应用固定化里氏木霉糖化玉米秆纤维素的研究   总被引:15,自引:0,他引:15  
采用多孔聚酯材料固定里氏木霉(TrichodermareeseiRutC30)菌丝细胞,将固定化细胞在生长限制条件下重复分批培养,使纤维酶的合成与玉米秆纤维原料的酶解糖化耦合在一个反应器中同时进行。在30℃、初始pH4.8、摇瓶转速150r/min的条件下,连续重复进行12次分批培养试验。每批玉米秆用量为60g/L,培养周期4.5d,共54d。培养液中含滤纸酶活力平均为0.70IU/ml,还原糖26.41g/L,糖化率达到理论值的89.11%。固定化菌丝形态正常,菌量保持在10g/L左右。在间歇添料条件下,玉米秆原料的总量可提高到120g/L,7d后还原糖浓度达52.81g/L,糖化率为89.20%。利用固定化里氏木霉同时产酶和糖化植物纤维原料,工艺简便、成本低廉、易于连续自动化操作,是一条有效利用可再生纤维素资源的新途径。  相似文献

High power ultrasound (HPU) represents a non-thermal processing method that has been rapidly researched and used in the last 10 years. The application of power ultrasound offers the opportunity to modify and improve some technologically important compounds which are often used in food products. One of them is starch. The aim of this research was to examine the effect of the high power ultrasound of 24 kHz frequency on rheological and some physical properties of corn starch. Various ultrasound treatments were used; an ultrasound probe set with different intensities (34, 55, 73 W cm−2) and treatment times (15 and 30 min) and ultrasound bath of 2 W cm−2 intensity and treatment times (15 and 30 min). Rheological parameters, turbidity and swelling power of corn starch suspensions were determined for native and ultrasonically treated corn starch suspensions. Differential scanning calorimetry was used in order to examine the pasting properties of corn starch. The results have shown that the ultrasound treatment of corn starch distorts the crystalline region in starch granules. The results of differential scanning calorimetry measurements have shown a decrease in enthalpy of gelatinization. A significant decrease in consistency coefficient (k) has also been observed. The consistency coefficient decreases stepwise jointly with the increasing ultrasound power. The increase in the swelling power is associated with water absorption capacity and corn starch granules solubility, respectively.  相似文献

为探明贵州玉米田昆虫群落结构和杀虫灯对害虫的防治效果,选取贵州玉米田3个具有代表性的点分别安装诱虫灯,进行诱集调查,并对近灯区(距离灯20 m以内)、远灯区(距离灯20~150 m)和对照区的昆虫种群消长情况进行田间调查。结果表明,贵州玉米田昆虫共有11目66科132种,主要为同翅目、鞘翅目、鳞翅目、膜翅目、直翅目、双翅目和半翅目类昆虫;灯诱共获得89个种类,其中害虫占64.77%,益虫占15.91%,中性昆虫占19.32%。田间调查结果显示,近灯区物种丰富度最高,对照区次之,远灯区最低;近灯区玉米螟Ostrinia furnacalis、黏虫Mythimna seperata (Walker)、灰飞虱Laodelphax striatellus和条赤须盲蝽Trigonotylus coelestialium (Kirkaldy)等趋光性昆虫的种群数量长期高于其他区域。综上所述,风吸式诱虫灯对玉米田害虫有较好的防治效果,为玉米田害虫的绿色防控提供了参考。  相似文献

