Factor F430 is a yellow compound of unknown structure present in methanogenic bacteria. It has recently been shown to contain nickel. In this communication the influence of the nickel concentration in the growth medium on the factor F430 content of Methanobacterium thermoautotrophicum and on the nickel content of factor F430 was studied. It was found: (1) The content of factor F430 in the cells was strongly dependent on the nickel concentration of the growth medium. Cells grown on media with 2.5 M NiCl2 contained 28 times as much factor F430 per g as those grown on media with 0.075 M NiCl2; (2) factor F430 was synthesized in nickel deprived cells only upon the addition of nickel Nickel uptake paralleled factor F430 synthesis; (3) independent of the nickel concentration in the growth medium, the extinction coefficient at 430 nm of factor F430 per mol nickel was always near 22,500 cm-1 (mol Ni)-1. Summary The neuronal pathways connecting the stomatogastric nervous system with the retrocerebral complex of the cockroach, Periplaneta americana, were investigated by means of axonal cobalt chloride iontophoresis. Somata in the hypocerebral ganglion and in the nervus recurrens sending their axons to different parts of the stomatogastric nervous system were traced. Some axons in the oesophageal nerve arise from large perikarya in the anterior part of the pars intercerebralis and pass via the NCCI to the corpora cardiaca and the oesophageal nerve. They form a profuse dendritic tree in the protocerebrum. Fibers of the NCC I and NCC II as well as the NCA I and NCA II enter the stomatogastric nervous system via the hypocerebral ganglion.  相似文献

Cerebral contusion, cortical laceration, intracerebral hematoma formation, and hemorrhagic cortical infarction cause extravasation of red blood cells, followed by hemolysis, decompartmentalization of iron, formation and deposition of hemosiderin, and an increased incidence of epilepsy. In this experiment, 10 microliter of an aqueous solution containing 100 mmol/L FeCl2, 100 mmol/L CoCl2, or 0.9% (wt/vol) NaCl were injected at a depth of 1.8 mm into rat isocortex. The rate of formation of fluorescent compounds was measured in chloroform-methanol extracts of isocortical homogenates. Significant increases in the quantity of fluorescent products of lipid peroxidation were found 120 min after the injection of 100 mmol/L FeCl2. Cobaltous chloride and saline injection had no effect on the levels of fluorescent products found in the cortical homogenates. Although the intracortical deposition of aqueous solutions containing CoCl2 or FeCl2 in rodent cortex causes acute epileptiform discharges, the epileptogenic effect of CoCl2 is transient, while the injection of iron salts causes persistent seizures. Since CoCl2 injection failed to cause formation of lipid peroxidation products while the isocortical injection of iron caused significant increase in fluorescence within the injected hemisphere, we suggest that the occurrence of iron-induced lipid peroxidation may be of importance in initiation of recurrent seizures in the rat.  相似文献

Summary The role of copper in bovine serum amine oxidase was investigated by studying the effect of copper-binding inhibitors on the reactions of the pyrroloquinoline quinone carbonyl and on the reaction with oxygen. Hydrazines and hydrazides were used as carbonyl reagents and one of the hydrazines, benzylhydrazine, which was found to behave as a pseudo-substrate, was used to probe the reaction with oxygen. The presence ofN,N-diethyldithiocarbamate, a chelator that binds copper irreversibly, did not prevent the reactions at the carbonyl, but slowed down their rate and modified the conformation of the adducts. The same happened to the reaction with oxygen, which was slowed down but not abolished. Copper, which was never seen in the reduced state, thus appears to control all reactions without being directly involved in the binding of either hydrazines or oxygen. The enzyme functionality was in fact preserved upon substitution of copper with cobalt. The specific activity of the cobalt-substituted enzyme was only reduced to about 40% the native amine oxidase value. A chitosan (CS) immobilized cobalt complex (CS–CoSalen) was prepared by immersing a CS film into the saturated aqueous solution of a cobalt(II) complex of N,N′-bis(salicylaldehyde)ethylenediimino (CoSalen). The CS–CoSalen was characterized, and it was found that CoSalen was immobilized onto CS through the coordination of the amino group of CS. The cobalt(II) ion in the CS–CoSalen has a five-coordination structure. Its vacant sixth coordination site was found to bind molecular oxygen reversibly. Electrochemical studies demonstrated that the CS–CoSalen is active for the catalysis of the electrochemical reduction of oxygen to hydrogen peroxide in an aqueous medium.  相似文献

