Abstract

Explorative experiments were done to investigate the possibility that tomato plants infected by Botrytis cinerea have a different emission of volatile organic compounds (VOC) than healthy plants. This was done by headspace analysis of volatiles emitted by detached leaves of infected and healthy plants. Principal component analysis (PCA) of GC-FID chromatograms revealed clearly separated clusters between infected and control leaves and identification of an interesting compound. In further analysis by GC-MS, the significantly distinctive component (p≤0.05) was identified as the sesquiterpene α-copaene. In previous work on herbivore damage, α-copaene was not distinctive, which may suggest that α-copaene may be specifically associated to fungal infections in tomato.  相似文献

The development of natural crop protection products as alternatives to the use of synthetic fungicides is currently popular. The aim of this study is to evaluate the antifungal effects of several essential oils against the fungal pathogens, Botrytis cinerea and Rhizopus stolonifer, under in vitro condition. Four essential oils (fennel, black caraway, peppermint and thyme) were each tested at five concentrations (0, 200, 400, 600 or 800 μl l?1). In vitro results showed that the essential oil of black caraway and fennel had the highest fungicidal effect against B. cinerea and R. stolonifer, respectively. The growth of B. cinerea was completely inhibited by the essential oil of black caraway at 400 μl l?1. Fennel oil perfectly inhibited growth of R. stolonifer fungus colonies at concentration higher than 600 μl L?1 in potato dextrose agar medium. Percentage of spores germination was the lowest in medium of Fennel and black caraway essential oils, and was the highest in Thyme ones. These results show that plant essential oils can have a strong effect on reducing post-harvest decay. These plant essential oils could provide an alternative to synthetic chemicals to control post-harvest phytopathogenic fungi on fruit.  相似文献

Compartmentalization of proteinases, amylases, and pH in the midgut of Nauphoeta cinerea Oliv. (Blattoptera:Blaberidae) was studied in order to understand the organization of protein and starch digestion. Total proteolytic activity measured with azocasein was maximal at pH 11.5 both in anterior (AM) and posterior (PM) halves of the midgut, but the bulk of activity (67%) was found in PM. Total AM and PM preparations were fractionated on a Sephadex G-50 column and further analysed by means of activity electrophoresis and specific inhibitors and activators. The major activity in PM was classified as an unusual SH-dependent proteinase with M(r) 24,000 and pH optimum with synthetic substrate BApNA at 10.0. The enzyme was 43-fold activated in the presence of 1 mM DTT, insensitive to synthetic inhibitors of serine (PMSF, TLCK, TPCK) and cysteine (IAA, E-64) proteinases, strongly inhibited by STI, and displayed four active bands on zymograms. In PM, activities of trypsin-like, chymotrypsin-like, subtilisin-like, and cysteine proteinases were observed. Aspartic and metalloproteinases were not detected. In AM, activity of unusual SH-dependent proteinase also dominated and activity of chymotrypsin-like proteinase was observed, but their levels were much lower than in PM. Distribution of amylase activity, exhibiting an optimum at pH 6.0, was quite the opposite. The major part of it (67%) was located in AM. Treatment of amylase preparation with proteinases from AM and PM reduced amylase activity twofold. pH of the midgut contents was 6.0-7.2 in AM, 6.4-7.6 in the first and 8.8-9.3 in the second halves of PM. Thus, pH in AM is in good agreement with the optimal pH of amylase, located in this compartment, but the activity of proteinases, including the ability to degrade amylase, in such an environment is low. Active proteolysis takes place in the second half of PM, where pH of the gut is close to the optimal pH of proteinases.  相似文献

