Plants have large families of proteins sharing a conserved eight-cysteine-motif (8CM) domain. The biological functions of
these proteins are largely unknown. EARLI1 is a cold responsive Arabidopsis gene that encodes a hybrid proline-rich protein (HyPRP) with a three-domain architecture:
a putative signal peptide at the N-terminus, a proline-rich domain (PRD) in the middle, and an 8CM domain at the C-terminus.
We report here that yeast cells expressing different EARLI1 genes had significantly higher rates of freezing survival than empty-vector transformed controls. Arabidopsis plants with
knocked down EARLI1 genes had an increased tendency for freezing-induced cellular damage. EARLI1-GFP fluorescence in transgenic plants and immunoblot
analyses using protoplasts suggested cell wall localization for EARLI1 proteins. Immunoblot analyses showed that EARLI1 proteins
form higher order complexes in plants, and that the PRD is a soluble and the 8CM an insoluble protein domain. We propose that
EARLI1 proteins have a bimodular architecture in which the PRD may interact with the cell wall and the 8CM domain with the

Osmotin is a pathogenesis-related protein exhibiting cryoprotective functions. Our aim was to understand whether it is involved
in the cold acclimation of the olive tree (Olea europaea L.), a frost-sensitive species lacking dormancy. We exposed olive trees expressing tobacco osmotin gene under the 35S promoter (35S:osm) [in the same manner as wild type (wt) plants] to cold shocks in the presence/absence of cold acclimation, and monitored
changes in programmed cell death (PCD), cytoskeleton, and calcium ([Ca2+]c) signalling. In the wt, osmotin was immunolocalized only in cold-acclimated plants, and in the tissues showing PCD. In the
35S:osm clones, the protein was detected also in the non-acclimated plants, and always in the tissues exhibiting PCD. In the non-acclimated
wt protoplasts exposed to cold shock, a transient decrease in phallotoxin signal suggests a temporary disassembly of F-actin,
a transient increase occurred instead in 35S:osm protoplasts exposed to the same shock. Transient increases in [Ca2+]c were observed only in the wt protoplasts. However, when F-actin was depolymerized by cytochalasin or latrunculin, and microtubules
by colchicine, increase in [Ca2+]c also occurred in the 35S:osm protoplasts. Successive cold shocks caused transient rises in [Ca2+]c and transient decreases in the phallotoxin signal in wt protoplasts. No change occurred in [Ca2+]c occurred in the 35S:osm protoplasts. The phallotoxin signal transiently increased at the first shock, but did not change after the subsequent shocks,
and an overall signal reduction occurred with shock repetition. Following acclimation, no cold shock-induced change in [Ca2+]c levels and F-actin signal occurred either in wt or 35S:osm protoplasts. The results show that osmotin is positively involved in the acclimation-related PCD, in blocking the cold-induced
calcium signalling, and in affecting cytoskeleton in response to cold stimuli. 相似文献

Central to the debate surrounding global climate change and Plio-Pleistocene hominin evolution is the degree to which orbital-scale climate patterns influence low-latitude continental ecosystems and how these influences can be distinguished from regional volcano-tectonic events and local environmental effects. The Pliocene Hadar Formation of Ethiopia preserves a record of hominin paleoenvironments from roughly 3.5 to 2.2 Ma at a temporal resolution relevant to evolutionary change within hominins and other taxa. This study integrates the high-resolution sedimentological and paleontological records at Hadar with climate proxies such as marine core isotope, dust, and sapropel records. Consistent cycling observed both between and within fluvial and lacustrine depositional environments prior to 2.9 Ma at Hadar appears to be predominantly climatic in nature. In contrast a significant change in depositional facies after 2.9 Ma to sequences dominated by conglomerate cut-and-fill cycles indicates a strong tectonic signature related to regional developments in the Main Ethiopian Rift. While specific events seen in marine proxy records may have parallels in the Hadar environmental archive, their overall patterns of high versus low variability may be even more relevant. For example, periods of relatively high-amplitude climate oscillations between 3.15 and 2.95 Ma may be linked to noted size-related morphological changes within the Hadar Australopithecus afarensis lineage and a significant increase in more arid-adapted bovid taxa. Increased aridity in East Africa during this period is also indicated by peaks in eolian dust in the marine core record. Conversely, the dominant lacustrine phase at Hadar ca. 3.3 Ma coincides with the least variable period in several climate proxy records, including marine core foraminifera delta(18)O values and eolian dust concentration. This phase is also coeval with low insolation variability and a very distinct and significant long-term period of low dust percentage in circum-Africa marine cores. 相似文献

