A transformation system for selected mature cork oak (Quercus suber L.) trees using Agrobacterium tumefaciens has been established. Embryos obtained from recurrent proliferating embryogenic masses were inoculated with A. tumefaciens strains EHA105, LBA4404 or AGL1 harbouring the plasmid pBINUbiGUSint [carrying the neomycin phosphotransferase II (nptII) and -glucuronidase (uidA) genes]. The highest transformation efficiency (4%) was obtained when freshly isolated explants were inoculated with A. tumefaciens strain AGL1. Evidence of stable transgene integration was obtained by PCR for the nptII and uidA genes, Southern blotting and expression of the uidA gene. Recent studies have demonstrated that growth rings are widespread in the roots of forbs, and there is evidence that the rings are formed annually. However, the annual nature and development of the growth rings has not yet been examined in comparative experimental studies. In this study growth rings were analysed in the main roots of four alpine forbs (Lotus alpinus, Trifolium thalii, Silene willdenowii and Potentilla aurea) that were grown in an alpine restoration experiment for 6 years. All individuals of L. alpinus and T. thalii, and some individuals of S. willdenowii showed six clearly demarcated growth rings, demonstrating that the rings were formed annually. P. aurea did not show distinguishable growth rings. In L. alpinus and T. thalii there were fluctuations in growth ring width that were consistent between individuals and also between species, and matched variations in climatic growth conditions. Results of the present study indicate that conclusions drawn from previous studies suggesting that growth rings in the roots of forb species are most likely formed annually are also valid for alpine plants. In terms of annual ring width patterns, this study also provides the first strong evidence for consistent responses of different forb species and individuals to commonly experienced variations in habitat conditions. 相似文献

A recently described phloem-bleeding technique was used to study seasonal changes in δ13C, sugar levels and the amino acid:sugar balance of phloem translocate of 2- to 3-year old trees of Eucalyptus globulus at a rain-fed site (Eulup) and a waste-effluent-irrigated site (Albany) in south-west Australia. δ13C of phloem sap from the Eulup site fluctuated widely between winter (−27.6‰) and peak summer stress (−20.2‰), compared with
a much smaller range of −28.4 to −26.3 at Albany. Seasonal changes in sugar concentrations in sap fluctuated closely with
those of phloem δ13C, with highest concentrations and least negative δ13C values at times of greatest soil water deficit. Molar ratios of amino acids to sugars in phloem sap were similar between
plantations in winter through to early summer. They then remained high at the nitrogen-rich effluent-treated site, but fell
dramatically once soils dried out at Eulup. Mature leaf dry matter sampled at peak yearly stress (early autumn) showed more
negative δ13C values than concurrently harvested phloem sap or recently initiated shoot apex dry matter, presumably because the sampled
foliage had laid down its structural carbon earlier under relatively unstressed winter/spring conditions. Differences between
Albany and Eulup were much greater for δ13C of phloem and new apical dry matter than for dry matter of mature foliage. Comparisons of δ13C signatures of phloem sap carbon with those of dry matter of nascent xylem tissues showed seasonal fluctuations in δ13C of phloem translocate which were mirrored a month or so later by those for xylem carbon. δ13C analyses of trunk growth rings from Eulup and Albany showed well-defined seasonal oscillations over the first 2 or 3 years
of growth until irrigation commenced at Albany. Fluctuations in δ13C at the latter site then became noticeably less pronounced than at Eulup. Future use of phloem sap δ13C and solute analyses for studying seasonal water and nutrient status of E. globulus is discussed.

