SYNOPSIS. Chloramphenicol cured Crithidia deanei of its endosymbiote. The derived aposymbiotic strain had additional growth requirements: purin (as adenine), heme, arginine, histidine, isoleucine, leucine, phenylalanine, threonine, tryptophan, valine, pyridoxine, riboflavin, and pantothenate and liver infusion (replaceable by high nicotinamide).  相似文献

Reversal of the growth inhibition of Crithidia fasciculata by allopurinol requires both a purine and a pyrimidine. Hypoxanthine is the most effective purine in the reversal. Cell-free extracts were prepared which were capable of the decarboxylation of orotidine 5′-phosphate. Other enzyme preparations carried out the phosphoribosylation of allopurinol. By the use of [4-14C] orotidine 5′-phosphate (enzymatically prepared), it was shown that allopurinol ribotide (enzymatically prepared), but not the free base, inhibits orotidine 5′-phosphate decarboxylase.  相似文献

To further investigate the phylogeny of protozoa from the order Kinetoplastida we have sequenced the small subunit (SSU) and a portion of the large subunit (LSU) nuclear rRNA genes. The SSU and LSU sequences were determined from a lizard trypanosome, Trypanosoma scelopori and a bodonid, Rhynchobodo sp., and the LSU sequences were determined from an insect trypanosomatid, Crithidia oncopelti, and a bodonid, Dimastigella trypaniformis. Contrary to previous results, in which trypanosomes were found to be paraphyletic, with Trypanosoma brucei representing the earliest-diverging lineage, we have now found evidence for the monophyly of trypanosomes. Addition of new taxa which subdivide long branches (such as that of T. brucei) have helped to identify homoplasies responsible for the paraphyletic trees in previous studies. Although the monophyly of the trypanosome clade is supported in the bootstrap analyses for maximum likelihood at 97% and maximum parsimony at 92%, there is only a small difference in ln-likelihood value or tree length between the most optimal monophyletic tree and the best suboptimal paraphyletic tree. Within the trypanosomatid subtree, the clade of trypanosomes is a sister group to the monophyletic clade of the nontrypanosome genera. Different groups of trypanosomes group on the tree according to their mode of transmission. This suggests that the adaptation to invertebrate vectors plays a more important role in the trypanosome evolution than the adaptation to vertebrate hosts. 【目的】近年来,熊蜂作为温室作物的理想授粉者在国外已被广泛利用,并且获得很好的经济效益和生态效益,所以国外熊蜂经常被进口用于设施农业。熊蜂短膜虫Crithidia bombi是熊蜂的一种重要寄生虫病,一旦随进口熊蜂传入,将给国内熊蜂蜂群带来严重危害,因此迫切需要建立一种熊蜂短膜虫检测方法。【方法】基于熊蜂短膜虫基因内转录间隔区(internal transcribed space,ITS)基因序列设计了一对引物(Cri-F/R),建立了熊蜂短膜虫的PCR检测方法,并对退火温度、引物浓度和循环个数等反应条件进行了优化,同时验证了该PCR方法的灵敏性、特异性和稳定性。【结果】以熊蜂短膜虫ITS基因保守区设计特异性引物建立的熊蜂短膜虫PCR检测方法是可行的。优化的PCR反应条件为:退火温度59℃,引物浓度0.5μmol/L,扩增循环数35次。对感染熊蜂短膜虫的熊蜂总DNA的灵敏度达到13.24×10-5ng/μL,并具有良好的特异性和稳定性。将该方法应用于熊蜂短膜虫的检测,整个检测过程不超过4 h,具有良好的适用性。【结论】研究建立了熊蜂短膜虫检测方法,能用于疫情监测和进境熊蜂的检验检疫。  相似文献

Floral landscapes comprise diverse phytochemical combinations. Individual phytochemicals in floral nectar and pollen can reduce infection in bees and directly inhibit trypanosome parasites. However, gut parasites of generalist pollinators, which consume nectar and pollen from many plant species, are exposed to phytochemical combinations. Interactions between phytochemicals could augment or decrease effects of single compounds on parasites. Using a matrix of 36 phytochemical treatment combinations, we assessed the combined effects of two floral phytochemicals, eugenol and thymol, against four strains of the bumblebee gut trypanosome Crithidia bombi. Eugenol and thymol had synergistic effects against C. bombi growth across seven independent experiments, showing that the phytochemical combination can disproportionately inhibit parasites. The strength of synergistic effects varied across strains and experiments. Thus, the antiparasitic effects of individual compounds will depend on both the presence of other phytochemicals and parasite strain identity. The presence of synergistic phytochemical combinations could augment the antiparasitic activity of individual compounds for pollinators in diverse floral landscapes.  相似文献

