The photosynthetic characteristics of coffee ( Coffea arabusta) plantlets cultured in vitro in response to different CO2 concentrations inside the culture vessel and photosynthetic photon flux (PPF) were investigated preliminarily. The estimation of net photosynthetic rate (Pn) of coffee plantlets involved three methods: (1) estimating time courses of actual Pn in situ based on measuring CO2 concentrations inside and outside the vessel during a 45-day period, (2) estimating Pn in situ at different CO2 concentrations and PPFs using the above measuring approach for 10-day and 30-day old in vitro plantlets, and (3) estimating Pn of a single leaf at different CO2 concentrations and PPFs by using a portable photosynthesis measurement system for 45-day old in vitro coffee plantlets. The results showed that coffee plantlets in vitro had relatively high photosynthetic ability and that the Pn increased with the increase in CO2 concentration inside the vessel. The CO2 saturation point of in vitro coffee plantlets was high (4500–5000 μmol mol-1); on the other hand, the PPF saturation point was not so high as compared to some other species, though it increased with increasing CO2 concentration inside the vessel. Molecular mobility in membranes of intracellular organelles is poorly understood, due to the lack of experimental tools applicable for a great diversity of shapes and sizes such organelles can acquire. Determinations of diffusion within the plasma membrane or cytosol are based mostly on the assumption of an infinite flat space, not valid for curved membranes of smaller organelles. Here we extend the application of FRAP to mitochondria of living cells by application of numerical analysis to data collected from a small region inside a single organelle. The spatiotemporal pattern of light pulses generated by the laser scanning microscope during the measurement is reconstructed in silico and consequently the values of diffusion parameters best suited to the particular organelle are found. The mobility of the outer membrane proteins hFis and Tom7, as well as oxidative phosphorylation complexes COX and F1F0 ATPase located in the inner membrane is analyzed in detail. Several alternative models of diffusivity applied to these proteins provide insight into the mechanisms determining the rate of motion in each of the membranes. Tom7 and hFis move along the mitochondrial axis in the outer membrane with similar diffusion coefficients (D = 0.7 μm2/s and 0.6 μm2/s respectively) and equal immobile fraction (7%). The notably slower motion of the inner membrane proteins is best represented by a dual-component model with approximately equal partitioning of the fractions (F1F0 ATPase: 0.4 μm2/s and 0.0005 μm2/s; COX: 0.3 μm2/s and 0.007 μm2/s). The mobility patterns specific for the membranes of this organelle are unambiguously distinguishable from those of the plasma membrane or artificial lipid environments: The parameters of mitochondrial proteins indicate a distinct set of factors responsible for their diffusion characteristics.  相似文献

关帝山华北落叶松、云杉、杨桦天然次生林植被类型是华北高海拔地带代表性的森林类型。以该地区上述3种典型的天然次生林群落为例,运用点格局分析中的单变量O-ring统计方法,分析各群落主要树种的种群空间分布格局;用双变量O-ring统计方法,分析各群落中树种间的空间关联性;并对同一树种在不同森林群落类型中种群空间分布格局和种间关联性的变化进行比较,以探讨华北山地次生林群落空间格局形成和种群维持与动态机制。研究结果表明,(1)各种群在相对小的尺度上聚集分布特征明显,随尺度增加树木种群的聚集性减弱并逐渐表现出随机分布的格局特征;(2)同一树种在不同森林群落类型中空间分布格局差异很大。红桦、白桦和山杨在阔叶林中呈明显的小尺度聚集分布格局,而在针叶林群落呈随机分布。在阔叶次生林中混生的小龄级华北落叶松呈现聚集性空间分布格局。在华北落叶松为优势树种的针叶林中,大龄级的华北落叶松表现出小尺度上均匀分布和较大尺度上随机分布的格局特征;(3)各群落中树种间的空间关联性主要表现在小尺度范围,随尺度加大空间关联性逐渐不明显;(4)在阔叶树占优势的次生林,小龄级华北落叶松与桦木的分布有较明显的空间正关联性,而在华北落叶松为优势树种的针叶林,桦与华北落叶松并未表现明显的空间关联性。种群分布格局和种群间的空间关联性随群落结构、空间尺度和龄级结构不同而有较大变化。  相似文献