The aim of this study was to investigate the effect of nitrogen availability on susceptibility of tomato leaves to the fungal pathogen Botrytis cinerea. Plants with varying nitrogen availability were grown by adding N daily in exponentially increasing amounts to a nutrient solution at different rates. Leaves of plants grown at low nitrogen availability had a high leaf C/N ratio (21 g g-1) and were about 2.5 times more susceptible to primary lesion formation by B. cinerea compared to plant grown at high nitrogen availability, which had a low leaf C/N ratio (11 g g-1). Leaf C/N ratio accounted for 95% of variation in susceptibility. This relationship between C/N ratio and susceptibility persisted when plants were grown with exponential P addition and optimal N supply, and was thus independent of plant growth rate or related factors. We could not explain the effect of nitrogen availability by variation in the most obvious N-based resistance compound α-tomatine because more susceptible leaves with a high C/N ratio contained more α-tomatine. These leaves also contained more soluble carbohydrates. The level of soluble carbohydrates correlated positively with susceptibility, independent of the growth method. We therefore suggest that the effect of N availability on susceptibility must be explained by variation in levels of soluble carbohydrates and speculate about the role of these carbohydrates in the infection process. The red light-induced antifungal substance(s) produced in broad bean was of relatively high molecular weight, water soluble, heat stable and fungi specific. Cellulose thin layer chromatography (TLC) of infection droplets of Botrytis cinerea or water droplets without spores of B. cinerea, recovered from inoculated broad bean leaflets kept under red light for 48 h, displayed inhibition zones at approximate Rf values of 0.0 and 0.6. Inhibition zones observed in cellulose TLC of water droplets were relatively faint compared to those of infection droplets. In a time-course study of accumulation of the antifungal substance(s), antifungal activity in both water and infection droplets recovered from red light irradiated broad bean leaflets occurred after 24 h irradiation. However, the antifungal activity in infection droplets was significantly higher than in water droplets. The antifungal substance(s) was less active against Botrytis fabae than B. cinerea.  相似文献

Cytoplasmic soluble proteins from ungerminated conidia of Botrytis cinerea exhibited cutinase activity, while cell wall binding proteins lacked this activity. Cutinase activity in proteins extracted from cell walls and cytoplasm of ungerminated conidia of Botrytis cinerea was determined using p-nitrophenyl butyrate (PNB) and TLC analysis of products derived from hydrolysis of [3H]cutin. Treatment of conidia with indoxyl acetate, a substrate indicative of non-specific esterase and cutinase activity, also gave a positive reaction in the cytoplasm of ungerminated conidia. The possible role of a putative constitutive cutinase in the cytoplasm of conidia in the early stages of infection of plants by B. cinerea is discussed.  相似文献

The new methylated grindelane diterpenoid, 7β ‐hydroxy‐8(17)‐dehydrogrindelic acid ( 1b ), together with the known 7α ‐hydroxy‐8(17)‐dehydrogrindelic acid ( 2a ), 6‐oxogrindelic acid ( 3a ), 4β ‐hydroxy‐6‐oxo‐19‐norgrindelic ( 4a ), 19‐hydroxygrindelic acid ( 5a ), 18‐hydroxygrindelic acid ( 6a ), 4α ‐carboxygrindelic acid ( 7a ), 17‐hydroxygrindelic acid ( 8a ), 6α ‐hydroxygrindelic acid ( 9a ), 8,17‐bisnor‐8‐oxagrindelic acid ( 10a ), 7α ,8α ‐epoxygrindelic acid ( 11a ), and strictanonic acid ( 12a ) as methyl esters were obtained from an Argentine collection of Grindelia chiloensis (Cornel .) Cabrera . Their structures and relative configurations were established on the basis of spectroscopic analysis. CHC l3 extract from the aerial parts and their pure compounds were evaluated for their antifungal and depigmenting effects. Methyl ester derivative of 10a ( 10b ) exhibited a remarkable mycelial growth inhibition against Botritis cinerea with an IC 50 of 13.5 μg ml?1. While the new grindelane 1b exerted a clear color reduction of the yellow‐orange pigment developed by Fusarium oxysporum against UV ‐induced damage.  相似文献

与水库水位低、繁殖群已经形成的2000-2001年相比, 苍鹭(Ardea cinerea) 的种群数量在水位正常、尚未繁殖的1990-1991年相对较低。该种鸟类在水库的3个地点取食。1990-1991年的8 月或9 月, 其数量达到最高值, 并有较高比例的个体出现在水库深水区的平坦的岸边。2000-2001 年的数量高峰在6 月份, 有大量的觅食个体出现在回水区, 在那里形成了新的生境(水塘、浅水区和沙洲)。苍鹭迁离或飞往繁殖地的活动在早晨或中午之前强于中午或下午。在其食物中共鉴别出17种鱼类, 其中数量最多的是圆鳍雅罗鱼和拟鲤。在苍鹭食物中, 游动性鱼类比例较高(2001年占32%, 2003年占25%), 表明它是在河流和水库之间的生态交错区取食的。所捕食鱼类的全长为4 cm到33 cm, 生物量在2-406 g之间。在2001 年和2003 年苍鹭食物中鱼类的全长未见明显差异。  相似文献
