Hybrid density functional theory with the B3LYP functional has been used to investigate the catalytic mechanism of catechol oxidase. Catechol oxidase belongs to a class of enzymes that has a copper dimer with histidine ligands at the active site. Another member of this class is tyrosinase, which has been studied by similar methods previously. An important advantage for the present study compared to the one for tyrosinase is that X-ray crystal structures exist for catechol oxidase. The most critical step in the mechanism for catechol oxidase is where the peroxide O–O bond is cleaved. In the suggested mechanism this cleavage occurs in concert with a proton transfer from the substrate. Shortly after the transition state is passed there is another proton transfer from the substrate, which completes the formation of a water molecule. An important feature of the mechanism, like the one for tyrosinase, is that no proton transfers to or from residues outside the metal complex are needed. The calculated energetics is in reasonable agreement with experiments. Comparisons are made to other similar enzymes studied previously.  相似文献

New copper(II) complexes [CuL2]2+ (L2=7,7,9-trimethyl-1,3,6,10,13-pentaazabicyclo[11,2,11.13]hexadec-9-ene) and [Cu2(L3)(H2O)2]4+ have been prepared by the reaction of [CuL1]2+ (L1=5,5,7-trimethyl-1,4,8,11,14-pentaazatetradce-7-ene) and formaldehyde. The mononuclear complex [CuL2]2+ has a square-planar coordination geometry with a 5-6-5-6 chelate ring sequence and is relatively stable even in low pH at room temperature. The dinuclear complex [Cu2(L3)(H2O)2]4+ consists of two unsaturated 15-membered pentaaza macrocyclic units (7,7,9-trimethyl-1,3,6,10,13-pentaazacyclopentadec-9-ene) that are linked together by a methylene group in a tilted face-to-face arrangement [Cu?Cu distance: 7.413(2) Å ]. Each macrocyclic unit of [Cu2(L3)(H2O)2]4+ contains one four-membered chelate ring and has a severely distorted octahedral coordination polyhedron. The dinuclear complex is quite stable in aqueous solutions containing an excess of formaldehyde or in dry acetonitrile but is decomposed to [CuL1]2+ and [CuL2]2+ in pure water.  相似文献

The variations in the coordination environment of Co(II), Cu(II) and Zn(II) complexes with the neutral, tridentate ligand bis[1-(cyclohexylimino)ethyl]pyridine (BCIP) are reported. Analogous syntheses were carried out utilizing either the M(BF4)2 · xH2O or MCl2 · xH2O metal salts (where M = Co(II), Cu(II) or Zn(II)) with one equivalent of BCIP. When the hydrated metal starting material was used, cationic, octahedral complexes of the type [M(BCIP)2]2+ were isolated as the tetrafluoroborate salt (4, 5). Conversely, when the hydrated chloride metal salt was used as the starting material, only neutral, pentacoordinate [M(BCIP)Cl2] complexes (1-3) formed. All complexes were characterized by X-ray diffraction studies. The three complexes that are five coordinate have distortions due mainly to the pyridine di-imine bite angle. The [Cu(BCIP)Cl2] (2) also exhibits deviations in the Cu(II)-Cl bond distances with values of 2.4242(9) and 2.2505(9) Å, which are not seen in the analogous Zn(II) and Co(II) structures. Similarly, the two six coordinate complexes (5, 6) are also altered by the ligand frame bite angle giving rise to distorted octahedral geometries in each complex. The [Cu(BCIP)2](BF4)2 (6) also exhibits Cu(II)-Nimine bond lengths that are on average 0.14 Å longer than those found in the analogous 5 coordinate complex, [Cu(BCIP)Cl2]. In addition to X-ray analysis, all complexes were also characterized by UV/Vis and IR spectroscopy with 1H NMR spectroscopy being used for the analysis of the Zn(II) analogue (3).  相似文献

The effects of excess copper (Cu) on the accumulation of hydrogen peroxide (H2O2) and antioxidant enzyme activities in roots of the Cu accumulator Elsholtzia haichowensis Sun were investigated. Copper at 100 and 300 μM significantly increased the concentrations of malondialdehyde and H2O2, and the activities of catalase (E.C. 1.11.1.6), ascorbate peroxidase (E.C. 1.11.1.11), guaiacol peroxidase (GPOD, E.C. 1.11.1.7) and superoxide dismutase (SOD, E.C. 1.15.1.1). Isoenzyme pattern and inhibitor studies showed that, among SOD isoforms, only copper–zinc superoxide dismutase (CuZn–SOD) increased. Excess Cu greatly increased the accumulation of superoxide anion (O2 ·−) and H2O2 in E. haichowensis roots. This study also provides the first cytochemical evidence of an accumulation of H2O2 in the root cell walls as a consequence of Cu treatments. Experiments with diphenyleneiodonium as an inhibitor of NADPH oxidase, 1,2-dihydroxybenzene-3,5-disulphonic acid as an O2 ·− scavenger, and N-N-diethyldithiocarbamate as an inhibitor of SOD showed that the source of H2O2 in the cell walls could partially be NADPH oxidase. The enzyme can use cytosolic NADPH to produce O2 ·−, which rapidly dismutates to H2O2 by SOD. Apoplastic GPOD and CuZn–SOD activities were induced in roots of E. haichowensis with 100 μM Cu suggesting that these two antioxidant enzymes may be responsible for H2O2 accumulation in the root apoplast.  相似文献

