根瘤菌对土壤铜、锌和镉形态分配的影响   总被引:13,自引:0,他引:13  
以湖南郴州红壤和河北巩义褐土为供试土壤。制备Cu、Zn、Cd污染土壤。接种大豆根瘤菌(Rhi-zobium fredii)HN01,用连续提取法浸提土壤中不同形态的重金属.结果表明。褐土接种根瘤菌后固相结合态Zn总量降低10%。专性吸附态、氧化锰结合态和有机结合态Zn减少达9%~26%.红壤中结合态Zn的总量变化不显著,但专性吸附态和氧化锰结合态Zn含量显著减少。交换态Zn含量显著增加.褐土中接种根瘤菌抑制了Cu向土壤溶液的释放,固相结合态Cu总量增加18%,可交换态、专性吸附态、氧化锰结合态和有机结合态的Cu增加20%~54%.接种根瘤菌对土壤中Cd的溶解没有明显的抑制或促进作用,但改变了红壤中各形态Cd的含量高低顺序.Cd污染红壤中可交换态和有机结合态Cd含量分别增加22%和11%,专性吸附态和氧化锰结合态Cd分别减少14%和29%.根瘤菌对不同类型重金属及不同土壤中重金属形态影响的差异主要与土壤pH降低有关.  相似文献

Equilibrium sorption isotherm for metal ions on tree fern   总被引:6,自引:0,他引:6  
A new sorbent system for removing heavy metal ions, such as Zn(II), Cu(II) and Pb(II), from aqueous solutions has been investigated. This new sorbent is tree fern, an agriculture product. Variables of the system include solution temperature and sorbent particle size. The experimental results were fitted to the Langmuir, Freundlich and Redlich–Peterson isotherms to obtain the characteristic parameters of each model. Both the Langmuir and Redlich–Peterson isotherms were found to well represent the measured sorption data. According to the evaluation using the Langmuir equation, the maximum sorption capacities of metal ions onto tree fern were 7.58 mg/g for Zn(II), 10.6 mg/g for Cu(II) and 39.8 mg/g for Pb(II). It was noted that an increase in temperature resulted in a higher metal loading per unit weight of the sorbent. Decreasing the particle sizes of tree fern led in an increase in the metal uptake per unit weight of the sorbent.  相似文献

Particulate methane monooxygenase (pMMO) has been exfoliated and isolated from membranes of the Methylosinus trichosporium IMV 3011. It appears that the stability of pMMO in the exfoliation process is increased with increasing copper concentration in the growth medium, but extensive intracytoplasmic membrane formed under higher copper concentration may inhibit the exfoliation of active pMMO from membrane. The highest total activity of purified pMMO is obtained with an initial concentration of 6 microM Cu in the growth medium. The purified MMO contains only copper and does not utilize NADH as electron donor. Treatment of purified pMMO with EDTA resulted in little change in copper level, suggesting that the copper in the pMMO is tightly bound with pMMO.  相似文献

Chronic (14 days) and acute (48 h) copper effects on the antioxidant defense system and some physiological variables of Laeonereis acuta (Polychaeta, Nereididae) were evaluated. In both assays, two nominal copper concentrations (chronic: C1=31.25 and C2=62.50 μg/l; acute: A1=250 and A2=500 μg/l) and one control group (Cc and Ac=0 μg/l) were tested. End points analyzed were antioxidant enzyme activity (catalase, CAT; superoxide dismutase, SOD; and glutathione S-transferase, GST), oxygen consumption, metahemoglobin concentration, and lipid peroxidation (LPO). In the chronic assay, CAT activity was significantly higher in worms exposed to both concentrations of copper tested (C1=3.36±0.07 U CAT/mg protein; C2=4.06 0.32 U CAT/mg protein) than in control worms (Cc=2.16±0.39 U CAT/mg protein). SOD activity was also increased in the two copper-exposed groups (C1=16.85±4.22 U SOD/mg protein; C2=38.19±4.31 U SOD/mg protein) than in control group (Cc=3.54±0.46 U SOD/mg protein). However, GST activity was increased only in worms exposed to the higher copper concentration (C2=0.022±9.10−4 U GST/mg protein) when compared to the other groups tested (Cc=0.012±3.10−3 U GST/mg protein; C1=0.016±9.10−4 U GST/mg protein). None of the physiological variables analyzed (oxygen consumption, metahemoglobin concentration, and lipid peroxidation) was affected by chronic copper exposure. In the acute assay, only GST activity was induced in worms exposed to copper. This induction was observed only in the A1 group (0.027±2.10−3 U GST/mg protein) when compared to Ac (0.017±2.10−3 U GST/mg protein) or A2 (0.016±7.10−4 U GST/mg protein) groups. On the other hand, lipid peroxidation was higher in A2 (481.9±49.2 nmol CHP/g ww) than in control worms (Ac=337.9±25.0 nmol CHP/g ww). Oxygen consumption was higher in worms acutely exposed to the lower copper concentration tested (A1=0.27±0.04 mg O2/g ww/h) than in the higher concentration (A2=0.14±0.01 mg O2/g ww/h). Changes in the swimming behavior of copper-exposed animals in both assays and edemas in the body wall of worms acutely exposed to copper were also observed. Results suggest that copper exposure favors reactive oxygen species generation and that enzymatic defense system is induced under chronic exposure, preventing oxygen consumption changes and lipid peroxidation and metahemoglobin formation. However, in acutely exposed worms, in spite of a transient peak of GST activity, no induction of antioxidant enzymes occurs, leading to morphological and physiological changes.  相似文献

