The overproduction of tumor necrosis factor-α (TNF-α) was suppressed by orally administering a perilla leaf extract (PLE). When mice were successively injected with OK-432, severe TNF-α was induced in the serum, but this elevated TNF-α level was reduced after an oral administration of PLE (400 μl/mouse). Oral administration of PLE also inhibited TNF-α production that was induced by muramyl dipeptide (500 μg/mouse) and OK-432 (3 KE/mouse). These characteristics were obtained from all strains of perilla. The inhibitory activity against TNF-α production was heat-stable, and the existence of several active molecules was suggested. When PLE was passed through an ultrafilter, the inhibitory activity against TNF-α production was collected in those fractions with a mass of 0.5 to 1 kDa and more than 10 kDa. When PLE was solvent-extracted, the strongest activity was recognized with aqueous preparation, although significant activity was also detected in preparations extracted with n-hexane and ethyl acetate. These findings suggest that the daily use of certain functional foods may be useful for controlling the host defense system.  相似文献

Dry forests are among the most endangered natural communities in the Hawaiian Islands. Most have been reduced to isolated trees and small forest fragments in which native tree species reproduce poorly. The replacement of native birds by introduced generalists may be contributing to dry forest decline through modification of seed dispersal patterns. To document seed dispersal by introduced birds, we conducted foraging observations on fleshy-fruited trees and measured seed rain under trees and in adjacent open areas for 1 year in a dry forest dominated by native trees. Although trees covered only 15.2 percent of the study area, 96.9 percent of the bird-dispersed seeds were deposited beneath them. The Japanese white-eye (Zosterops japonicus) was the principal dispersal agent. Among bird-dispersed seeds, those of the invasive tree Bocconia frutescens accounted for 75 percent of all seeds collected beneath trees (14.8 seeds/m2/yr) and the invasive shrub Lantana camara accounted for 17 percent. Although nearly 60 percent of the reserve's native woody species possess fleshy fruits, introduced birds rarely disperse their seeds. Native trees accounted for <8 percent of all bird-dispersed seeds and are consequently experiencing dispersal failure by falling directly under parent trees. Smaller-seeded non-native plants, in contrast, may be benefiting from dispersal by introduced birds. Current dispersal patterns suggest that these readily disseminated non-native plants may eventually replace the remaining native flora.  相似文献

An automated method for rapid and convenient measurement of L-glutamate has been developed by using a discrete analyzer, EEL Auto Chemist. It is based on the colorimetric measurement of NADH produced on a mole-mole basis by enzymatic dehydrogenation of L-glutamate using L-glutamate dehydrogenase from bovine liver. The values of L-glutamate obtained by this method were well agreed with those obtained by the routine Waruburg mano-metric method using L-glutamate decarboxylase from Escherichia coli.  相似文献

Normal roots of Capsicum frutescens were excised from tissue-cultured plants into half strength Murashige and Skoog's medium with 2.23 μM naphthalene acetic acid. Maximum growth of cultured roots was 6.5 g fresh weight 40 ml-1, as recorded on day 20. Even though normal roots were unable to accumulate capsaicin, they contained other phenylpropanoid intermediates and vanillylamine, as detected by HPLC analysis. Normal roots of Capsicum frutescens were treated with ferulic acid and protocatechuic aldehyde in order to study their biotransformation ability. Ferulic acid, which is the nearest precursor to vanillin, when fed at concentrations of 1 and 2 mM led to the accumulation of vanilla flavour metabolites, vanillin being the major one. In cultures treated with 1 and 2 mM ferulic acid, maximum vanillin accumulation of 12.3 and 16.4 μM was observed, on day 6 after precursor addition, respectively. Feeding of ferulic acid and β-cyclodextrin complex (2 mM each) enhanced the accumulation of biotransformed products. Moreover, vanillin accumulation was recorded as 24.7 μM on day 6 after precursor addition, which was 1.5 times higher than in cultures fed with ferulic acid (2 mM) alone. When ferulic acid was fed along with β-cyclodextrin (1 mM each) to cultures growing in a three-litre bubble column bioreactor, the maximum vanillin production of 10.7 μM was obtained; other vanilla flavour metabolites were also formed after 9 days of precursor addition. Root cultures could also biotransform protocatechuic aldehyde wherein a maximum vanillin production of 7.9 μM was recorded on day 6 after precursor addition. The bioconversion efficiency was observed to be 5–7% in case of ferulic acid fed cultures and 3.2% in case of protocatechuic aldehyde fed cultures suggesting the possible channelling of precursors to alternate biosynthetic pathways such as lignin.  相似文献

Abstract

Salt stress is one of the major environmental factors limiting crop growth and yield. To understand the effect of salt stress on plant growth, we investigated the response of three perilla varieties (Suyin 1, Ziye 7, and Ziye 10) to NaC1 exposure at concentrations of 0, 50, 100, 150, 200, and 250 mM in terms of seed germination, seedling growth, root activity, contents of soluble sugar, proline, and malondialdehyde (MDA), and peroxidase (POD) enzyme activity. Germination characteristics, such as the percentage of seed germination, tended to decrease with increasing NaC1 concentrations. After three weeks of salt stress, the three varieties exhibited different salt tolerance in terms of seed germination, seedling growth, and physiological changes: seedling growth was inhibited to various degrees, seedling vigor and root activities decreased, and MDA, proline, and soluble sugar contents increased with increasing NaCl concentrations. POD enzyme activity, a protective mechanism against salt stress, increased at low NaC1 concentrations in Suyin1 (0–150 mM) and Ziye 7 (0–100 mM), and then decreased at higher NaCl concentrations. In Ziye 10, on the other hand, POD activity dropped significantly with increasing NaCl concentrations. These results suggest that among the three varieties Suyin 1 is more salt tolerant than Ziye 7 and Ziye 10, and that Ziye 10 is the most sensitive to salt stress.  相似文献

