C3larvin toxin is a new member of the C3 class of the mono-ADP-ribosyltransferase toxin family. The C3 toxins are known to covalently modify small G-proteins, e.g. RhoA, impairing their function, and serving as virulence factors for an offending pathogen. A full-length X-ray structure of C3larvin (2.3 Å) revealed that the characteristic mixed α/β fold consists of a central β-core flanked by two helical regions. Topologically, the protein can be separated into N and C lobes, each formed by a β-sheet and an α-motif, and connected by exposed loops involved in the recognition, binding, and catalysis of the toxin/enzyme, i.e. the ADP-ribosylation turn–turn and phosphate–nicotinamide PN loops. Herein, we provide two new C3larvin X-ray structures and present a systematic study of the toxin dynamics by first analyzing the experimental variability of the X-ray data-set followed by contrasting those results with theoretical predictions based on Elastic Network Models (GNM and ANM). We identify residues that participate in the stability of the N-lobe, putative hinges at loop residues, and energy-favored deformation vectors compatible with conformational changes of the key loops and 3D-subdomains (N/C-lobes), among the X-ray structures. We analyze a larger ensemble of known C3bot1 conformations and conclude that the characteristic ‘crab-claw’ movement may be driven by the main intrinsic modes of motion. Finally, via computational simulations, we identify harmonic and anharmonic fluctuations that might define the C3larvin ‘native state.’ Implications for docking protocols are derived. 相似文献
Summary A short-term experiment was conducted to examine the relationships among the branching coral Porites porites, algal epibionts, and a facultative crab associate Mithrax sculptus in Belize, Central America. Initial field observations suggested that coral colonies supporting resident crabs generally had lower algal cover than colonies without crabs. The hypothesis was tested that Mithrax significantly depresses host coral algal cover and thereby indirectly affects host survivorship and growth. Crab accessibility to an array of coral colonies, similarly covered with algal epibionts, was manipulated in three treatments. Results strongly support the hypothesis, with significant differences in algal cover (primarily Dictyota spp.) noted among treatments after only one month. Caged heads with crabs included and uncaged natural controls allowing crabs free access averaged less than 10% cover, whereas mean algal cover exceeded 75% where crabs were excluded. The uncaged treatment, in which crabs were allowed free access to Porites heads was not significantly different from the crab inclusion treatment. Collectively, these results demonstrate that under natural conditions, crabs can have pronounced effects on host corals by reducing fouling algal epibionts. Furthermore, these facultative coral associates may have more important, albeit localized effects on Caribbean corals than has been suggested previously. 相似文献
The bioerosive potential of the intertidal chiton Acanthopleura gemmata on One Tree Reef was determined by quantification of CaCO3 in daily faecal pellet production of individuals transplanted into mesocosms after nocturnal-feeding forays. Mean bioerosive
potential was estimated at 0.16 kg CaCO3 chiton−1 yr−1. Bioerosion rates were estimated for populations on two distinct chiton habitats, reef margin (0.013 kg CaCO3 m−2 yr−1) and beachrock platform (0.25 kg CaCO3 m−2 yr−1). Chiton density on the platform was orders of magnitude greater than on the reef margin. The surface-lowering rate (0.16
mm m−2 yr) due to bioerosion by the beachrock population is a substantial contribution to the total surface-lowering rate of 2 mm
m−2 yr−1 previously reported for One Tree Reef across all erosive agents. At high densities, the contribution of A. gemmata to coral reef bioerosion budgets may be comparable to other important bioeroders such as echinoids and fish. 相似文献
In order to understand feeding ecology and habitat use of coral reef fish, fatty acid composition was examined in five coral reef fishes, Thalassoma lunare, Lutjanus lutjanus, Abudefduf bengalensis, Scarus rivulatus and Scolopsis affinis collected in the Bidong Island of Malaysian South China Sea.
Results
Proportions of saturated fatty acids (SAFA) ranged 57.2% 74.2%, with the highest proportions in fatty acids, the second highest was monounsaturated fatty acids (MUFA) ranged from 21.4% to 39.0% and the proportion of polyunsaturated fatty acids (PUFA) was the lowest ranged from 2.8% to 14.1%. Each fatty acid composition differed among fishes, suggesting diverse feeding ecology, habitat use and migration during the fishes’ life history in the coral reef habitats.
