For animal development it is necessary that organs stop growing after they reach a certain size. However, it is still largely unknown how this termination of growth is regulated. The wing imaginal disc of Drosophila serves as a commonly used model system to study the regulation of growth. Paradoxically, it has been observed that growth occurs uniformly throughout the disc, even though Decapentaplegic (Dpp), a key inducer of growth, forms a gradient. Here, we present a model for the control of growth in the wing imaginal disc, which can account for the uniform occurrence and termination of growth. A central feature of the model is that net growth is not only regulated by growth factors, but by mechanical forces as well. According to the model, growth factors like Dpp induce growth in the center of the disc, which subsequently causes a tangential stretching of surrounding peripheral regions. Above a certain threshold, this stretching stimulates growth in these peripheral regions. Since the stretching is not completely compensated for by the induced growth, the peripheral regions will compress the center of the disc, leading to an inhibition of growth in the center. The larger the disc, the stronger this compression becomes and hence the stronger the inhibiting effect. Growth ceases when the growth factors can no longer overcome this inhibition. With numerical simulations we show that the model indeed yields uniform growth. Furthermore, the model can also account for other experimental data on growth in the wing disc.  相似文献

While the use of the Internet for the exchange of scientific data was characterised by exclusivity during its pioneer era, the active employment of the medium today, by a broad social spectrum of users in the exchange of information, for dialogue and in the accumulation of knowledge, displays an almost unbounded inclusion. Blo and online encyclopaedias based on the 'Wikipedia' model have contributed to the formation of a marketplace in which the free expression of opinions and the relaying of information occur. Counted among the ideas which have been popularised in the wake of this phenomenon, "lay journalism" and the "wisdom of the masses" are seen to be integral to the new 'web 2.0'. Consequently, the ever-increasing information disseminated in the web has been diluted in quality and authenticity, resulting in the presentation of new challenges to online science journalism. In reference to the public debate surrounding green gene technology, the communications platform bioSicherheit.de, which receives more than one million visitors per year, will be examined as an example of an agent that retrieves and mobilises information on biological safety research and that successfully has established itself as an intermediary between the scientific community and the broader public.  相似文献

In sports analytics, an understanding of accurate on-field 3D knee joint moments (KJM) could provide an early warning system for athlete workload exposure and knee injury risk. Traditionally, this analysis has relied on captive laboratory force plates and associated downstream biomechanical modeling, and many researchers have approached the problem of portability by extrapolating models built on linear statistics. An alternative approach would be to capitalize on recent advances in deep learning. In this study, using the pre-trained CaffeNet convolutional neural network (CNN) model, multivariate regression of marker-based motion capture to 3D KJM for three sports-related movement types were compared. The strongest overall mean correlation to source modeling of 0.8895 was achieved over the initial 33% of stance phase for sidestepping. The accuracy of these mean predictions of the three critical KJM associated with anterior cruciate ligament (ACL) injury demonstrate the feasibility of on-field knee injury assessment using deep learning in lieu of laboratory embedded force plates. This multidisciplinary research approach significantly advances machine representation of real-world physical models with practical application for both community and professional level athletes.  相似文献

Non-native disulfide isomers of α-conotoxins are generally inactive although some unexpectedly demonstrate comparable or enhanced bioactivity. The actions of “globular” and “ribbon” isomers of α-conotoxin AuIB have been characterized on α3β4 nicotinic acetylcholine receptors (nAChRs) heterologously expressed in Xenopus oocytes. Using two-electrode voltage clamp recording, we showed that the inhibitory efficacy of the ribbon isomer of AuIB is limited to ∼50%. The maximal inhibition was stoichiometry-dependent because altering α3:β4 RNA injection ratios either increased AuIB(ribbon) efficacy (10α:1β) or completely abolished blockade (1α:10β). In contrast, inhibition by AuIB(globular) was independent of injection ratios. ACh-evoked current amplitude was largest for 1:10 injected oocytes and smallest for the 10:1 ratio. ACh concentration-response curves revealed high (HS, 1:10) and low (LS, 10:1) sensitivity α3β4 nAChRs with corresponding EC50 values of 22.6 and 176.9 μm, respectively. Increasing the agonist concentration antagonized the inhibition of LS α3β4 nAChRs by AuIB(ribbon), whereas inhibition of HS and LS α3β4 nAChRs by AuIB(globular) was unaffected. Inhibition of LS and HS α3β4 nAChRs by AuIB(globular) was insurmountable and independent of membrane potential. Molecular docking simulation suggested that AuIB(globular) is likely to bind to both α3β4 nAChR stoichiometries outside of the ACh-binding pocket, whereas AuIB(ribbon) binds to the classical agonist-binding site of the LS α3β4 nAChR only. In conclusion, the two isomers of AuIB differ in their inhibitory mechanisms such that AuIB(ribbon) inhibits only LS α3β4 nAChRs competitively, whereas AuIB(globular) inhibits α3β4 nAChRs irrespective of receptor stoichiometry, primarily by a non-competitive mechanism.  相似文献

