Supramolecular organization involving a polyanionic glycan in a bacterial capsule (hyaluronan, HA, in streptococcal capsules) is revealed, for the first time, by electron-histochemical methodology previously used to elucidate ultrastructure in extracellular matrix. Capsular HA filaments thereby revealed closely resemble aligned linear structures demonstrated by similar technology in HA solution. These parallel arrays, spontaneously formed, are based on HA tertiary structures (beta sheet-like) which are stabilized by hydrophobic and hydrogen bonds. HA tertiary structures in aqueous solutions resist shear stress as shown by rheo-NMR. Thus, supramolecular HA wrapping covering many cells probably stabilizes chains of bacteria. Streptococci possibly templated the ordered structures since eukaryotic B6 cell HA did not produce similarly organized envelopes. Supramolecular organization in streptococcal and pneumococcal capsules are compared. Their glycans are very similar but the potential for HA-like tertiary structures is not present in the pneumococcal type 3 polysaccharide and chains of cells are not formed to the same extent by pneumococci. We suggest that the streptococcal capsule exemplifies a simple extracellular matrix analogous to those in animal connective tissues, which contain glycans (chondroitin, keratan, and dermochondan sulfates) of the HA family, capable of undergoing aggregation to similar tertiary structures.  相似文献

It is still unclear whether the paradoxical arteriovenous carboxyhemoglobin (COHb) difference found in critical illness is due to increased COHb production by the lung, or whether this gradient is caused by technical artifacts using spectrophotometry. In healthy and matched endotoxemic sheep, blood gases were analyzed with a standard ABL 625 and the updated version, an ABL 725. The latter one was accurately calibrated for COHb wavelengths (SAT 100) to eliminate the FCOHb dependency on oxygen tension. All endotoxemic sheep exhibited a hypotensive-hyperdynamic circulation and a pulmonary hypertension. Interestingly, arteriovenous COHb difference occurred in both healthy and endotoxemic sheep (P<0.001 each). Arterial and central venous COHb concentrations determined with the ABL 625 were significantly lower than those measured with the ABL 725 (P<0.001 each). We conclude that (a) arteriovenous COHb difference per se does not reflect critical illness and (b) measurements with an ABL 625 underestimate COHb concentrations.  相似文献

Effects of Mg2+ ions on thermally induced conformational transitions in the synthetic poly(dA)·poly(dT) and poly(dA)·2poly(dT) were studied in the buffered solutions (pH 6.9), containing 0.1 or 1 M NaCl at polynucleotide concentration of 0.1–0.3 mM (in nucleic bases). The experiments consist of measurements of the UV absorption and intensity of conventional visible static light scattering. The diagram of conformational transitions in the poly(dA)–poly(dT)–Mg2+ system was constructed on a basis of experimental data obtained. Anomalously strong light scattering, like critical opalescence, has been revealed at 0.1 M NaCl and [Mg2+]≥20 mM in the melting range of both polynucleotides, which eventually disappeared after the completion of polymer strands separation. The effect presumably is caused by a fluctuation process of polymer strands complexing which arises at a certain concentration of Mg2+ ions.  相似文献

The water content, the survival time at various relative humidities(r.h.) and the critical equilibrium activity of unfed adultDermacentor marginatus and D.reticulatus ticks were investigated at a constant temperature of20 °C. It was also examined whether these ticks use liquidwaterto compensate water loss. Both Dermacentor spp. showed nosignificant differences in water content in relation to body mass. The meanwater content of D. marginatus and D.reticulatus was 54.6% and 54.7%, respectively, in females and 56.3%and 57.0%, respectively, in males. The survival time of unfed adults prolongedwith decreasing saturation deficits. On average, males survived longer thanfemales and D. marginatus ticks survived mostly longerthanD. reticulatus ticks. The 50% mortality period rangedbetween 40 d at 33% r.h. and 420 d at 95% r.h. in D.marginatus, and between 43 d at 33 r.h. and 366 d at 95% r.h. inD. reticulatus. The critical equilibrium activity of unfedadults was estimated to be 0.84 for both species and was independent of sex.When dehydrated adult D. marginatus and D.reticulatus ticks were offered liquid water, only a few slightlygained weight while most further lost weight. Liquid water was not attractivefor dehydrated or non-dehydrated ticks and drinking was not observed. Aftersubmerging in water for 2 d, most of the dehydrated ticks had gained weight.  相似文献

When exposed to severely hypoxic water, many teleosts skim the better oxygenated surface layer (aquatic surface respiration, ASR). Information is scarce concerning the thresholds triggering ASR and its cardio-respiratory consequences. To assess the ambient conditions leading to ASR and to evaluate its effects on cardio-respiratory function, we exposed specimens of Piaractus mesopotamicus to gradual hypoxia (water oxygen tension ranging from 120 to 10 torr) with or, alternatively, without access to the surface. Concurrently, ASR, cardiac and respiratory frequencies, O2 uptake and gill ventilation were monitored. With surface access, ASR developed below the critical tension for O2 uptake (34 torr) by normal gill ventilation. Moreover, the time spent in ASR increased with prolonged hypoxic exposure to a maximum of 95% of total time. Without surface access, the species exhibited hypoxic bradycardia, that had not occurred in the group with fully developed ASR. Even without ASR, P. mesopotamicus recovered readily from hypoxic exposure, showing that this species possesses a number of mechanisms to cope with environmental hypoxia.  相似文献