Recent research in Europe, Africa, and Southeast Asia suggests that we can no longer assume a direct and exclusive link between anatomically modern humans and behavioral modernity (the 'human revolution'), and assume that the presence of either one implies the presence of the other: discussions of the emergence of cultural complexity have to proceed with greater scrutiny of the evidence on a site-by-site basis to establish secure associations between the archaeology present there and the hominins who created it. This paper presents one such case study: Niah Cave in Sarawak on the island of Borneo, famous for the discovery in 1958 in the West Mouth of the Great Cave of a modern human skull, the 'Deep Skull,' controversially associated with radiocarbon dates of ca. 40,000 years before the present. A new chronostratigraphy has been developed through a re-investigation of the lithostratigraphy left by the earlier excavations, AMS-dating using three different comparative pre-treatments including ABOX of charcoal, and U-series using the Diffusion-Absorption model applied to fragments of bones from the Deep Skull itself. Stratigraphic reasons for earlier uncertainties about the antiquity of the skull are examined, and it is shown not to be an 'intrusive' artifact. It was probably excavated from fluvial-pond-desiccation deposits that accumulated episodically in a shallow basin immediately behind the cave entrance lip, in a climate that ranged from times of comparative aridity with complete desiccation, to episodes of greater surface wetness, changes attributed to regional climatic fluctuations. Vegetation outside the cave varied significantly over time, including wet lowland forest, montane forest, savannah, and grassland. The new dates and the lithostratigraphy relate the Deep Skull to evidence of episodes of human activity that range in date from ca. 46,000 to ca. 34,000 years ago. Initial investigations of sediment scorching, pollen, palynomorphs, phytoliths, plant macrofossils, and starch grains recovered from existing exposures, and of vertebrates from the current and the earlier excavations, suggest that human foraging during these times was marked by habitat-tailored hunting technologies, the collection and processing of toxic plants for consumption, and, perhaps, the use of fire at some forest-edges. The Niah evidence demonstrates the sophisticated nature of the subsistence behavior developed by modern humans to exploit the tropical environments that they encountered in Southeast Asia, including rainforest. 相似文献

Hatchlings of the painted turtle, Chrysemys picta, hibernate terrestrially and can survive subfreezing temperatures by supercooling or by tolerating the freezing of their
tissues. Whether supercooled or frozen, an ischemic hypoxia develops because tissue perfusion is limited by low temperature
and/or freezing. Oxidative stress can occur if hatchlings lack sufficient antioxidant defenses to minimize or prevent damage
by reactive oxygen species. We examined the antioxidant capacity and indices of oxidative damage in hatchling C. picta following survivable, 48 h bouts of supercooling (−6°C), freezing (−2.5°C), or hypoxia (4°C). Samples of plasma, brain, and
liver were collected after a 24 h period of recovery (4°C) and assayed for Trolox-equivalent antioxidant capacity (TEAC),
thiobarbituric acid reactive substances (TBARS), and carbonyl proteins. Antioxidant capacity did not vary among treatments
in any of the tissues studied. We found a significant increase in TBARS in plasma, but not in the brain or liver, of frozen/thawed
hatchlings as compared to untreated controls. No changes were found in the concentration of TBARS or carbonyl proteins in
supercooled or hypoxia-exposed hatchlings. Our results suggest that hatchling C. picta have a well-developed antioxidant defense system that minimizes oxidative damage during hibernation. 相似文献

A total of 35 Brazilian isolates of Clostridium difficile from faecal stools and four isolates from hospital environments were analyzed by PCR ribotyping. A whole cell protein profile
(as an alternative for serogrouping), in vitro toxin production and susceptibility to vancomycin, metronidazole and clindamycin
were also investigated. All strains were typeable by both phenotypic and genotypic methods, and a total of 13 different PCR
ribotypes were identified, of which seven (132, 133, 134, 135, 136, 142 and 143) were considered new types and accounted for
78.5% of all samples evaluated (including hospital environments). A non-toxigenic C. difficile PCR ribotype 133 was detected in all children groups examined (inpatients, outpatients and healthy children), whilst toxigenic
PCR ribotypes 015, 131, 134 and 135 were associated mostly with symptomatic children. Serogroups G and D were disseminated
both in patients from the community and from the pediatric hospital, with group G prevalent among outpatient children. All
strains were susceptible to vancomycin and metronidazole but high levels of resistance to clindamycin were found, especially
among serogroups G and D. Co-existence of different ribotypes and serogroups in the same individual was observed. The new

