To understand relationships between protein sequence and stability, we often compare data from proteins that differ by the substitution of one amino acid. Frequently, an amino acid change causes the cooperative denaturation transitions to shift to lower temperatures, diminishing the signal from the native state. Here we show that apparent stability changes, i.e., the free energy of denaturation, deltaGD, can also be caused by a deficiency of points in the low temperature end of the transition. Von Bertalanffy growth parameters were estimated for silver hake Merluccius bilinearis within four regions of the United States continental shelf ecosystem of the northwest Atlantic Ocean during 1975–1980, 1982–1987 and 1988–1992. Differences in silver hake growth were found between the Middle Atlantic and southern Georges Bank regions ( P <0–01) over all three time periods. Growth also differed between northern Georges Bank and the Gulf of Maine ( P <0–01) during 1975–1980 and 1982–1987, and significant differences between southern Georges Bank and northern Georges Bank were found only during 1982–1987. Silver hake in the Gulf of Maine exhibited larger asymptotic sizes, but attained their asymptotic size at slower rates (L00 = 47 cm and K =0–33) in comparison to silver hake in the Middle Atlantic ( L00 = 43 cm and K =0–51), while growth parameter estimates for silver hake on Georges Bank were intermediate between these. Although significant differences in growth of silver hake between the various regions are consistent with separate stocks in the U.S. continental shelf ecosystem, the growth characteristics from the different regions are inconsistent with the presently assigned stock boundaries. Schistosoma mansoni schistosomules prepared from cercariae by seven in vitro techniques had not all reached the same state of development at the end of the incubation period as scored by seven parameters: water tolerance; Cercarienhüllen Reaktion; presence of the glycocalyx; condition of the surface membrane; nuclear state; granule migration; and cryopreservability. At the end of the specific incubation period for each technique, the level of development was judged with respect to schistosomules which had developed in situ for 1 hr after penetration of the ear skin of mice. In descending order of their correspondence to in vivo schistosomules, those derived in vitro (by the procedures listed) ranked as follows: first, penetration of dried rat skin; second, centrifuging and vortexing, or incubation in serum-supplemented medium; and third, syringe passage, omnimixing, centrifuging, and incubating, or incubating alone. The only treatment common to all techniques was incubation in 37 C culture medium for 2 hr or more. This is suggested as the stimulus for the cercaria-to-schistosomule transformation. 相似文献

Numerous behavioural and morphological markers were found during the last instar of Trichoplusia ni which permit selection of highly synchronous groups of larvae for physiological or biochemical experiments. Growth parameters were also examined and it was found that the occurrence of a 6th instar was associated with a head-capsule width below the critical threshold of 1.66 mm. Starvation experiments indicated that the critical body weight triggering the first release of prothoracicotropic hormone (PTTH) was not the same for all larvae and was associated closely with a critical ratio of body size: head-capsule size and/or body size: initial body size at the beginning of the instar. The time of prothoracicotropic hormone-ecdysone release in preultimate instars was also associated with a similarly calculated ratio. The ratio was very similar from instar to instar. Neck or thoracic-abdominal ligation of larvae attaining various markers provided indications of times of release of critical amounts of prothoracicotropic hormone, ecdysone and juvenile hormone. The time of peak juvenile-hormone-esterase (JHE) activity in the haemolymph during the prepupal stage was determined with these markers. 相似文献

An investigation into the probable secondary structure of the myelin basic protein was carried out by the application of three procedures currently in use to predict the secondary structures of proteins from knowledge of their amino acid sequences. In order to increase the accuracy of the predictions, the amino acid substitutions that occur in the basic protein from different species were incorporated into the predictive algorithms. It was possible to locate regions of probable alpha-helix, beta-structure, beta-turn, and unordered conformation (coil) in the protein. One of the predictive methods introduces a bias into the algorithm to maximize or minimize the amounts of alpha-helix and/or beta-structure present; this made it possible to assess how conditions such as pH and protein concentration or the presence of anionic amphiphilic molecules could influence the protein's secondary structure. The predictions made by the three methods were in reasonably good agreement with one another. They were consistent with experimental data, provided that the stabilizing or destabilizing effects of the environment were taken into account. According to the predictions, the extent of possible alpha-helix and beta-structure formation in the protein s severely restricted by the low frequency and extensive scattering of hydrophobic residues, along with a high frequency and extensive scattering of residues that favor the formation of beta-turns and coils. Neither prolyl residues nor cationic residues per se are responsible for the low content of alpha-helix predicted in the protein. The principal ordered conformation predicted is the beta-turn. Many of the predicted beta-turns overlap extensively, involving in some cases up to 10 residues. In some of these structures it is possible for the peptide backbone to oscillate in a sinusoidal manner, generating a flat, pleated sheetlike structure. Cationic residues located in these structures would appear to be ideally oriented for interaction with lipid phosphate groups located at the cytoplasmic surface of the myelin membrane. An analysis of possible and probable conformations that the triproline sequence could assume questions the popular notion that this sequence produces a hairpin turn in the basic protein. 相似文献