The reaction of Cp*2NbBH4 (Cp* = η5-C5Me5) with Ru3(CO)12 gave a mixture of compounds, from which only [Cp*2Nb(CO)2]2[Ru6(CO)16C] (1) could be characterized by spectroscopic and crystallographic methods. 11B NMR spectroscopy proved that interstitial boron may be present in other Ru6 clusters, but these compounds did not crystallize. The reaction of Cp*2NbBH4 with Co2(CO)8 gave among others the salts [Cp*2Nb(CO)2]2[Co6(CO)15C] (4) and [Cp*2Nb(CO)2]3[Co13(CO)24C2] (5), which were examined by X-ray diffraction studies. The true nature of the interstitial atoms in 5 was deduced from electrochemical investigations, which reveal similar redox properties as for the already known [Co13(CO)24C2]3− anion.  相似文献

The specific insertion of a divalent metal ion into tetrapyrrole macrocycles is catalyzed by a group of enzymes called chelatases. Distortion of the tetrapyrrole has been proposed to be an important component of the mechanism of metallation. We present the structures of two different inhibitor complexes: (1) N-methylmesoporphyrin (N-MeMP) with the His183Ala variant of Bacillus subtilis ferrochelatase; (2) the wild-type form of the same enzyme with deuteroporphyrin IX 2,4-disulfonic acid dihydrochloride (dSDP). Analysis of the structures showed that only one N-MeMP isomer out of the eight possible was bound to the protein and it was different from the isomer that was earlier found to bind to the wild-type enzyme. A comparison of the distortion of this porphyrin with other porphyrin complexes of ferrochelatase and a catalytic antibody with ferrochelatase activity using normal-coordinate structural decomposition reveals that certain types of distortion are predominant in all these complexes. On the other hand, dSDP, which binds closer to the protein surface compared to N-MeMP, does not undergo any distortion upon binding to the protein, underscoring that the position of the porphyrin within the active site pocket is crucial for generating the distortion required for metal insertion. In addition, in contrast to the wild-type enzyme, Cu2+-soaking of the His183Ala variant complex did not show any traces of porphyrin metallation. Collectively, these results provide new insights into the role of the active site residues of ferrochelatase in controlling stereospecificity, distortion and metallation.  相似文献

Cobalt hydroxide nanoparticles were prepared onto a carbon ceramic electrode (CHN|CCE) using the cyclic voltammetry (CV) technique. The modified electrode was characterized by X-ray diffraction and scanning electron microscopy. The results showed that CHN with a single-layer structure was uniformly electrodeposited on the surface of CCE. The electrocatalytic activity of the modified electrode toward the oxidation of insulin was studied by CV. CHN|CCE was also used in a homemade flow injection analysis system for insulin determination. The limit of detection (signal/noise [S/N] = 3) and sensitivity were found to be 0.11 nM and 11.8 nA/nM, respectively. Moreover, the sensor was used for detection of insulin in human serum samples. This sensor showed attractive properties such as high stability, reproducibility, and high selectivity.  相似文献

Copper(II)-zinc(II) bimetallic imidazolate metal-organic framework compounds of composition CuaZnbIm2(a + b) (Im = C3H3N2), including Cu2ZnIm6 (1), were prepared in high yields from the metal oxides under mild aqueous conditions using a novel acid catalysis method. Mild acidic hydrothermal treatment of paramagnetic 1 (≥120 °C) gave diamagnetic Cu(I)-containing Cu2ZnIm4 (2) in high yield. The formation mechanism of 2 involves electron transfer from Im to Cu(II), with concomitant formation of the unusual cyclotriimidazole, C9H6N6. Air-stable 2, characterized by single-crystal X-ray diffraction, crystallized in the tetragonal space group , with a = b = 10.9623(3), c = 6.3231(4) Å, α = β = γ = 90°, V = 759.86(6) Å3, and Z = 1.  相似文献

Summary The axonal connections between the medulla terminalis ganglionic X-organ (MTGXO) and the sinus gland are traced by iontophoretic application of cobalt dye to the neurosecretory system in the eyestalks of the crayfish, Orconectes limosus. The MTGXO consists of about 15 large perikarya, forming a distinct subgroup of neurosecretory cells in the medulla terminalis and giving rise to a prominent fibre bundle.
