Low temperature spectrophotometry was used to study the phototransformation of Pfr to Pr in 1000–7000 g pelletable fractions extracted from dark grown pea ( Pisum sativum L. cv. Alaska) epicotyls which had been irradiated with red and then far-red light. At -170°C, far-red irradiation of the pelletable phytochrome which had been pre-irradiated with saturating fluence of red light before freezing caused formation of an intermediate (named I660 ), the difference spectrum of which showed a marked ab-sorbance decrease at 740 nm and a concomitant small increase at about 660 nm. The inermediate I660 was converted to another intermediate (I660 ) when it was warmed above -80°C. The difference spectrum of this intermediate showed a positive peak at 670 nm. This intermediate was photoconverted to Pfr by red irradiation and also underwent dark reversion to Pfr at -60°C. I660 formed Pr if the temperature was above -10°C. The basic features of the phytochrome intermediates resemble those obtained in vivo and in degraded purified phytochrome. 相似文献

Summary The survival of Azolla was studied in an artificial system which simulated the soil/water interface and the desiccation of
soil during a fallow period in lowland rice culture. Tests with non-sporulating and sporulating Azolla fronds showed that
Azolla only survives with sporulated fronds. At their reappearance the Azolla fronds already harboured the Anabaena endophyte.
A detailed light microscopic and transmission electron microscopic study of macro- and micros-porocarp formation and development
revealed that the endophyte is transmitted by the macrosporocarps and not by the microsporocarps. The Anabaena cells within
the macrosporocarps are found just below the indusium cap. These cells are not nitrogen-fixing akinetes. The free-living Anabaena
cells at the stem apex and below the overarching developing leaves do not bear heterocysts and accordingly are non nitrogen-fixing.
During the development of the leaf the Anabaena enters the leaf cavity, but later the pore of this, cavity closes and the
imprisoned cyanobacteria are lysed before the leaf decays. As the Azolla leaves age a nitrogen-fixing capability is successively
built up concomittantly with the production of heterocysts. Heterocyst frequencies of 40–50% can be found inAnabaena azollae. Usually a gradient of nitrogen-fixing capacity occurs along the Azolla rhizome with two distinct peaks at leaf number 7/8

Seedlings of 14 species were grown for 14–28 days on nutrient solution with 6 mmol.l−1 NH4 as the sole nitrogen source. Solutions acidity was were kept constant at pH 4.0, 5.0, 6.0 and 7.0 by continuous titration
with diluted KOH. The following species were used: barley, maize, oats sorghum, yellow and white lupin, pea, soybean, carrot,
flax, castor-oil, spinach, sugarbeet and sunflower. Most plant species grew optimally at pH 6.0 with slight reductions at
pH 5.0. Growth of many species was severely inhibited at pH 4.0, but this inhibition was not observed with the legume and
cereal species. Yield depressions at pH 4.0 relative to pH 6.0 were well correlated with the respective relative decreases
of the K concentration in their roots (P<0.002). In the roots of two species (sunflower and flax) total N concentrations were also strongly reduced at pH 4.0. apparently,
the interactions between uptake of K, NH4 and H ions become the prevalent problem at suboptimal pH. At pH 7.0, yields were also considerably decreased, with the exception
of the lupines. At this pH, the roots of the growth inhibited plants were characterized by increased levels of total N and
free NH4. It is thought that the binding capacity of the roots for NH4 is an important factor in preventing NH4/NH3 toxicity at supraoptimal pH. 相似文献