以不同基因型紫苏为材料,研究150 mmol/L NaCl胁迫对紫苏种子萌发,幼苗生长及其生理指标的影响.结果显示:(1)盐胁迫下,不同基因型紫苏种子发芽率和发芽势均比对照显著降低,种子发芽指数、活力指数等发芽指标下降;不同基因型紫苏幼苗地上部和地下部干重均有不同程度降低,高油1(Y1)、紫野1(Z1)、紫野2(Z2)、紫野3(Z3)和紫野4(Z4)幼苗组织含水量均比对照显著降低,且幼苗存活率低而盐害指数显著高于其他基因型品种.(2)盐胁迫下不同基因型紫苏幼苗根系活力均比对照显著降低,其中Y1降幅最大,紫野6(Z6)和紫野7(Z7)降幅较小,可溶性糖含量均比对照显著提高.基因型Y1、Z1和Z2幼苗中MDA含量显著高于对照,表明其膜脂过氧化程度较高.(3)基因型苏引1(S1)、Z6、Z7、紫野8(Z8)、紫野9旱(ZY9)和紫野10(Z10)幼苗叶片中SOD、CAT和POD活性均显著高于对照,Y1、Z1、Z2、Z3和Z4幼苗中SOD和CAT活性与对照差异不显著,且Y1、Z2和Z4幼苗POD活性较对照显著降低,表明其抗氧化能力较差.研究发现,基因型S1、Z6和Z7在盐胁迫下其种子发芽能力、耐盐能力及幼苗渗透调节能力和活性氧清除能力均较强,表现出较强的耐盐性;基因型Y1、Z1、Z2、Z3和Z4受盐胁迫影响较大,为盐敏感基因型;Z8、Z9、ZY9和Z10为中等耐盐性基因型.  相似文献

Abstract. 1. Much has been learned in recent years regarding the influence of environmental conditions on top‐down and bottom‐up effects acting on insect herbivores. Temporal variation in environmental conditions, however, has gone largely unstudied in spite of undoubtedly strong influences in most systems. 2. A 2‐year study was conducted to examine the legacy effects of previous manipulations of host plant quality and parasitism pressure on the top‐down and bottom‐up effects influencing population densities of the salt marsh planthopper Pissonotus quadripustulatus. 3. For 10 months in 1998, a 2 × 2 factorial experiment was carried out, in which host plant quality was increased by the addition of nitrogen fertiliser, and parasitism pressure was decreased through the use of yellow sticky traps. This was followed by 2 months in the winter with no treatment applications. Treatments were then reversed in 1999 for a further 10 months. 4. In 1998, fertilisation treatments increased plant quality significantly, which resulted in strong effects on P. quadripustulatus density. Parasitism reduction treatments had weaker and time‐dependent effects on the herbivore, increasing planthopper density in late summer and autumn. 5. After 2 months without treatments, previous fertilisation treatments were still influencing all response variables measured significantly. The legacy effects of fertilisation persisted for at least 7 months for the host plant, and at least 5 months for the herbivore and parasitoid. 6. Fertilisation treatments in 1999 increased P. quadripustulatus density by approximately the same percentage as in 1998, suggesting that previous reductions in parasitism had no influence on herbivore responses to increased nutrients. Parasitism reduction treatments in 1999, however, resulted in greater increases in herbivore density than in 1998, suggesting that previous increases in nutrients enabled greater responses to reductions in top‐down pressure. 7. The results show that the top‐down effects of parasitism attenuated more quickly than did the bottom‐up effects of increased plant quality through greater nutrient availability. They also suggest that the recent history of nutrient status in an ecosystem may be important in determining the relative strengths of top‐down and bottom‐up forces.  相似文献

In vitro regeneration from leaf, cotyledon and hypocotyl explants of six cultivars belonging to three species of Capsicum was achieved by direct organogenesis. The cultivar Umorok showed the best response while Meiteimorok, Haomorok, Mashingkha and Uchithi showed intermediate response and the cultivar Chiengpi was the least responsive. Leaf and cotyledon explants regenerated more shoots than hypocotyl explants and the maximum number of shoots were produced on Murashige and Skoog (1962) medium containing 8.8 μM 6-benzylaminopurine (BAP) with 11.4 μM indole-3-acetic acid (IAA). Elongation of shoot buds derived from different explants was achieved on medium containing 2.8 μM IAA and the elongated shoots were rooted on medium containing 2.8 or 5.7 μM IAA and 2.4 or 4.9 μM indole-3-butyric acid (IBA). Four-week old rooted plantlets were hardened and transplanted to the soil. The plantlets showed 90 % survival during transplantation.  相似文献

Fine structural events in the mature stylar transmitting tissue ofsweet pepper (Capsicum frutescens L. var. grossum Bally) have been investigated byelectron microscope with regard to pollen tribe grwoth. The transmitting tissue consists of parenchymatous cells with large intercellular spaces filled with an electron densesubstance. The pollen tubes grow through the intercellular spaces in the intercellular substance. Cells of this tissue are rich in organelles, especially the rough and tubular ER, and numerous lomasomes near the plasma membrane. It is demonstrated that they function as secretory cells. On the other hand, the fact that the transmitting tissue contains many large amyloplastids with several starch grains in the cytoplasm and numerous globular protein bodies in vacuoles, indicates that the transmitting tissue mayhave some nutritive value for the growth of pollen tubes. The results obtained from this observation are in agreement with those of mostspecies reported by other authors and support the conclusion that the transmitting tissue is not a collenchyma tissue and the nature of intercellular substances is essentially same as that of the middle lamella.  相似文献