粗木质残体(Coarse woody debris,CWD)的空间格局反映了森林群落的死亡格局和干扰格局,在一定程度上体现了群落内林木的死亡过程。采用相邻网格法对色季拉山急尖长苞冷杉(Abies georgei var.smithii)原始林1 hm2固定样地内CWD进行调查,从CWD类型、腐烂等级、径级3个方面对CWD空间分布格局进行分析。结果表明:样地内CWD总密度为582株/hm2,倒木占55.33%,是CWD的主要输入形式。CWD密度在腐烂等级上的分布可用多项式拟合(R2=0.9973),在径级上的分布可用指数衰减模型拟合(R2=0.9746),且在不同类型、腐烂等级及径级上的分布差异较大。在50 m尺度内,CWD整体表现为小尺度的集群分布和中、大尺度的随机分布。在3种CWD分类中,仅有大枯枝、Ⅰ级腐烂、径级ⅠCWD在小尺度或中尺度表现为较强的集群分布,其余则均以随机分布为主,只是在个别尺度达到或接近集群分布。不同类型CWD间整体关联不显著,只有枯立木与大枯枝在0-21 m尺度内达到显著负关联。CWD空间分布格局是急尖长苞冷杉原始林的重要结构特征,在很大程度上决定着林下植物群落及林型自然更新格局。  相似文献

Larix principis-rupprechtii forest is an important vegetation formation and has a large distribution area in Luya Mountain Reserve,China.Spatial pattern analysis on individual trees in different age-classes of Larix principisrupprechtii was made in this paper.Here,we employed the technique of point pattern analysis,which could analyze patterns under all scales along a gradient.It was based on spatial mapped points of individual distribution.The results of this study showed that the densities of the five age-classes varied in the order:age-class 3>age-class 4>age-class 5>age-class 2>age-class 1.Although age-classes 1 and 2 have much fewer individuals than other three age-classes do,the population was stable at present.However,it would be necessary to take some measures for improving population regeneration for a long-time view.The individuals of all age-classes focused on clumping distribution in space;however,their distribution pattern varied with the change of scale.This mainly depended on biological features of Larix principis-rupprechtii and forest environments,but it also meant that the scale was an important factor in controlling spatial distribution pattern of tree individuals.The feature of clumping distribution became more significant with the increase of age.The relationships between individuals in different age-classes were almost all significantly correlated with each other.These associations became more significant within the older age-classes.This suggested that the individuals of different age-classes were interdistributed,by which the population could get benefits in resource utilization.The technique of point pattern analysis is effective and easy to be used in species pattern study.Its results are more closer to the reality,especially for community structure.  相似文献

Expansins are a family of proteins with plant cell wall remodeling‐activity, which bind cell wall components through hydrophobic and electrostatic interactions. A shallow area on the surface of the protein serves as the polysaccharide binding site (PBS) and it is composed of conserved residues. However, electric charge differences on the opposite face of the PBS produce basic, neutral, or acidic proteins. An analysis of forty‐four bacterial expansins, homologues of BsEXLX1, revealed two main groups defined by: (a) the presence or absence of disulfide bonds; and (b) by the proteins isoelectric point (pI). We determined the location of the residues responsible for the pI on the structure of representative expansins. Our results suggest that the electric charge at the opposite site of the PBS may help in substrate differentiation among expansins from different species; in addition, electrostatic polarization between the front and the back of the molecule could affect expansin activity on cellulose. 长江经济带是重大国家战略发展区域,优化其国土空间开发保护格局是实现生态文明建设的重要内容。研究基于“核心点-发展轴-战略区”的框架,通过国土空间利用强度表征指标及夜间灯光数据识别发展核心点,借助有序加权平均算法模拟多种决策愿景下的空间发展阻力面,运用最小累积阻力模型和电路理论模型识别空间发展轴线及其内部的开发保护战略区,定量构建不同决策偏好的空间格局优化方案,突破了传统研究中单一目标和定性分析的局限。结果表明:长江经济带国土空间发展核心点共63个,约占区域总面积的5%,呈现自东向西递减的空间分布特征;城镇扩张、协同发展和生态约束三类情景下发展轴线均为61条,所覆盖的县级单元分别占总数的36.6%、36.5和36.7%,呈现出“一横三纵”的格局特征;城镇扩张、协同发展和生态约束三类情景下开发战略区分别占轴线面积的4.5%、4.9%、4.8%,多分布于长江干流及其支流沿线。保护战略区分别占轴线面积的17.1%、13.9%、15.3%,主要分布于长江南岸区域。在此基础上形成的国土空间开发保护格局基础架构,能够为落实长江经济带“共抓大保护、不搞大开发”提供决策参考。  相似文献
