The present research was conducted in district Jhang, Pakistan, to evaluate the concentration of metals/metalloids in soil and pumpkin (Cucurbita maxima) irrigated with domestic wastewater. Data revealed that the levels of metals and metalloids in soil samples from two different sites were below the safe limits except Cd, whereas, in the vegetable, the concentrations of As, Se, Ni, Mo, Pb, Mn, and Cu were above the safe limits. The levels of 12 metals and metalloids in the soil were ranged between 0.14 to 22.76 mg/kg at site-I and 0.16 to 22.13 mg/kg at site-II. The levels of these metals in the vegetable were found 0.35 to 61.13 mg/kg at site-I and 0.31 to 53.63 mg/kg at site-II. The transfer factor at both sites was highest for As and Co. The pollution load index recorded for Se, Cu, Cd, Mo, Pb, and Co was greater than 1. The daily intake of As, Mn, and Mo was above the oral reference dose, which reflects that the intake of pumpkin is not safe for the inhabitants of the selected sites. The control measures should be taken to phytoextract heavy metals and metalloids from polluted sites so as to reduce the health risks.  相似文献

Premise

Domestication of plant species results in phenotypic modifications and changes in biotic interactions. Most studies have compared antagonistic plant-herbivore interactions of domesticated plants and their wild relatives, but little attention has been given to how domestication influences plant-pollinator interactions. Floral attributes and interactions of floral visitors were compared between sister taxa of the genus Cucurbita (Cucurbitaceae), the domesticated C. moschata, C. argyrosperma ssp. argyrosperma and its wild progenitor C. argyrosperma ssp. sororia in the place of origin.

Methods

We conducted univariate and multivariate analyses to compare floral morphological traits and analyzed floral reward (nectar and pollen) quantity and quality between flowers of wild and domesticated Cucurbita taxa. Staminate and pistillate flowers of all three taxa were video recorded, and visitation and behavior of floral visitors were registered and analyzed.

Results

Most floral morphological characteristics of flowers of domesticated taxa were larger in both staminate and pistillate flowers. Staminate and pistillate flowers presented distinct correlations between floral traits and integration indices between domesticated and wild species. Additionally, pollen quantity and protein to lipid ratio were greater in domesticated species. Cucurbit pollen specialists, Eucera spp., had the highest probability of visit for all Cucurbita taxa.

Conclusions

We provide evidence that floral traits of domesticated and wild Cucurbita species experienced different selection pressures. Domesticated Cucurbita species may have more resources invested towards floral traits, thereby increasing attractiveness to pollinators and potentially plant reproductive success. Wild ancestor plant populations should be conserved in their centers of origin to preserve plant-pollinator interactions.  相似文献

选用西葫芦(Cucurbita pepo)品种“阿兰”一代为试验材料,研究了外源Ca2+处理对高温强光交叉胁迫下西葫芦幼苗生长特征、光合特性及叶绿素荧光参数的影响.结果表明:高温强光胁迫下,5~20 mmol·L-1 Ca2+处理的西葫芦幼苗具有较高的株高和较大的叶面积,其叶绿素、类胡萝卜素含量及光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)、PSⅡ最大光化学效率(Fv/Fm)、PSⅡ实际光化学效率(ΦPSⅡ)和光化学猝灭系数(qP)均较高,而胞间CO2浓度(Ci)和非光化学猝灭系数(NPQ)较低,其中以10 mmol·L-1Ca2+处理效果最好.说明5~20 mmol·L-1Ca2+处理能有效缓解高温强光对西葫芦光合机构的不可逆伤害,使其保持较快的光合电子传递速率和较高的PSⅡ电子传递活性.Ca2+处理浓度超过40 mmol·L-1时对高温强光胁迫没有缓解效应.  相似文献

Enzymes catalyzing the deamidation of seed storage proteins were found in germinating seeds of kidney beans ( Phaseolus vulgaris L. cv. Moldavian) and squash ( Cucurbita pepo L. cv. Mozoleevskaya). They were partially purified and characterized. The properties of these enzymes closely resemble those of the protein deamidase in germinating wheat grains. The detection of similar protein deamidases in germinating seeds of plants belonging to phytogenetically unrelated species indicates that protein deamidases are of considerable physiological significance.  相似文献

Thirty-six clones were recovered fromCucurbita maxima genomic DNA which had been enriched for rDNA and cleaved at the unique repeat unitHind III site. Twenty-nine of these, which contain complete rDNA units, were compared to a standard whose intergenic spacer (IGS) nucleotide sequence has been determined. Twenty-one are identical in length and restriction site pattern. Eight which differ from the standard in length do so because of addition or deletion of varying numbers of IGS subrepetitive units of two different classes, with four of the length variants being different in both of these classes. Seven clones were isolated which contain incomplete repeat units, six of which are composites of rDNA and non-rDNA material. They have been cleaved at the unique rDNAHind III site at one end and at a non-rDNAHind III site at the other. We consider it most likely that these are derived from the termini of repeat unit tandem arrays, although other explanations are possible. Twelve individual plants of two different cultivars were examined for heterogeneity of IGS length distribution. They all appear to be identical in this regard.  相似文献