Snow or glacial algae are found on all continents, and most species are in the Chlamydomonadales (Chlorophyta) and Zygnematales (Streptophyta). Other algal groups include euglenoids, cryptomonads, chrysophytes, dinoflagellates, and cyanobacteria. They may live under extreme conditions of temperatures near 0°C, high irradiance levels in open exposures, low irradiance levels under tree canopies or deep in snow, acidic pH, low conductivity, and desiccation after snow melt. These primary producers may color snow green, golden-brown, red, pink, orange, or purple-grey, and they are part of communities that include other eukaryotes, bacteria, archaea, viruses, and fungi. They are an important component of the global biosphere and carbon and water cycles. Life cycles in the Chlamydomonas–Chloromonas–Chlainomonas complex include migration of flagellates in liquid water and formation of resistant cysts, many of which were identified previously as other algae. Species differentiation has been updated through the use of metagenomics, lipidomics, high-throughput sequencing (HTS), multi-gene analysis, and ITS. Secondary metabolites (astaxanthin in snow algae and purpurogallin in glacial algae) protect chloroplasts and nuclei from damaging PAR and UV, and ice binding proteins (IBPs) and polyunsaturated fatty acids (PUFAs) reduce cell damage in subfreezing temperatures. Molecular phylogenies reveal that snow algae in the Chlamydomonas–Chloromonas complex have invaded the snow habitat at least twice, and some species are polyphyletic. Snow and glacial algae reduce albedo, accelerate the melt of snowpacks and glaciers, and are used to monitor climate change. As a result of the genetic selection for prolificacy and the improvements in the environment and farms management, litter size has increased in the last few years so that energy requirements of the lactating sow are greater. In addition, selection for feed efficiency of growing pigs is also conducted in maternal lines, and this has led to a decrease in appetite and feed intake that is extended to the lactation period, so the females are not able to obtain the necessary energy and nutrients for milk production and they mobilize their energetic reserves. When this mobilization is excessive, reproductive and health problems occur which ends up in an early sow culling. In this context, it has been suggested to improve feed efficiency at lactation through genetic selection. The aim of this study is to know, in a Duroc population, the genetic determinism of sow feed efficiency during lactation and traits involved in its definition, as well as genetic and environmental associations between them. The studied traits are daily lactation feed intake (dLFI), daily sow weight balance (dSWB), backfat thickness balance (BFTB), daily litter weight gain (dLWG), sow residual feed intake (RFI) and sow restricted residual feed intake (RRFI) during lactation. Data corresponded to 851 parities from 581 Duroc sows. A Bayesian analysis was performed using Gibbs sampling. A four-trait repeatability animal model was implemented including the systematic factors of batch and parity order, the standardized covariates of sow weight (SWf) and litter weight (LWs) at farrowing for all traits and lactation length for BFTB. The posterior mean (posterior SD) of heritabilities were: 0.09 (0.03) for dLFI, 0.37 (0.07) for dSWB, 0.09 (0.03) for BFTB, 0.22 (0.05) for dLWG, 0.04 (0.02) for RFI and null for RRFI. The genetic correlation between dLFI and dSWB was high and positive (0.74 (0.11)) and null between dLFI and BFTB. Genetic correlation was favourable between RFI and dLFI and BFTB (0.71 (0.16) and −0.69 (0.18)), respectively. The other genetic correlations were not statistically different from zero. The phenotypic correlations were low and positive between dLFI and dSWB (0.27 (0.03), dSWB and BFTB (0.25 (0.04)), and between dLFI and dLWG (0.16 (0.03)). Therefore, in the population under study, the improvement of the lactation feed efficiency would be possible either using RFI, which would not have unfavourable correlated effects, or through an index including its component traits. 相似文献

Bees represent an important element for the preservation of plant biodiversity. During their activities, bees cover a large area around the hive and their products strictly reflect the distinctive traits of these environments. In honey, the floral diversity of the areas inspected by bees is revealed by their pollen spectra. The present work characterized 460 Italian unifloral honeys by melissopalynological and physicochemical analyses. Obtained data contributed to: confirm the botanical identity of the samples; determine the plant biodiversity revealed in each honey; differentiate the samples of the same monofloral typology in relation to their geographical origin; and identify peculiar floristic associations in the Italian regions. Moreover, gas chromatographic analysis was carried out on rare unifloral honey to study their sugar profiles. This work provided the scientific base to create a valid tool able to assign a territorial brand and an authenticity guarantee to the honey, protecting the consumers from adulterations. 相似文献
