Biting midges of the genus Culicoides (Diptera: Ceratopogonidae) play important roles in the transmission of viral diseases affecting wild and domestic ruminants and horses, including Bluetongue (BT) and African horse sickness (AHS) respectively. In southern Europe, BT has been largely transmitted by the classical Afro-Asian vector Culicoides imicola Kieffer. However, other species such as C. obsoletus Meigen, C. scoticus Downs & Kettle and C. pulicaris Linné may also be involved in BTV transmission. As a consequence of the discovery of C. imicola followed by BTV-2 outbreaks on the island of Corsica in October 2000, further studies on these biting midges have been carried out. To better characterize the evolution and phylogenetic relations of Culicoides, molecular analysis in parallel with a morphology-based taxonomic approach were performed. Phylogenetic analyses of French Culicoides species were undertaken using the ribosomal DNA (rDNA) internal transcribed spacer 1 (ITS1) as a molecular target. This region was shown to be useful in understanding evolutionary and genetic relationships between species. Construction of several trees showed that molecular phylogeny within the genus Culicoides correlates not only with morphological-based taxonomy but also with ecological patterns.  相似文献

Switzerland is historically recognized by the Office Internationale des Epizooties as free from bluetongue disease (BT) because of its latitude and climate. With bluetongue virus (BTV) moving north from the Mediterranean, an entomological survey was conducted in Switzerland in 2003 to assess the potential of the BTV vectors present. A total of 39 cattle farms located in three geographical regions, the Ticino region, the Western region and the region of the Grisons, were monitored during the vector season. Farms were located in areas at high risk of vector introduction and establishment based on the following characteristics: annual average temperature > 12.5 degrees C, average annual humidity >or= 60%, cattle farm. Onderstepoort black light traps were operated at the cattle farms generally for one night in July and one night in September. A total of 56 collections of Culicoides (Diptera: Ceratopogonidae) were identified morphologically. Only one single individual of Culicoides (Avaritia) imicola, the major Old World vector of BTV, was found in July 2003 in the Ticino region, one of the southernmost regions of Switzerland. In the absence of further specimens of C. imicola from Switzerland it is suggested that this individual may be a vagrant transported by wind from regions to the south of the country where populations of this species are known to occur. Alternative potential BTV vectors of the Culicoides (Culicoides) pulicaris and Culicoides (Avaritia) obsoletus complexes were abundant in all sampled regions with individual catches exceeding 70 000 midges per trap night.  相似文献

This study was carried out in the northeast region of Konya in Turkey, between April and December 2004, to determine the vertical distribution of Culicoides (Diptera: Ceratopogonidae) larvae and pupae. Larvae and pupae were collected using the saturated sugar flotation technique in mud samples taken from a range of depths of up to 20 cm. In total, 282 adult Culicoides were obtained. The number of larvae and pupae decreased as the depth increased; at 2.5 cm, 97 Culicoides larvae and 73 Culicoides pupae were found. Larvae and pupae of C. nubeculosus (Meigen) and C. puncticollis (Becker) were mostly found in organic matter-rich breeding sites, while larvae and pupae of C. gejgelensis Dzhafarov and C. festivipennis Kieffer were mostly found in organic matter-poor breeding sites. Culicoides larvae and pupae were obtained largely in July and August.  相似文献

Abstract  A study of responses by Culicoides spp. to light traps with light-emitting diodes (LEDs) confirmed that the livestock virus vector, Culicoides brevitarsis Kieffer, was preferentially attracted to green light. Four species shown previously to respond to blue light exhibited significantly higher responses to ultraviolet (UV) light. Field trials comparing green LEDs with incandescent lights in New South Wales, Northern Territory and East Timor confirmed the superiority of green LEDs for catching C. brevitarsis . The green LED traps also had a significant advantage over incandescent traps for a wide range of Culicoides species. These included: species whose ultimate preference was for UV, most species that are proven or potential vectors of viruses affecting Australian livestock and native animals and the main species affecting humans in northern Australia. Use of green LEDs has been adopted for trapping C. brevitarsis , especially in its marginal and low-density areas, and for detecting incursions of new Culicoides spp. into Australia. Use could be possible for species in other insect Orders.  相似文献