Conclusions
Diets of the coral fish species might vary among species in spite of that each species are living sympatrically. Differences in fatty acid profiles might not just be considered with respect to the diets, but might be based on the habitat and migration. 相似文献
The present study aimed to investigate the spatial structure of fish communities at juvenile and adult stages on coral reefs
at Kudaka Island (Ryukyu Archipelago, Japan) and to relate spatial patterns in the structure of the fish communities to gradients
in environmental variables. Diurnal visual censuses allowed us to record 2,602 juveniles belonging to 60 species and 1,543
adults belonging to 53 species from October to December 2005. The distribution of species highlighted that the juvenile community
was organised into three distinct assemblages, rather than exhibiting gradual change in community structure along the cross-reef
gradient. Correlations between spatial patterns of juvenile community and environmental variables revealed that the most significant
factors explaining variation in community structure were coral rubble and coral slab. In contrast, the adult community was
organised into one assemblage, and the most significant variation factors in community structure were depth, live coral in
massive form, live coral in branched form, dead coral and sand. Overall, the present study showed that most juvenile and adult
coral reef fish at Kudaka Island exhibited striking patterns in their distribution and depth and some biological factors (e.g.,
abundance of live coral, dead coral and coral rubble) might exert considerable influence on the distribution of fishes. 相似文献
In laboratory experiments, blackfly larvae collected from a lake outlet, a woodland and a meadow stream were tested for size
selection of latex beads of < Ito > 100 μm diameters. 3 suspensions of varying proportions for each size category were supplied
to these blackfly larvae in common experiments. Comparisons between the size frequency distributions of particles supplied
and the particle compositions in the larval guts showed intra- and interspecific differences and were quantified by calculating
Jacobs' electivity index. In all species selection of larger particles increased with the larger larval instars. Although
there was a positive selectivity of small particles in some cases, the ingested proportion of large particles increases volumes
and biomasses of gut content and may be more important for larval growth than small particles. 相似文献
A morphological study of the midgut and salivary glands of second and third instars of Gasterophilus intestinalis (De Geer) (Diptera: Oestridae) was conducted by light, scanning and transmission electron microscopy. The midgut is anteriorly delimited by a proventriculus, without caeca, and is composed of posterior foregut and anterior midgut tissue from which a double‐layered peritrophic matrix is produced. The midgut can be divided into anterior, median and posterior regions on the basis of the structural and physiological variations of the columnar cells which occur along its length. Two other types of cell were identified: regenerative cells scattered throughout the columnar cells, and, more rarely, endocrine cells of two structural types (closed and open). Different secretion mechanisms (merocrine, apocrine and microapocrine) occur along the midgut epithelium. Abundant microorganisms are observed in the endoperitrophic space of the anterior midgut. The origin and nature of these microorganisms remain unknown. No structural differences are observed between the second and third instar midguts. The salivary glands of G. intestinalis second and third instars consist of a pair of elongated tubular structures connected to efferent ducts which unite to form a single deferent duct linked dorsally to the pharynx. Several intermediate cells, without cuticle, make the junction with the salivary gland epithelium layer. Cytological characteristics of the gland epithelial cells demonstrate high cellular activity and some structural variations are noticed between the two larval stages. 相似文献
The objective of this study was to determine whether individuals of Lymnaea stagnalis naturally parasitised with trematode larvae had different thermal preferences from non-parasitised ones, and what was the influence of constant temperature on biology of the host and the parasite.
The snails under study were placed in and oblong thermal gradient and their preferences were recorded during 24 h. Non-parasitised individuals chose a rather high temperature (25 °C). In the group of parasitised snails, only individuals which released cercariae showed statistically significant differences in thermal preference than the non-larvae releasing ones. Parasitised snails kept in constant temperatures lived shorter than non-parasitised ones. At high temperatures, they released more cercariae per day than at low temperatures. Temperature had a great influence on the longevity and viability of cercariae.
The thermal behaviour of snails parasitised with trematode larvae is not a symptom of behavioural fever but probably can play a therapeutic role in the host. 相似文献
Structural refuges within which prey can escape from predators can be an important limiting resource for the prey. In a manner
that resembles the childhood game of musical chairs, many prey species rapidly retreat to shared, unguarded refuges whenever
a predator threatens, and only when refuges are relatively abundant do all prey individuals actually escape. The key feature
of this process is that the per capita prey mortality rate depends on the ratio of prey individuals to refuges. We introduce
a new class of mortality functions with this feature and then demonstrate statistically that they describe field mortality
data from a well-studied coral reef fish species, the Caribbean bridled goby Coryphopterus glaucofraenum, substantially better than do several mortality functions of more conventional form.

Diseases are causing alarming declines in reef‐building coral species, the foundation blocks of coral reefs. The emergence of these diseases has occurred simultaneously with large increases in the abundance of benthic macroalgae. Here, we show that physical contact with the macroalga Halimeda opuntia can trigger a virulent disease known as white plague type II that has caused widespread mortality in most Caribbean coral species. Colonies of the dominant coral Montastraea faveolata exposed to algal transplants developed the disease whereas unexposed colonies did not. The bacterium Aurantimonas coralicida, causative agent of the disease, was present on H. opuntia sampled close to, and away from diseased corals, indicating that the alga serves as a reservoir for this pathogen. Our results suggest that the spread of macroalgae on coral reefs could account for the elevated incidence of coral diseases over past decades and that reduction of macroalgal abundance could help control coral epizootics. 相似文献