Bioavailability is integral in mediating the delicate balance between nutritive and potentially toxic levels of copper in fish diets. Brush-border membrane vesicles isolated from freshwater rainbow trout intestine were used to characterise apical copper absorption, and to examine the influence of the amino acid histidine on this process. In the absence of histidine, a low affinity, high capacity copper uptake mechanism was described. However, when expressed as a function of ionic copper (Cu2+), absorption was linear, rather than saturable, suggesting that the saturable curve was an artifact of copper speciation. Conversely, in the presence of l-histidine (780 μM) saturable uptake was characterised. The uptake capacity discerned (J max of 354 ± 81 nmol mg protein−1 min−1) in the presence of histidine indicated a significantly reduced capacity for copper transport than that in the absence of histidine. To determine if copper uptake was achievable through putative histidine uptake pathways, copper and histidine were incubated in the presence of tenfold greater concentrations of amino acids proposed to block histidine transporters. Accounting for changes in copper speciation, significant inhibition of uptake by glycine and lysine were noted at copper levels of 699 and 1,028 μM. These results suggest that copper–histidine complexes may be transportable via specific amino acid-transporters in the brush-border membrane.  相似文献

The new Cu/Hg bimetallic thiocyanato-bridged coordination polymer, [Cu(2-bzpy)Hg(SCN)4]n, has been synthesized and characterized by elemental analysis, magnetic measurements, EPR, and X-ray crystallography. Dinuclear copper(I) complexes with bridging bis(dicyclohexylphosphino)methane (dcpm) or bis(diphenylphosphino)methane (dppm) and 2,2′-bipyridine or 2-[N-(2-pyridyl)methyl]amino-5,7-dimethyl-1,8-naphthyridine (L), [Cu2(bpy)2(dppm)2](BF4)2 (1), [Cu2(bpy)2(dcpm)](BF4)2 (2), [Cu2(L)(dppm)](BF4)2 (3) and [Cu2(L)(dcpm)](BF4)2 (4) were prepared, and their structures were determined by X-ray crystal analysis. Two-, three-, and four-coordinate copper(I) centers are found in these complexes. Compounds 3 and 4 show close CuI?CuI separations of 2.664(3) and 2.674(1) Å, respectively, whereas an intramolecular copper-copper distance of 3.038 Å is found in 2 having only dcpm as an additional bridge. Powdered samples of 1, 3, and 4 display intense and long-lived phosphorescence with λmax at 533, 575, and 585 nm at room temperature, respectively. In the solid state, 2 exhibits only a weak emission at 555 nm. The time-resolved absorption and emission spectra of these complexes were investigated. The difference in the emission properties among complexes 1-4 suggests that both CuI?CuI distances and coordination environment of the copper(I) centers affect the excited-state properties.  相似文献

The nuclearity, bonding and H-bonded networks of copper(I) halide complexes with thiophene-2-carbaldehyde thiosemicarbazones {(C4H3S)HC2N3-N(H)-C1(S)N1HR} are influenced by R substituents at N1 atom. Thiophene-2-carbaldehyde-N1-methyl thiosemicarbazone (HttscMe) or thiophene-2-carbaldehyde-N1-ethyl thiosemicarbazone (HttscEt) have yielded halogen-bridged dinuclear complexes, [Cu2(μ-X)21-S-Htsc)2(Ph3P)2] (Htsc, X: HttscMe, I, 1; Br, 2; Cl, 3; HttscEt, I, 4; Br, 5; Cl, 6), while thiophene-2-carbaldehyde-N1-phenyl thiosemicarbazone (HttscPh) has yielded mononuclear complexes, [CuX(η1-S-HttscPh)2] (X, I, 7a; Br 8; Cl, 9) and a sulfur bridged dinuclear complex, [Cu2(μ-S-HttscPh)21-S-HttscPh)2I2] 7b co-existing with 7a in the same unit cell. These results are in contrast to S-bridged dimers [Cu2(μ-S-Httsc)21-Br)2(Ph3P)2] · 2H2O and [Cu2(μ-S-Httsc)21-Cl)2(Ph3P)2] · 2CH3CN obtained for R = H and X = Cl, Br (Httsc = thiophene-2-carbaldehyde thiosemicarbazone) as reported earlier. The intermolecular CHPh?π interaction in 1-3 (2.797 Å, 1; 3.264 Å, 2; 3.257 Å, 3) have formed linear polymers, whereas the CHPh?X and N3?HCH interactions in 4-6 (2.791, 2.69 Å, 5; 2.776, 2.745 Å, 6, respectively) have led to the formation of H-bonded 2D polymer. The PhN1H?π, interactions (2.547 Å, 8, 2.599 Å, 9) have formed H-bonded dimers only. The Cu?Cu separations are 3.221-3.404 Å (1-6).  相似文献

采用病情指数调查、DAS-ELISA和生理生化指标测定的方法,研究了铜(Cu)元素对烟草抗马铃薯Y病毒脉坏死株系(Potatovirus Y-vein necrosis strain,PVYN)的室内效果及对植株抗病性的诱导作用。结果表明:喷施不同浓度的铜元素均能延迟烟株显症和叶脉出现褐色坏死时间,降低发病程度,烟株体内病毒含量也明显减少,当Cu元素浓度为0.8mg/L时,效果最显著。单独喷施Cu元素(0.8mg/L)和喷施Cu元素后再接种PVYN的处理,均能提高烟株体内叶绿素a、叶绿素b含量及苯丙氨酸解氨酶活性;另外,单独喷施Cu元素处理烟株后能诱导总酚和类黄酮含量的提高,但喷施Cu元素后接种PVYN的烟株中总酚和类黄酮的含量在接种处理后3d内迅速上升高于对照,随后下降,在6d后虽又呈缓慢上升趋势,但仍明显低于对照。总酚和类黄酮的含量与叶脉褐变密切相关,因此总酚和类黄酮含量的降低可减轻植株褐变。研究表明,Cu元素具有诱导抗病性、增强烟草对PVYN侵染的抵抗能力。  相似文献