Cucurbita maxima (pumpkin) is a typical cucurbit that is susceptible to gummy stem blight. In recent years, this major fungal disease has decimated pumpkin yields in Northeast China (Heilongjiang Province) in increasingly numerous outbreaks with more rapid spread in recent years. After culturing fungi on potato dextrose agar medium, we conducted a systematic study of the growth and morphological characteristics of various purified strains of Cucurbita gummy stem blight (GSB) fungus from across Northeast China. Subsequently, DNA samples of 30 isolates with distinct hyphal variations were subjected to internal transcribed spacer (ITS) sequencing. Sequence analysis resulted in identification of the isolates as Stagonosporopsis cucurbitacearum and demonstrated that this is a dominant and widely distributed fungal species in this region. Subsequently, multi‐site phylogenetic analysis assigned the 30 aforementioned strains to two genotypes that aligned to seven phenotypic types of Cucurbita GSB fungi. By analysing ITS conserved sequences of these phenotypically diverse groups, we found that Cucurbita GSB pathogens broadly shared two motifs that contained sequence variations unique to two groups in addition to common identical motifs. Ultimately, this study provided useful data for rapid and accurate identification of S. cucurbitacearum and diagnosis in early symptoms of Cucurbita GSB. This work also provides tools to explore the distribution and regularity of GSB outbreaks spatially and temporally across Northeast China.  相似文献

Basipetal auxin transport along the zucchini hypocotyl requires the cell-to-cell translocation of this hormone. Several authors have demonstrated that naphthylphthalamic acid (NPA) blocks auxin efflux. It is believed that NPA binds to a regulatory protein that modulates the activity of the carrier rather than to the carrier itself. A few regulatory mechanisms have been suggested but all lack strong supporting experimental evidence. Four classes of protein tyrosine kinase inhibitors (PTKIs) have been used to investigate the NPA perception pathway. First, six synthetic tyrphostins, inhibitors of the epidermal growth factor receptor (EGF-R), were shown to displace NPA on plasmalemma fractions. The most potent compound was (3,4-dihydroxybenzylidene)-thiocyanoacetamide (tyrphostin A47). The inactive counterpart of tyrphostin A47, (4-methoxybenzylidene)-malononitrile (tyrphostin A1), was unable to displace NPA. Tyrphostins did not inhibit auxin efflux but were antagonists of the inhibition of auxin efflux by NPA. Again, tyrphostin A47 was the most effective and tyrphostin A1 was inactive. Second, the flavonoid genistein, also an EGF-R inhibitor, showed an effect on NPA binding and NPA antagonism similar to the one for the tyrphostins. Daidzein, the inactive counterpart of genistein, was ineffective in displacing NPA and in antagonizing its inhibition of auxin efflux. Two other PTKIs, curcumin and lavendustin A, displayed similar characteristics. Calmodulin antagonists, protein serine/threonine kinase inhibitors, and phosphatase inhibitors were inactive. These results suggest that the NPA binding protein may be related to mammalian tyrosine kinases and may regulate the auxin efflux carrier by phosphorylation.  相似文献

Auxin and elicitors reportedly activate phospolipase A. A number of inhibitors known to inhibit animal phospholipase A2 were tested for their ability to inhibit hormone and fusicoccin-induced growth. To this end, growth induced by indolyl-3-acetic acid and 2,4-dichlorophenoxyacetic acid in hypocotyl segments of etiolated zucchini (Cucurbita pepo L.) seedlings was determined in the presence of the inhibitors nordihydroguajaretic acid (NDGA), aristolochic acid, 5,8,11,14-eicosatetraynoic acid (ETYA), PBx (a prostaglandin derivative), and oleylethyl phosphocholine. Each chemical proved inhibitory to auxin-induced growth, oleylethyl phosphocholine being the least effective. The effects of the first three inhibitors were investigated in more detail. Growth induced by 10 μM 2,4-dichlorophenoxyacetic acid or 1 μM indolyl-3-acetic acid was inhibited 50% by about 30–50 μM NDGA, by about 25 μM aristolochic acid, and by about 10–20 μM EYTA. Growth inhibition was reversible and became apparent 0.5–1 h after inhibitor addition. Growth induced by 0.5 or 1 μM fusicoccin was much less inhibited by NDGA and by ETYA, whereas aristolochic acid was only slightly less effective on fusicoccin-induced than on auxin-induced growth. These three inhibitors were also tested for their effects on gibberellin-induced growth in light-grown peas (Pisum sativum L.) and on cytokinin-induced expansion growth in excised cotyledons from radish (Raphanus sativum L.) seedlings. In both tests, aristolochic acid had toxic side-effects although gibberellin-induced growth was still apparent. In the gibberellin test, neither NDGA at up to 100 μM nor ETYA at 80 μM was inhibitory to hormone-induced growth. Moreover, 40 μM ETYA was not inhibitory to kinetin-induced growth. We hypothesize that the selectivity of phospholipase A2 inhibitors for auxin-induced growth implies a different signal transduction pathway for each of the different signal substances tested, and that auxins might use fatty acid(s) and/or lysophospholipid(s) or their derivatives as the preferred second messengers. Rinsing roots of Cucurbita pepo with an essentially boron-free nutrient solution results in a significant reduction of the cell wall elasticity modulus as determined by the cell presure probe (measuring always in the same cells). This points to a role of boron in crosslinking cell wall macromolecules. Longer exposure to boron deficient conditions, however, increased again and above the initial values. This trend became visible between 20 and 30 minutes after starting the deficiency treatment. There is no indication for a loss of turgor during the time course of the experiments, suggesting that boron primarily affects cell wall physical properties and that membrane leakiness, as observed by other groups, is a secondary event. Several testable hypotheses for the action of boron are presented.  相似文献