Seasonally dry tropical forests are an important global climate regulator and represent one of main drivers of carbon sink dynamics. However, projections of climate change suggest future productivity losses and negative impacts on forest functioning. Understanding the interaction between climate variability and tree growth responses between species with different growth strategies represents a crucial challenge to forecast ecosystem functioning in the future. Here we used tree ring chronology to evaluate changes in growth and climate sensitivity of two tropical tree species that co-occur in a seasonally dry tropical forest in Brazil: Cedrela odorata and Ceiba glaziovii. Using Pearson correlations and linear regressions we explored how growth variability is correlated with local (precipitation, temperature) and global (ocean temperature and El Niño Southern Oscillation - ENSO) climatic factors. Tree growth was closely related with precipitation in C. odorata (r = 0.59) and C. glaziovii (0.24). Differences were found at monthly level, which C. odorata showing greater sensitivity in the beginning of rainy season. The South Atlantic Temperature was positively correlated with C. odorata, while ENSO was negatively correlated. Our results showed a dominant effect of precipitation on tree growth and suggest that are different growth strategies among species, which C. odorata being the most sensitive to drought and C. glaziovii more adapted with parenchyma in trunk. Therefore, C. odorata is probably more vulnerable to the deleterious effects of future climate change than C. glaziovii. Our findings highlight the importance of understanding the climate sensitivity of different seasonally dry tropical forest species, which is critical to predicting carbon dynamics in tropical regions. As tree rings can reveal various information regarding climate and environmental factors, increasing research is being conducted on them. Although tree ring analysis software such as Windendro has been applied, research on the development of analysis software using image preprocessing algorithms and deep learning is recently being attempted as computer vision technology. In this study, Mask R-CNN and linear interpolation were applied from images collected using a smartphone (SM-G973, Samsung, Suwon) and a scanner (CanoScan 9000F, Canon, Otaku) to propose an effective method for detecting tree ring boundaries. Pitch pine (Pinus rigida), Korean pine (Pinus koraiensis), white birch (Betula platyphylla), and cork oak (Quercus variabilis) were selected as tree species. Of the 300 images, 240 were classified as training data and 60 as validation data. As a result of learning, smartphones detected 86.0 % (381 ring boundaries) of the rings in pitch pine, 82.1 % (367 ring boundaries) in Korean pine, 84.7 % (309 ring boundaries) in white birch, and 78.7 % (318 ring boundaries) in cork oak. The scanner detected 93.2 % (413 ring boundaries) of the rings in pitch pine, 90.8 % (405 ring boundaries) in Korean pine, 88.2 % (322 ring boundaries) in white birch, and 89.4 % (361 ring boundaries) in cork oak. In particular, the smartphone showed satisfactory results of 84.7 % and 78.7 % for detecting tree ring boundaries of white birch and cork oak, where the boundaries of the rings were unclear. In the annual growth analysis results, both smartphones and scanners were statistically insignificant, and there was no difference compared with those of Windendro. Therefore, Mask R-CNN might be an effective approach for tree ring boundary detection as it showed satisfactory results, even with smartphones. In addition, although there was distortion in cases where images were acquired with a circular lens, there was no statistically significant difference from Windendro results. Summary The bark texture of Melia azedarach L. changes from smooth to furrowed as trees age. In trees that were cut down, those with smooth bark sprouted below the cut from suppressed buds; trees with thick, furrowed bark sprouted at the edge of the cut surface from adventitious buds. The trees that had thin, furrowed bark sprouted mainly at the edge of the cut from adventitious buds, but sometimes also from suppressed buds in cracks. Cork oak ( Quercus suber L.) zygotic embryos, endosperm and ovules were treated with different concentrations of 2,4-D for induction of somatic embryos. Plant material was collected during the embryo development season, from June to September. Immature embryos proved to be the most reactive initial explant. Callus and somatic embryos developed a few weeks after the beginning of the 2,4-D treatment. For embryo development experiments, different growth regulators and cold and desiccation treatments were tested. Cork oak (Quercus suber L.) is a monoecious wind-pollinated species with a protandrous system to ensure cross-pollination. To the best of our knowledge,
this report provides the first insight into the sexual reproduction cycle in this species. The cork oak flowering season extends
from April until the end of May. Our results show that, at anthesis, the pistillate flower is not completely formed and ovules
are just starting to develop. Pollen reaching the dry stigmatic surface adheres to the receptive cells, germinates and penetrates
the epidermis in aproximately 24 h, and grows through the intercellular spaces of a solid transmitting tissue. In cross-pollination,
a sequential arrest of pollen tubes was observed along the style, providing preliminary evidence for a pollen tube competition
mechanism. As a consequence, few pollen tubes reach the basal portion of the style. Furthermore, pollen tube growth is a discontinuous
process since tubes are arrested in the basal portion of the style about 10–12 days after pollination. While tubes are latent,
the ovarian loculus starts to develop from an emerging mass of sporogeneous cells which later will differentiate into the
placenta and ovules. One and a half months after pollination ovules complete their differentiation, tubes resume growth and
fertilisation occurs. Ovular abortion is frequent at this stage, and only one ovule will successfully mature during autumn
into a monospermic seed.

Since the XIX century, there is a controversy about the existence of biennial maturation of the acorns in Quercus suber L. While some authors recognised biennial cycles as an adaptation to habitats with short vegetative periods, other authors discarded the biennial pattern. Successive flowering events from spring to autumn and annual acorn ripening are proposed as an explanation of the multiple acorn crops typical of Iberian forests. To clarify this discussion, the presence of annual and biennial acorns was assessed in seven cork oak stands, covering a wide range of environmental conditions. In each stand, 100 individuals were sampled once in spring and once in autumn. Biennial acorns were observed with variable frequencies in all populations. There was a significant and positive relationship between latitude and the percentage of trees with biennial acorns within northern and central populations. On the contrary, this trend was not significant among southern populations. The hypothesis that the presence of biennial acorns in Quercus suber is related to individual female flowering phenology was confirmed in four populations located in the southwestern Iberian Peninsula. Unregarding local differences in the distribution of phenological stages anticipated trees bore significantly less biennial acorns than delayed individuals of the same stand. This result is coherent with the idea that the length of the vegetative period plays a crucial role in the frequency of annual and biennial acorn ripening patterns. The relationship between annual and biennial ripening cycles and the multiple acorn crops is discussed. 相似文献