AIM: To assess how completely the diversity of anoxygenic phototrophic bacteria (APB) was sampled in natural environments. METHODS AND RESULTS: All nucleotide sequences of the APB marker gene pufM from cultures and environmental clones were retrieved from the GenBank database. A set of cutoff values (sequence distances 0.06, 0.15 and 0.48 for species, genus, and (sub)phylum levels, respectively) was established using a distance-based grouping program. Analysis of the environmental clones revealed that current efforts on APB isolation and sampling in natural environments are largely inadequate. Analysis of the average distance between each identified genus and an uncultured environmental pufM sequence indicated that the majority of cultured APB genera lack environmental representatives. CONCLUSIONS: The distance-based grouping method is fast and efficient for bulk functional gene sequences analysis. The results clearly show that we are at a relatively early stage in sampling the global richness of APB species. Periodical assessment will undoubtedly facilitate in-depth analysis of potential biogeographical distribution pattern of APB. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first attempt to assess the present understanding of APB diversity in natural environments. 1. Within mainstream ecological literature, functional structure has been viewed as resulting from the interplay of species interactions, resource levels and environmental variability. Classical models state that interspecific competition generates species segregation and guild formation in stable saturated environments, whereas opportunism causes species aggregation on abundant resources in variable unsaturated situations. 2. Nevertheless, intrinsic functional constraints may result in species-specific differences in resource-use capabilities. This could force some degree of functional structure without assuming other putative causes. However, the influence of such constraints has rarely been tested, and their relative contribution to observed patterns has not been quantified. 3. We used a multiple null-model approach to quantify the magnitude and direction (non-random aggregation or divergence) of the functional structure of a vertebrate predator assemblage exposed to variable prey abundance over an 18-year period. Observed trends were contrasted with predictions from null-models designed in an orthogonal fashion to account independently for the effects of functional constraints and opportunism. Subsequently, the unexplained variation was regressed against environmental variables to search for evidence of interspecific competition. 4. Overall, null-models accounting for functional constraints showed the best fit to the observed data, and suggested an effect of this factor in modulating predator opportunistic responses. However, regression models on residual variation indicated that such an effect was dependent on both total and relative abundance of principal (small mammals) and alternative (arthropods, birds, reptiles) prey categories. 5. In addition, no clear evidence for interspecific competition was found, but differential delays in predator functional responses could explain some of the unaccounted variation. Thus, we call for caution when interpreting empirical data in the context of classical models assuming synchronous responses of consumers to resource levels. 相似文献

Understanding patterns and processes involved in changes in soil and vegetation after agricultural abandonment is a key issue for management policies leading to land restoration and reclamation in Mediterranean semiarid environments. We selected a number of active and abandoned fields in two regions of olive groves in Catalonia (NE Spain), in order to analyze changes in certain soil properties and vegetation cover variables, as well as their interrelationships. The soil chemical properties considered, summarized into a single PCA axis, showed significant spatial (regional) differences and no temporal (successional) pattern, indicative of the strong influence of the parent rock material. The soil physical variables examined also showed significant regional variability, but such differences could be partially explained by successional changes. The variability at the landscape level and the observed successional trends for soil physical properties are interpreted as a result of both the influence of the bedrock and contrasting management practices prior to abandonment in each region. Although we did not find significant regional or successional variability in a global measure of plant cover, we did find significant and different patterns of variability for each of the main plant functional groups considered. Regional and post-agricultural variability in soil physical properties seem to be the prime factors determining the abundance of the main plant functional groups. 相似文献

Production of recombinant proteins at low temperatures is one strategy to prevent formation of protein aggregates and the use of an expensive inducer such as IPTG. We report on the construction of two expression vectors both containing the cold-inducible des promoter of Bacillus subtilis, where one allows intra- and the other extracellular synthesis of recombinant proteins.