Abstract.  1. Monthly light trap collections over 2 years at 39 sites across South Africa captured over 3 000 000 Culicoides of 86 species, some of them known vectors of African horse sickness and bluetongue viruses.
2. To aid disease control and risk evaluation, cluster analysis and association measurements were carried out on species present at each site. Six species occurred at almost all sites. From the remaining 80 species, eight clusters were identified at > 48% similarity.
3.  Culicoides citroneus and C. krameri were characteristic of cluster 8, C. loxodontis of cluster 4, and two numbered, but un-named, species, C. #119 and C. #89 , of cluster 5.
4. Multiple anova and multiple discriminant analysis on the climatic and geographic parameters of sites in each cluster gave two significant discriminant functions that explained 91.5% of the variance between clusters. Function 1 contrasted sites with high cold cloud duration and high vegetation index with sites with high wind speeds. Function 2 contrasted upland clusters that also had high cold cloud duration with hotter, lower altitude sites.
5. Species associations were explained in terms of biotic provinces, based on ecogeographic factors that have previously been useful to describe communities of birds.
6. None of the clusters was ideal for siting an African horse sickness-vector free quarantine station as C. imicola and C. bolitinos , the main vectors, were found in all clusters but in very low abundance in cluster 5. The analysis provides a firm basis for studying the changing patterns of species distribution under varying climatic influences.  相似文献

Effective surveillance is essential for protecting livestock from Culicoides biting midges and the viruses they transmit. The objective of this study was to determine how the baiting system used in traps (UV, incandescent light, incandescent light with CO2, and incandescent light with CO2 and 1‐octen‐3‐ol) influences estimates of midge population abundance, parity, and diel activity. This was achieved through a standardized trapping protocol conducted in three habitats in Sweden. UV light traps caught the most Culicoides species and more C. obsoletus complex females than incandescent light traps. Traps baited with CO2 plus 1‐octen‐3‐ol caught more female C. impunctatus than incandescent light traps. No consistent effect of bait type was found on C. obsoletus parity rate, as estimated from the proportion of midges with presence or absence of pigmentation. Midge activity, as reflected by trap catches, peaked between ‐3 h and +3 h relative to sunset, with UV traps catching significantly more female C. obsoletus complex and C. impunctatus at and after sunset than before sunset. We conclude that baiting system can influence biting midge collections, even using identical traps. Effective surveillance may require more than one bait type and kairomones to attract species that do not feed exclusively on cattle.  相似文献

To find out the effect of feed materials on the nutrient composition of Protaetia brevitarsis larvae (Pbl), three different feed materials were prepared and fed to the Pbl populations. After the Pbl population reached the third instar stage, they were boiled and dried for the nutrient analysis. The results showed that the nutrient composition of Pbl varied in response to the feed materials. Therefore, it is critical to set a feed material standard to guarantee the efficacy of Pbl medicinal property.  相似文献

Abstract.  Two experiments were undertaken to estimate the transmission rates of bluetongue virus (BTV) serotype 1 between a biting midge vector, Culicoides sonorensis (Wirth & Jones) (Ceratopogonidae), and a natural host, sheep. In an experiment to measure the transmission rate from vector to host (V→H), six batches of one, five and 20 intrathoracically infected midges were fed on a total of 18 bluetongue (BT)-naïve sheep. The sheep were then monitored for 21 days for clinical signs of BT, viraemia and antibody response. All sheep fed on by five or 20 midges and five of six sheep fed on by just one midge showed signs of BT, were viraemic and developed antibody. The sixth sheep fed on by a single infected midge did not show signs of BT or have detectable viraemia; it did, however, develop a weak antibody response. A bite from a single infected midge is therefore able to transmit BTV to naïve sheep with 80–100% efficiency. Sheep fed upon by larger numbers of infected midges took less time to reach maximum viraemia and developed stronger antibody responses. Sheep exposed to greater amounts of BTV in feeding midges developed a higher level of viraemia and stronger antibody responses. In a second experiment to measure the transmission rate from host to vector (H→V), batches of up to 500 uninfected female C. sonorensis fed every 1–2 days on two experimentally infected sheep during the course of infection. Of 3929 engorged midges that were individually titrated after surviving the extrinsic incubation period, only 23 (0.6%) were infected with BTV. Viraemia in the sheep extended for up to 19 days post-inoculation. No infected midges, however, were detected from 14 days post-infection.  相似文献