Abstract Incorporation of [1-14 C]acetate by the trypanosomid Crithidia fasciculata showed that ergosterol biosynthesis occurs de novo in this protozoon, via lanosterol and 31-norlanosterol. No cycloartenol could be detected, indicating that this biosynthesis pathway is rather similar to those of other non-photosynthetic organisms (animals, fungi). From the point of view of sterol biosynthesis, C. fasciculata is not related to other ergosterol-synthesising protozoa, such as the hitherto examined phytoflagellates and soil amoebae, which synthesise their sterols via cycloartenol, like photosynthetic organisms (plants, algae).  相似文献

1. Social insect castes and sexes differ in many ways, including morphology, behavior, and sometimes ploidy level. Recent studies have found that consuming sunflower pollen reduces the gut pathogen Crithidia bombi in workers of the common eastern bumble bee (Bombus impatiens). Here, this work is extended to the reproductive individuals that represent colony fitness – males and queens – to assess if the medicinal effects of sunflower pollen vary with bee caste and sex. 2. This study examined the effect of sunflower pollen compared to a diverse wildflower pollen mix on infection in worker, male, and daughter queen commercial B. impatiens. Bees were infected, fed either sunflower pollen or wildflower pollen for 7 days, and then infection levels were assessed. 3. Compared to wildflower pollen, sunflower pollen dramatically reduced Crithidia infection in workers and daughter queens, but not males. Infection levels were very low for both diets in males; this could be due to low pollen consumption or other mechanisms. 4. Reducing Crithidia infection in young queens before they undergo hibernation is important for population dynamics since infected queens are less likely to survive hibernation, and those that do are less likely to successfully establish a nest the following spring. Because sunflowers bloom in late summer when new queens are emerging, sunflowers could provide an important dietary component for queens during this critical life stage. Deepening our understanding of how diet impacts pathogens in reproductive bees, as well as workers, is crucial to maintain healthy pollinator populations.  相似文献

Summary As in other trypanosomatids, the cell membrane of Crithidia fasciculata overlies a single layer of microtubules. Each microtubule possesses a large number of periodically arranged drumstick-like appendages and adjacent microtubules are joined by fibrillar connectives. Anteriorly, the microtubules gradually taper to terminate just before or just after entering the reservoir. An attempt is made to correlate microtubule tapering with maintenance of form of the truncated anterior end of the cell. Smooth and coated vesicles are proliferated from the Golgi saccules and the prominent contractile vacuole lies nearby. The single mitochondrion is extensive and expanded at one point to form a capsule for the kinetoplast. The cristae are predominantly plate-like but other configurations do occur. The cytostome, a shallow invagination of the reservoir membrane, is found between two constrictions in the reservoir wall. Supporting the cytostome are several microtubules which penetrate deeply into the cytoplasm. Ingestion of ferritin occurs by pinocytosis from the cytostome and by coated vesicle formation from the reservoir membrane. Digestion probably occurs in multivesicular bodies which contain acid phosphatase activity.  相似文献

Species interaction networks, which play an important role in determining pathogen transmission and spread in ecological communities, can shift in response to agricultural landscape simplification. However, we know surprisingly little about how landscape simplification‐driven changes in network structure impact epidemiological patterns. Here, we combine mathematical modelling and data from eleven bipartite plant‐pollinator networks observed along a landscape simplification gradient to elucidate how changes in network structure shape disease dynamics. Our empirical data show that landscape simplification reduces pathogen prevalence in bee communities via increased diet breadth of the dominant species. Furthermore, our empirical data and theoretical model indicate that increased connectance reduces the likelihood of a disease outbreak and decreases variance in prevalence among bee species in the community, resulting in a dilution effect. Because infectious diseases are implicated in pollinator declines worldwide, a better understanding of how land use change impacts species interactions is therefore critical for conserving pollinator health.  相似文献

Trypanosomatid diversity in Heteroptera was sampled using a culture-independent approach based on amplification and sequencing of Spliced Leader RNA gene repeats from environmental samples. By combining the data collected herein with that of previous work, the prevalence of parasites was found to be 22%-23%. Out of approximately 170 host species investigated nearly 60 were found to harbor trypanosomatids. The parasites found were grouped by cluster analysis into 48 typing units. Most of these were well separated from the known groups and, therefore, likely represent new trypanosomatid species. The sequences for each typing unit serve as barcodes to facilitate their recognition in the future. As the sampled host species represent a minor fraction of potential hosts, the entire trypanosomatid diversity is far greater than described thus far. Investigations of trypanosomatid diversity, host-specificity, and biogeography have become feasible using the approach described herein.  相似文献