Two experiments were conducted to test for the recovery of brucella organisms from uterine flushings and harvested embryos of sero-positive embryo donor females. In Experiment I, 16 sero-positive cows were superovulated with FSH treatments and artificially inseminated at 12, 24 and 36 hours following the onset of estrus with brucella-free semen. At 48 hours after the onset of estrus, one half the potential donor females were administered an intrauterine inoculation of 3.3 to 4.6 × 104Brucellaabortus (strain 2308) organisms while the remainder received a control inoculation. In Experiment II, the same 16 cows were similarly administered superovulatory treatments and inseminated following estrus. The uterine inoculation was increased to 1.5 to 2.5 × 108 organisms administered 48 hours following estrus. Samples of recovered flushing medium and homogenized embryo residues were placed into a validated invitro culture system to detect the presence of brucella bacteria. Uterine flushings and embryos recovered from 31 females exhibiting estrus following FSH treatments were free from either field strain or the inoculated B.abortus (strain 2308) contamination. The flushings obtained from a single female, which did not respond with estrus following FSH treatment but was inoculated at appointment, did contain B.abortus which was identified as the inoculated strain 2308 and not field strain organisms. These results indicate that brucella contamination of flushing media and harvested embryos will not likely be incurred when collecting embryos from sero-positive donor females. These findings offer further encouragement for the use of embryo transplantation as a method to produce brucella-free offspring from infected cows.  相似文献

The receptor tyrosine kinase Met is crucial for the genetic program causing cancer progression and metastasis. Its nodal function during aggressiveness and resistance acquisition poses Met inhibition as an obligatory step in anti-cancer targeted therapy. Here, we applied a “Met-focussed” forward chemical biological screen to discover new agents antagonizing Met-triggered biological functions. The identified new scaffold, JLK1360, has a dual mechanism of action towards Met: it impairs Met signalling and also prevents its restoration after degradation. Docking and molecular dynamics provide evidences on the interacting mode of JLK1360 within the Met ATP-binding pocket. Moreover, computational and biochemical studies also highlighted that JLK1360 has a good degree of selectivity towards Met than other RTKs tested. Altogether, these findings demonstrate that the approach we have applied is a powerful strategy to identify compounds with combined properties towards a chosen target. Our studies show how integration of chemistry, biology and computational analysis can provide robust strategies to identify new inhibitory scaffolds suitable for further development of anti-cancer targeted therapies.  相似文献

The selenoenzyme glutathione peroxidase cannot account for all the physiological effects of selenium in rat liver. Therefore, a study was carried out with the ultimate aim of identifying selenoproteins other than glutathione peroxidase. The incorporation of 75Se, given as 75SeO32?, into centrifugally separated fractions of selenium-deficient and control rat livers was determined. In selenium-deficient liver much less 75Se was incorporated into the 105,000g supernatant fraction than in controls, so this fraction was studied further by gel filtration, ion-exchange, and hydroxylapatite chromatography. Selenoglutathione peroxidase and another selenoprotein, called 75Se-P, were separated and identified. Both these selenoproteins were also found in plasma. Selenium deficiency had opposite effects on incorporation of 75Se by these proteins. It decreased 75Se incorporation by glutathione peroxidase at 3 and 72 h after 75Se injection but increased 75Se incorporation by 75Se-P. This suggests that 75Se-P competes for available selenium better than does glutathione peroxidase when the element is in short supply. Apparent molecular weights of 75Se-P from liver and plasma determined by gel filtration were, respectively, 83,000 and 79,000, which indicate proteins smaller than glutathione peroxidase. Cycloheximide pretreatment of the rat blocked 75Se incorporation into plasma 75Se-P. These experiments establish the existence of a selenoprotein, 75Se-P, in rat liver and plasma which is chromatographically distinct from glutathione peroxidase and which incorporates 75Se differently from glutathione peroxidase. 75Se-P may account for some of the physiological effects of selenium.  相似文献

鱼类游泳动力学分析研究对解决鱼道等工程应用中水力学设计方面的关键问题有着重要的意义,利用计算机技术对鱼类游泳动力学进行分析有助于研究目标鱼类的生理特性、游泳能力及其与水力环境因子的响应关系。基于MATLAB软件对我国特有鱼类鲢幼鱼进行游泳动力学分析,借助鲢幼鱼游泳时的摆尾行为,得到不同水流速度下鲢幼鱼的摆尾频率、摆尾幅度、游泳速度和加速度;对比人工计数和手动跟踪分析方法,从实际操作复杂程度和实验数据准确性的角度,分析各数据采集方法的优劣性。结果表明基于Matlab软件采用跟踪鱼的身体中线的思路能更高效的获取大量的运动参数,比如摆尾频率、摆尾幅度、游泳速度和加速度等指标。文章介绍了一种基于Matlab开发的鱼类游泳动力学分析方法,有助于为以后鱼类游泳动力学研究提供依据。  相似文献