Ceruloplasmin (CP) oxidises low density lipoprotein (LDL). The oxidising potential depends on the formation of Cu(+)-CP which is redox-cycled during oxidation. Homocysteine (HCY) reduces free Cu(2+), potentiating its cell-damaging property. We show that HCY enhanced LDL oxidation by CP, but did not activate the LDL oxidising potential of Cu(2+)-diamine oxidase. Selective removal of the redox-active Cu(2+) abolished the LDL oxidase activity of CP. However, HCY partially restored the LDL oxidase activity of redox-copper depleted CP, indicating that the remaining six copper atoms in CP may also be involved in the process. Spectroscopic and oxidation inhibition studies using the Cu(+)-reagent bathocuproine revealed that HCY induced Cu(+)-CP formation, thus promoting its LDL oxidase activity.  相似文献

Cysteine (Cys) residues are major causes of crystallin disulfide formation and aggregation in aging and cataractous human lenses. We recently found that disulfide linkages are highly and partly conserved in β- and γ-crystallins, respectively, in human age-related nuclear cataract and glutathione depleted LEGSKO mouse lenses, and could be mimicked by in vitro oxidation. Here we determined which Cys residues are involved in disulfide-mediated crosslinking of recombinant human γD-crystallin (hγD). In vitro diamide oxidation revealed dimer formation by SDS-PAGE and LC-MS analysis with Cys 111-111 and C111-C19 as intermolecular disulfides and Cys 111-109 as intramolecular sites. Mutation of Cys111 to alanine completely abolished dimerization. Addition of αB-crystallin was unable to protect Cys 111 from dimerization. However, Cu2+-induced hγD-crystallin aggregation was suppressed up to 50% and 80% by mutants C109A and C111A, respectively, as well as by total glutathionylation. In contrast to our recently published results using ICAT-labeling method, manual mining of the same database confirmed the specific involvement of Cys111 in disulfides with no free Cys111 detectable in γD-crystallin from old and cataractous human lenses. Surface accessibility studies show that Cys111 in hγD is the most exposed Cys residue (29%), explaining thereby its high propensity toward oxidation and polymerization in the aging lens.  相似文献

Summary Leek plants were preinoculated with a mixed inoculum ofGlomus caledonium, Glomus fasciculatum andGlomus sp., and transplanted to Dazomet disinfected and untreated field plots of moderate P deficiency. Successive harvests were made until 99 days after transplanting. Preinoculated leeks attained marketable weights 25 days earlier than uninoculated leeks from untreated soil and their final dry matter yields were 5.7 and 1.5 times as high as those of uninoculated leeks from disinfected and untreated soil, respectively. Phosphorus concentration in preinoculated leeks remained highest for at least 22 and 75 days after transplanting in untreated and disinfected soil, respectively. Preinoculation had a similar, although smaller, influence on Cu and Zn concentrations. Infection levels produced by introduced and indigenous VA endophytes in leeks reached plateaus of 90% and 40%, respectively, 47 days after transplanting. It is concluded that VAM is essential to leeks grown in moderately P deficient soils, and the potential for inoculating seedlings in commercial leek production is discussed.  相似文献

A novel outer mitochondrial membrane protein containing [2Fe-2S] clusters, mitoNEET was first identified through its binding to the anti-diabetic drug pioglitazone. Pioglitazone belongs to a family of drugs that are peroxisome proliferator-activated receptor (PPAR) gamma agonists, collectively known as glitazones. With the lack of pharmacological tools available to fully elucidate mitoNEET's function, we developed a binding assay to probe the glitazone binding site with the aim of developing selective and high affinity compounds. We used multiple thiazolidine-2,4-dione (TZD), 2-thioxothiazolidin-4-one (TTD), and 2-iminothiazolidin-4-one (ITD) compounds to establish several trends to enhance ligand development for the purpose of elucidating mitoNEET function.  相似文献

The interaction of four fluorescent compounds containing thiophene and benzoxazole moieties combined with an alanine residue with alkaline, alkaline-earth, transition and post-transition metal ions was explored. The highly fluorescent heterocyclic alanine derivatives are strongly quenched in the solid state after complexation with the paramagnetic metal ions Cu2+ and Ni2+, and with the diamagnetic Hg2+. Absorption and steady-state fluorescence titrations reveal a selective interaction with Cu2+, Ni2+ and Hg2+. In all cases the formation of mononuclear or dinuclear metal complexes in solid state and in solution are postulated. DFT calculations on the mercury(II) complexes confirm the formation of dinuclear species. Our results suggest that one metal ion is coordinated by the chelate group formed by the amine and the protonated carboxylic groups present in the amino acid residue while a second metal ion is directly linked to the chromophore. As parent compound, L4 shows no interaction with Cu2+ and Ni2+ salts. However, the interaction with Hg2+ induces a strong quenching and a red shift of the fluorescence emission.  相似文献

应用脱氧核糖降解法研究了离体条件下Cu,Zn-SOD与H2O2反应产生·OH,并对其机理进行了探讨。H2O2可使Cu,Zn-SOD失活,在失活过程中有·OH产生,甲酸钠和苯甲酸钠均能不同程度地保护Cu,Zn-SOD和降低H2O2与Cu,Zn-SOD反应中·OH的产额;热失活SOD也可和H2O2反应生成·OH,且效能高于活性Cu,Zn-SOD;用螫合剂脱去Cu,Zn-SOD的金属辅基后,脱辅基的SOD蛋白不能和H2O2反应产生·OH;Cu2+和H2O2反应产生·OH的效率很高,而Zn2+产生·OH的效率很低。实验结果提示Cu,Zn-SOD与H2O2反应产生的·OH可能是SOD活性中心的Cu2+与H2O2发生Fenton反应的结果.  相似文献