Regime shifts are abrupt transitions between alternate ecosystem states including desertification in arid regions due to drought or overgrazing. Regime shifts may be preceded by statistical anomalies such as increased autocorrelation, indicating declining resilience and warning of an impending shift. Tests for conditional heteroskedasticity, a type of clustered variance, have proven powerful leading indicators for regime shifts in time series data, but an analogous indicator for spatial data has not been evaluated. A spatial analog for conditional heteroskedasticity might be especially useful in arid environments where spatial interactions are critical in structuring ecosystem pattern and process. We tested the efficacy of a test for spatial heteroskedasticity as a leading indicator of regime shifts with simulated data from spatially extended vegetation models with regular and scale‐free patterning. These models simulate shifts from extensive vegetative cover to bare, desert‐like conditions. The magnitude of spatial heteroskedasticity increased consistently as the modeled systems approached a regime shift from vegetated to desert state. Relative spatial autocorrelation, spatial heteroskedasticity increased earlier and more consistently. We conclude that tests for spatial heteroskedasticity can contribute to the growing toolbox of early warning indicators for regime shifts analyzed with spatially explicit data.  相似文献

This paper is a survey paper on stochastic epidemic models. A simple stochastic epidemic model is defined and exact and asymptotic (relying on a large community) properties are presented. The purpose of modelling is illustrated by studying effects of vaccination and also in terms of inference procedures for important parameters, such as the basic reproduction number and the critical vaccination coverage. Several generalizations towards realism, e.g. multitype and household epidemic models, are also presented, as is a model for endemic diseases.  相似文献

We determine the critical patch size below which extinction occurs for populations living in one-dimensional habitats surrounded by completely hostile environments in the presence of environmental fluctuations. The population dynamics is reformulated in terms of a stochastic reaction–diffusion equation and is reduced to a deterministic equation that incorporates the systematic contributions of the noise. We obtain bifurcation diagrams and relations for the mean population density at the stationary state, the critical patch size, and the mean number of individuals in the habitat. The effect of the noise differs, depending on whether it affects the net growth rate or the intraspecific competition term. Fluctuations in the net growth rate decrease the critical patch size, whereas fluctuations in the competition term do not change the critical patch size. We compare our analytical results with numerical solutions of the stochastic partial differential equations and show that our procedure proves useful in dealing with reaction–diffusion equations with multiplicative noise.  相似文献

A series of quaternary ammonium compounds (QUATS) derived from l-Phenylalanine have been synthesized and their antibacterial efficiencies were determined against various strains of Gram-positive and Gram-negative bacteria. The antibacterial activity increased with increasing chain length, exhibiting a cut-off effect at C14 for Gram-positive and C12 for Gram-negative bacteria. The l-Phenylalanine QUATS displayed enhanced antibacterial properties with a higher cut-off point compared to their corresponding l-Phenylalanine ester hydrochlorides. The CMC was correlated with the MIC, inferring that micellar activity contributes to the cut-off effect in antibacterial activity. The hemolytic activities (HC50) of the QUATS against human red blood cells were also determined to illustrate the selectivity of these QUATS for bacterial over mammalian cells. In general, the MIC was lower than the HC50, and assessment of the micellar contribution to the antibacterial and hemolytic evaluation in TBS as a common medium confirmed that these QUATS can act as antibacterial, yet non-toxic molecules at their monomer concentrations. The interaction of the QUATS with the phospholipid vesicles (1,2-dipalmitoyl-sn-glycero-3-phosphocholine, DPPC) in the presence of 1-anilino-8-naphthalene sulfonate (ANS) and 1,6-diphenyl-1,3,5-hexatriene (DPH) as fluorescence probes showed that the presence of the quaternary ammonium moiety causes an increase in hydrophobic interactions, thus causing an increase in antibacterial activity.  相似文献

Coping with cold winter conditions is a major challenge for many insects.In early spring we observed newly emerged Drosophila subobscura, which had overwintered as larvae and pupae. As temperatures increase during spring these flies are faced with higher minimum and maximum temperatures in their natural microhabitat. Thus, there is a potential costly mismatch between winter and early spring acclimatization and the increased ambient temperatures later in adult life.We obtained individuals from a natural Danish population of D. subobscura and acclimated them in the laboratory to 20 °C for one generation, and compared critical thermal maximum (CTmax) and minimum (CTmin) to that of individuals collected directly from their natural microhabitat. The two populations (laboratory and field) were subsequently both held in the laboratory at 20 °C and tested for their CTmax and CTmin every third day for 28 days.At the first day of testing, field acclimatized D. subobscura had both higher heat and cold resistance compared to laboratory flies, and thereby a considerable larger thermal scope. Following transfer to the laboratory, cold and heat resistance of the field flies decreased over time relative to the laboratory flies. Despite the substantial decrease in thermal tolerances the thermal scope remained larger for field acclimatized individuals for the duration of the experiment.We conclude that flies acclimatized to their natural microhabitat had increased cold resistance, without a loss in heat tolerance. Thus while a negative correlation between cold and heat tolerance is typically observed in laboratory studies in Drosophila sp., this was not observed for field acclimatized D. subobscura in this study. We suggest that this is an adaptation to juvenile overwintering in temperate cold environments, where developmental (winter) temperatures can be much lower than temperatures experienced by reproducing adults after emergence (spring). The ability to gain cold tolerance through acclimatization without a parallel loss of heat tolerance affects thermal scope and suggests that high and low thermal tolerance act through mechanisms with different dynamics and reversibility.  相似文献