Sodium hydroxide and its derivatives are used as pulping reagents, wherein the spent NaOH is recovered in salt form and reused. In this study, use of low concentration NaOH (1–5%) in pretreatment of corn stover and hybrid poplar was investigated. It was done with the understanding that NaOH can be recovered. One of the main objectives in this study is to explore the potential of H2O2 with NaOH for pretreatment of high lignin substrate such as hybrid poplar. Pretreatment time has not been optimized in this study but held constant at 24 h. Corn stover, after treatment with NaOH under moderate conditions, attains near quantitative glucan digestibility. On the other hand, hybrid poplar requires treatment at higher temperature and NaOH concentration to attain acceptable level of digestibility. Supplementation of hydrogen peroxide in the pretreatment significantly raises delignification and digestibility of hybrid poplar. It was also helpful in retaining the carbohydrates in the treated solids. Retention of hemicellulose after pretreatment provides a significant economic benefit as it eliminates the need for detoxifying hemicellulose sugars. As the residual xylan remaining after pretreatment is an impediment to enzymatic digestion of glucan, supplementation of xylanase has significantly increased the digestibility of glucan as well as xylan of the treated hybrid poplar. © 2010 American Institute of Chemical Engineers Biotechnol. Vegetation is a major environmental factor influencing habitat selection in bird species. High resolution mapping of vegetation cover is essential to model the distribution of populations and improve the management of breeding habitats. However, the task is challenging for grassland birds because microhabitat variations relevant at the territory scale cannot be measured continuously over large areas to delineate areas of higher suitability. Remote sensing may help to circumvent this problem. We addressed this issue by using SPOT 5 imagery and phytosociological data. We mapped grassland vegetation in a floodplain using two methods. We (i) mapped the continuous Ellenberg index of moisture and (ii) identified 5 vegetation classes distributed across the wetness gradient. These two methods produced consistent output maps, but they also provided complementary results. Ellenberg index is a valuable proxy for soil moisture while the class approach provided more information about vegetation structure, and possibly trophic resources. In spite of the apparent uniformity of meadows, our data show that birds do not settle randomly along the moisture and vegetation gradients. Overall birds tend to avoid the driest vegetation classes, i.e. the highest grounds. Thus, vegetation maps based on remote sensing could be valuable tools to study habitat selection and niche partition in grassland bird communities. Seed-borne fungus Penicillium duclauxii was examined in this study to investigate its capability of synthesizing silver nanoparticles (Ag-NPs). In vitro experiments were conducted using corn-grain contaminating fungal isolate. Ag-NPs detection and characterization were assayed by the aid of spectroscopic techniques. Spectroscopy (energy dispersive), X-ray diffraction, transmission electron-microscope and optical absorption dimensions were employed.Ag-NPs with biosynthesized were used to test invitro against Bipolaris sorghicola; the cause of target leaf spot disease on sorghum plants. The myco-synthesis of Ag NPs using Penicillium duclauxii was proved in this study. Moreover, Bipolaris sorghicola was successfully inhibited by such Ag NPs in vitro.  相似文献

This study investigated the interaction between polyphenols with different cereal sources and their effects on performance, serum biochemistry, corticosterone levels and carcass characteristics in broiler chickens reared under hot-humid environment. Newly hatched coloured broiler chicks (n = 240) were randomly divided into six groups with five replicates of two different cereal sources, namely corn and broken rice-sorghum combined, and three levels of pomegranate peel polyphenol extracts (PPP) 0, 50 and 100 mg/kg in each cereal groups. Birds were maintained under standard management conditions for six weeks during hot-humid environment (Temperature: 29–36 °C; Relative Humidity: 69–80%). Fortnight body weight and feed consumption were recorded and serum biochemical constituents were estimated at 28 and 42 days of age. The body weight gain was significantly (p < 0.05) higher in broken rice-sorghum diet than in corn diet. The supplementation of polyphenols increased the skin and shank pigmentation. Serum protein, lipids and minerals showed significant difference due to cereals, polyphenols and their interactions. Corn-fed birds had significantly higher visceral organs weight than the alternate grain-fed broilers. The serum corticosterone levels were significantly reduced in the PPP supplemented groups than the control, it could be concluded that, supplementation of 50 mg/kg polyphenol extract from pomegranate peels improved production performance in broilers fed broken rice-sorghum as cereal source under hot-humid conditions.  相似文献

Water-soluble corn silk polysaccharides (CSPS) were chemically modified to obtain their sulfated, acetylated and carboxymethylated derivatives. Chemical characterization and bioactivities of CSPS and its derivatives were comparatively investigated by chemical methods, gas chromatography, gel filtration chromatography, scanning electron microscope, infrared spectroscopy and circular dichroism spectroscopy, scavenging DPPH free radical assay, scavenging hydroxyl radical assay, ferric reducing power assay, lipid peroxidation inhibition assay and α-amylase activity inhibitory assay, respectively. Among the three derivatives, carboxylmethylated polysaccharide (C-CSPS) demonstrated higher solubility, narrower molecular weight distribution, lower intrinsic viscosity, a hyperbranched conformation, significantly higher antioxidant and α-amylase inhibitory abilities compared with the native polysaccharide and other derivatives. C-CSPS might be used as a novel nutraceutical agent for human consumption.  相似文献

对粘性丝胞酵母进行紫外诱变,获得一株产油率较高的菌株,较原菌株提高了1.53倍。将该菌株接种于用1%硫酸和酶水解处理并浓缩至还原糖浓度为5%的玉米秸秆水解液中培养,生长较好。通过四因素三水平正交实验,确定培养条件为初始pH值7.0、接种量1%、发酵温度30℃、发酵时间5 d时产油率最高。对最佳产油条件进行验证,测得油脂含量为21.3%。从而为利用农业废弃物大规模生产微生物油脂提供了试验数据。气相色谱-质谱联用分析仪显示油脂脂肪酸组成为棕榈酸28.36%,油酸55.86%,10-十八烯酸9.23%,硬脂酸6.70%,可以作为原料生产生物柴油。  相似文献
