In tissues and/or organs of mesenchymal origin, the vasculature is usually well developed. However, there are certain hypovascular tissues that exhibit powerful anti-angiogenic resistance, implying the presence of tissue-type specific inhibitors of angiogenesis. Hyaline cartilage is one example, and several anti-angiogenic factors have been purified from cartilage. We previously identified chondromodulin-I (ChM-I) as a tissue-specific inhibitor of angiogenesis in fetal bovine cartilage. ChM-I is specifically expressed in the avascular regions of the growth-plate and cartilaginous bone rudiments in embryos. Recently, we cloned a novel type II transmembrane protein, tenomodulin (TeM), having a domain homologous to ChM-I at its C-terminus. TeM turned out to be expressed specifically in other hypovascular structures in the mesenchyme, such as the epimysium, tendon, and ligaments. In this overview, we discuss the structural characteristics of this class of anti-angiogenic molecules and their pathophysiological role in the control of vascularity. 相似文献

Elaboration of size and shape in multicellular organisms involves coordinated cell division and cell growth. In higher plants, continuity of cell layer structures exists from the shoot apical meristem (SAM), where organ primordia arise, to mature aboveground organs. To unravel the extent of inter-cell layer coordination during SAM and aboveground organ development, cell division in the epidermis was selectively restricted by expressing two cyclin-dependent kinase inhibitor genes, KRP1/ICK1 and KRP4, driven by the L1 layer-specific AtML1 promoter. The transgenes conferred reduced plant size with striking, distorted lateral organ shape. While epidermal cell division was severely inhibited with compensatory cell size enlargement, the underlying mesophyll/cortex layer kept normal cell numbers and resulted in small, packed cells with disrupted cell files. Our results demonstrate the autonomy of cell number checkpoint in the underlying tissues when epidermal cell division is restricted. Finally, the L1 layer-specific expression of both KRP1/ICK1 and KRP4 showed no effects on the structure and function of the SAM, suggesting that the effects of these cyclin-dependent kinase inhibitors are context dependent. 相似文献

of short time intervals (TE) of 2 h are strongly coupled to the circadian rhythm in sleepwakefulness. There is a linear correlation
between the number of hours humans stay awake (α) and their estimation of 2 h intervals. The coupling of TE to α appears to

The mechanisms that regulate neuronal function are a sum of genetically determined programs and experience. The effect of
experience on neuronal function is particularly important during development, because early-life positive and adverse experience
(stress) may influence the still “plastic” nervous system long-term. Specifically, for hippocampal-mediated learning and memory
processes, acute stress may enhance synaptic efficacy and overall learning ability, and conversely, chronic or severe stress
has been shown to be detrimental. The mechanisms that enable stress to act as this “double-edged sword” are unclear. Here,
we discuss the molecular mediators of the stress response in the hippocampus with an emphasis on novel findings regarding
the role of the neuropeptide known as corticotropin-releasing hormone (CRH). We highlight the physiological and pathological
roles of this peptide in the developing hippocampus, and their relevance to the long-term effects of early-life experience

The Photosystem II complex (PSII) is susceptible to inactivation by strong light, and the inactivation caused by strong light is referred to as photoinactivation or photoinhibition. In photosynthetic organisms, photoinactivated PSII is rapidly repaired and the extent of photoinactivation reflects the balance between the light-induced damage (photodamage) to PSII and the repair of PSII. In this study, we examined these two processes separately and quantitatively under stress conditions in the cyanobacterium Synechocystis sp. PCC 6803. The rate of photodamage was proportional to light intensity over a range of light intensities from 0 to 2000 μE m−2 s−1, and this relationship was not affected by environmental factors, such as salt stress, oxidative stress due to H2O2, and low temperature. The rate of repair also depended on light intensity. It was high under weak light and reached a maximum of 0.1 min−1 at 300 μE m−2 s−1. By contrast to the rate of photodamage, the rate of repair was significantly reduced by the above-mentioned environmental factors. Pulse-labeling experiments with radiolabeled methionine revealed that these environmental factors inhibited the synthesis de novo of proteins. Such proteins included the D1 protein which plays an important role in the photodamage-repair cycle. These observations suggest that the repair of PSII under environmental stress might be the critical step that determines the outcome of the photodamage-repair cycle. 相似文献

Dobutamine is a β-adrenergic agonist with an affinity higher for receptor expressed in the heart (β1) than for receptors expressed in the arteries (β2). When systemically administered, it increases cardiac demand. Thus, dobutamine unmasks abnormal rhythm or ischemic areas potentially at risk of infarction. Monitoring of heart function during a cardiac stress test can be performed by either ecocardiography or cardiac catheterization. The latter is an invasive but more accurate and informative technique that the former.Cardiac stress test induced by dobutamine and monitored by cardiac catheterization accomplished as described here allows, in a single experiment, the measurement of the following hemodynamic parameters: heart rate (HR), systolic pressure, diastolic pressure, end-diastolic pressure, maximal positive pressure development (dP/dtmax) and maximal negative pressure development (dP/dtmin), at baseline conditions and under increasing doses of dobutamine.As expected, in normal mice we observed a dobutamine dose-related increase in HR, dP/dtmax and dP/dtmin. Moreover, at the highest dose tested (12 ng/g/min) the cardiac decompensation of high fat diet-induced obese mice was unmasked. 相似文献

A Monte Carlo procedure is proposed for testing homogeneity of variances in linear models. The method is applicable to a variety of common experimental designs. It is valid when errors are independently normally distributed. Under nonnormality the test is expected to behave robust in a similar fashion as Levene's test. The reconstitution of bovine cardiac troponin from its subunits has been investigated using hydrodynamic techniques. Gel filtration (Sephacryl S-300) and sedimentation velocity experiments indicate that troponin-C and troponin-I from a stable binary complex (1:1 mole ratio) with an apparent Stokes' radius of 36 Å (frictional ratio = 1.6). Troponin-C and troponin-T do not interact significantly while troponin-I and troponin-T undergo partial complex formation. The effect of subunit ratio on the reconstitution of whole troponin has been examined by SDS—polyacrylamide gel electrophoresis and gel filtration and the results suggest that native troponin contains the subunits in an equimolar ratio. 相似文献