Summary General recursion formulae for the coefficient of inbreeding under a cyclical mating system were derived in which one male and one female are selected from each of the n families per generation (population size N = 2 n). Each male is given the family number of his sire in each generation, while his mate comes from another family, varying systematically in different generations. Males of the r-th family in generations 1, 2, 3,..., t = n–1 within each cycle mate with females from families r+1, r+2, r+3,..., r+t to produce generations 2, 3, 4,..., t+1=1, respectively. The change in heterozygosity shows a cyclical pattern of rises and falls, repeating in cycles of n–1 generations. The rate of inbreeding oscillates between <-3% to >6% in different generations within each cycle, irrespective of the population size. The average rate of inbreeding per generation is approximately 1/[4 N-(Log2N+1)], which is the rate for the maximum avoidance of inbreeding. The mean dimensions of thecis N-methyl peptide unit have been arrived at by analysing the crystal structure data on compounds containing such units. These
dimensions can be used as standard in conformational studies on cyclic peptides. While the bonds meeting at C are almost coplanar,
those meeting at N show a slight pyramidal disposition. A comparison of the dimensions of the normal and N-methylatedcis peptide units show that there are perceptible differences in the parameters connected with N. In addition, the flexibility
of thecis peptide unit has been analysed by studying the distribution of the parameters in different classes of compounds such as cyclic
di, tri and higher peptides. The salient features are: (i) The angle CαCN in cyclic dipeptide and the angle CδNCα in higher peptides tend to be lower, when the peptide unit is associated with a prolyl residue; (ii) in cyclic tripeptides
the internal anglesviz., CαCN and CNCα are significantly larger thereby increasing the intra-annular space; (iii) the bond Cα-C is distinctly shorter when it occurs in cyclic dipeptides. Both Lys-166 and His-291 of ribulosebisphosphate carboxylase/oxygenase fromRhodospirillum rubrum have been implicated as the active-site residue that initiates catalysis. To decide between these two candidates, we resorted
to site-directed mutagenesis to replace Lys-166 and His-291 with several amino acids. All 7 of the position-166 mutants tested
are severely deficient in carboxylase activity, whereas the alanine and serine mutants at position 291 are ∼40% and ∼18% as
active as the native carboxylase, essentially ruling out His-291 in theRhodospirillum rubrum carboxylase (and by inference His-298 in the spinach enzyme) as a catalytically essential residue. The ability of some of
the mutant proteins to undergo carbamate formation or to bind either ribulosebisphosphate or a transition-state analogue remains
largely unimpaired. This implies that Lys-166 is not required for substrate binding; rather, the results corroborate the earlier

Of the three Brachionus species used in aquaculture, Brachionus rubens, B. calycilorus and B. plicatilis, the latter is most widely used in raising marine fish and shrimp larvae due to its tolerance to the marine environment. In freshwater aquaculture the use of B. rubens and B. calycilorus is limited, probably because inert food products are readily available as feed for freshwater larvae.The rotifer Brachionus plicatilis is used in large numbers as the first food organism in intensive cultures of marine fish and shrimp larvae. An adequate supply of these rotifers relies on mass cultures. The reproductive rate of rotifers in these cultures depends on food quality and quantity, salinity, temperature and pH of the medium. Removal of waste products from culture tanks leads to higher and more efficient production of rotifers over extended periods of time. Rotifers have to be enriched with polyunsaturated fatty acids, which are essential for proper development and survival of marine fish and shrimp larvae.The future use of preserved rotifers and their resting eggs may help to overcome unforeseen failures of live cultures and may lead to more efficient use of these organisms in raising freshwater and marine fish and shrimp larvae. 相似文献

Individual size, rate of growth, and mode and frequency of asexual reproduction are life-history traits of primary importance for sea anemones. These traits determine sexual reproductive output, affect an individual's probability of survival, and are crucial in adapting an individual to its environmental surroundings. The sea anemone Metridium senile (L.) is highly variable in ecological distribution and life history, including rate of growth, individual size, and rate of asexual reproduction. Gonad size (measured as cross-sectional area of gonadal tissue) increases with body weight, so individuals should grow as large and as rapidly as possible to maximize individual sexual reproductive output. Cessation of growth and small body size in intertidal populations suggest that growth is constrained by genetic or environmental conditions. The growth of intertidal individuals transplanted to harbor-float panels demonstrated that growth limits are imposed by environmental factors, most probably limited food and feeding time and damage from wave exposure (which stimulates fragmentation). Individuals in harbor-float populations, which are continuously immersed, grow much larger, and large individuals comprise a greater proportion of the population than in the intertidal zone. The highest rate of fragmentation observed was on harbor-float panels. Patterns of growth and asexual reproduction provide adaptive advantages for M. senile. For harborfloat individuals, large individual size increases gamete production and may increase feeding efficiency. For intertidal individuals, asexual reproduction allows growth despite individual size constraints and rapid population growth, with specific advantages resulting from clone formation. 相似文献
