3-Deazaadenosine and 5′-deoxy-5′-isobutylthio-3-deazaadenosine (3-deaza-SIBA) inhibits replication of both herpes simplex type 1 virus and the RNA type C virus, HL-23. Oncogenic transformation caused by SV40 and HL-23 are also blocked by either compound. Both compounds exhibit relatively low cytotoxicity at the anti-viral concentrations. 相似文献

Control of cell proliferation is vital for the normal development of the neural retina. Gap junctional communication has been implicated in the control of retinal cell proliferation. We have previously shown that the expression of the gap junction protein Connexin 43 closely correlates with the first wave of cell proliferation in the retina. Preventing its expression using antisense oligonucleotides in the developing eye and surrounding tissues, produces a reduction in cell number and the formation of a small eye. In order to examine this in more detail we have developed a new means of manipulating connexin expression in the developing chick embryo. We have generated pIRES vectors which use cyclomegalovirus (CMV) to promote the expression of a green fluorescent protein (EGFP) and either wild type Cx43 or a dominant negative form of this connexin. Following injection of these constructs into the ventricles of the stage 10-11 chick embryo they can be incorporated into one side of the chick brain or optic vesicle using an electroporation technique, leaving the other side as a control. EGFP expression can be seen on the electroporated side of the chick brain within 24hours. Expression of the dominant negative construct in cultures of chick limb bud mesenchyme results in total block of cascade blue transfer when injected into transfected cells. Expression of both wild type and dominant negative constructs in the developing chick retina perturbs the normal development of the eye. 相似文献

The purpose of this study was to investigate the biological effect of miR‐16 on myocarditis and the underlying molecular mechanism. H9c2 cells were treated with 10 µg/mL lipopolysaccharide (LPS) for 12 hours to form a myocarditis injury model. We observed that LPS treatment distinctly decreased the level of miR‐16 in H9c2 cells. Upregulation of miR‐16 increased cell proliferation and reduced cell apoptosis. Then, CD40 was predicted and verified as a target gene of miR‐16 by TargetScan and luciferase reporter assay, respectively. Furthermore, the messenger RNA and protein expression of CD40 are negatively regulated by miR‐16. The relative expression of inflammatory factors was dramatically decreased by the miR‐16 mimic. Cells cotransfected with miR‐16 mimic and si‐CD40 could significantly abolish the injury of cardiomyocytes caused by myocarditis. Our study illustrated that the upregulation of miR‐16 has a protective effect on LPS‐damaged H9c2 cells, which may be achieved by regulating CD40 and the nuclear factor kappa B pathway. 相似文献

Immune checkpoint inhibitors improved the survival rate of patients with unresectable melanoma. However, some patients do not respond, and variable immune‐related adverse events have been reported. Therefore, more effective and antigen‐specific immune therapies are urgently needed. We previously reported the efficacy of an immune cell therapy with immortalized myeloid cells derived from induced pluripotent stem cells (iPS‐ML). In this study, we generated OX40L‐overexpressing iPS‐ML (iPS‐ML‐Zsgreen‐OX40L) and investigated their characteristics and in vivo efficacy against mouse melanoma. We found that iPS‐ML‐Zsgreen‐OX40L suppressed the progression of B16‐BL6 melanoma, and prolonged survival of mice with ovalbumin (OVA)‐expressing B16 melanoma (MO4). The number of antigen‐specific CD8+ T cells was higher in spleen cells treated with OVA peptide‐pulsed iPS‐ML‐Zsgreen‐OX40L than in those without OX40L. The OVA peptide‐pulsed iPS‐ML‐Zsgreen‐OX40L significantly increased the number of tumor‐infiltrating T lymphocytes (TILs) in MO4 tumor. Flow cytometry showed decreased regulatory T cells but increased effector and effector memory T cells among the TILs. Although we plan to use allogeneic iPS‐ML in the clinical applications, iPS‐ML showed the tumorgenicity in the syngeneic mice model. Incorporating the suicide gene is necessary to ensure the safety in the future study. Collectively, these results indicate that iPS‐ML‐Zsgreen‐OX40L therapy might be a new method for antigen‐specific cancer immunotherapy. 相似文献

Atherosclerosis is a chronic inflammatory disease arising due to an imbalance in lipid metabolism and maladaptive immune response driven by the accumulation of cholesterol-laden macrophages in the artery wall. Interactions between monocytes/macrophages and endothelial cells play an essential role in the pathogenesis of atherosclerosis. In our current study, nitric oxide synthase 1 (NOS1)-derived nitric oxide (NO) has been identified as a regulator of macrophage and endothelial cell interaction. Oxidized LDL (OxLDL) activates NOS1, which results in the expression of CD40 ligand in macrophages. OxLDL-stimulated macrophages produce some soluble factors which increase the CD40 receptor expression in endothelial cells. This increases the interaction between the macrophages and endothelial cells, which leads to an increase in the inflammatory response. Inhibition of NOS1-derived NO might serve as an effective strategy to reduce foam cell formation and limit the extent of atherosclerotic plaque expansion. 相似文献
