp. sativus resulted in an insignificant reduction of all genetic variability and genetic distance estimates. Although they are less variable genetically, cultivated forms maintain a high proportion of observed heterozygosity. Relative to the overall genetic variability of the species, samples from four common cultivars Red Cored Chantenay, Scarlet Nantes, Danvers Half Long and A Plus demonstrated a high degree of genetic similarity. This is attributed to the recent development of orange cultivars and the limited gene pool utilized in their development.  相似文献

The leafhopper Empoasca vitis (Göthe) (Homoptera: Cicadellidae) can cause economic damage in European vineyards. Egg parasitoids, in particular Anagrus atomus (Linnaeus) (Hymenoptera: Mymaridae), are the most important natural enemies of the leafhopper. In four different years, leaves of ten grapevine cultivars, which were grown in a vineyard of north-eastern Italy, were collected at the end of the leafhopper 2nd generation to determine the total number of E. vitis eggs per leaf and the percentage of the E. vitis eggs parasitized. These data were analysed for correlation with leaf density and foliar pubescence. The E. vitis eggs per leaf and the percentage of eggs parasitized by Anagrus spp. were significantly influenced by the cultivar. The number of E. vitis eggs per leaf was positively correlated to leaf density, but it was not influenced by leaf hair density. The parasitization rate by Anagrus spp. was affected by foliar pubescence, especially by erect hairs on the veins. These findings could be used for integrated pest management. In particular, (1) the more susceptible cultivars can be used as early indicators of leafhopper infestation, (2) agronomic practices, that reduce leaf density, could decrease the E. vitis population level and (3) clones with glabrous leaves would favour egg parasitoid activity.  相似文献

采用10份花生品种材料,对通过数据库查询设计合成的SSR引物和其他作者发表的SSR引物及STS引物进行了评估,并基于品种间简单匹配系数做了聚类分析。获得62个能产生多态性片段的SSR和STS引物对,总共获得427条带,其中291条(68.1%)为多态性带。平均每对引物产生6.88条带,4.69条为多态性带;多态性条带比率为16.7%~100%,PIC值为0.254~0.952,平均为0.760。9对SSR/STS引物,即Lec-1、Ah4-26、Ah4-4、SsS14、SHPAL-1、PG71、PG43、PM36、PG22,对所采用的10份花生品种区分率达到10096。说明SSR和STS标记应用于花生品种鉴定有效。  相似文献

Oilseed rape (Brassica napus) is one of the major oilseed crops in the world but is vulnerable to attack by many pathogens and insect pests. In addition to the host plant genotype, micro-organisms present in the rhizosphere and within plant tissues affect the susceptibility to plant pathogens. While rapid progress has been achieved concerning the concept of plant resistance genes, information on the role of the microbial community in plant protection is less apparent. We have studied the endophytic bacterial populations present in different tissues of oilseed rape and also analysed several cultivars (Express, Libraska, Maluka and Westar), which differ in their susceptibility to the wilt pathogen Verticillium longisporum. The population diversity was studied using agar plating assay, fatty acid methyl ester analysis and functional characterisation of isolated strains. Our work shows that already in the seeds there exists diversity in populations as well as in the total microbial load between two of the four tested cultivars. About 50% of the strains isolated from cultivars Express and Libraska showed moderate to strong direct inhibition of V. longisporum. The diversity of the endophytic flora isolated from oilseed rape and its implications in crop protection are discussed.  相似文献

简要回顾了中国桂花(Osmanthus fragrans Lour.)的研究历史,对桂花品种研究的现状和存在的问题进行了评述,介绍了申报桂花品种国际登录工作的意义和目前的工作进展。桂花品种的现代分类始于20世纪80年代,除了传统的形态分类以外,同工酶、分子标记等技术已用于桂花品种亲缘关系的研究中,但目前仍缺乏合理的品种分类原则和分类系统,品种调查、记载和命名比较混乱,因此应加强桂花品种形态学的基础研究。  相似文献
