Summary The influence of vesicular-arbuscular mycorrhizal symbiosis on cytokinin activity in Citrus jambhiri Lush, seedlings was investigated. C. jambhiri inoculated with cultures of Glomus caledonium (Nicol. and Gerd.), G. epigaeum (Dan. and Trappe), G. etunicatum (Becker and Gerd.), G. fasciculatum Thaxt. (Gerd, and Trappe) or G. mosseae (Nicol and Gerd.) was grown from seed for 105 days in a glasshouse. Cytokinin activity in roots and leaves of seedlings was analyzed using high-performance liquid chromatography, mass spectrometry and a bioassay. Seedling leaf tissue had greater cytokinin activity than root tissue. Zeatin, zeatin riboside, and their dihydro- and glucoside derivatives were isolated from leaves of 105-day-old seedlings inoculated with G. fasciculatum and G. mosseae. Cytokinin activity in roots and leaves was associated with differences in seedling total dry weight and vesicular-arbuscular mycorrhizal colonization. The ribose moiety and the saturated side chain apparently influence cytokinin transport and physiological activity in Citrus seedlings.  相似文献

Summary Salt-tolerant cell lines of citrus rootstock (Poncirus trifoliata cv Pomeroy) were selected by subculturing embryo-derived calli on media containing sublethal concentrations of NaCl (5 and 10 g/l). Selected lines showed a normal growth in the presence of salt at the concentrations used for selection, and salt tolerance persisted after a passage on a salt-free media. Their K+ and Ca2+ content remained higher than in control cells for increasing NaCl concentration in the medium, suggesting a modification of cell membrane permeability as the main cause of NaCl tolerance. Shoots and plants regenerated from selected cell lines showed improved growth and salt tolerance. Calli induced from these plants tolerated a salt concentration of 10 g/l, indicating the persistance of the selected trait.  相似文献

Sour orange trees have been grown from the seedling stage out-of-doors at Phoenix, Arizona, USA, in open-top enclosures with clear plastic walls for 3.5 years. For the last 3 years of this period, half of the trees have been continuously exposed to air enriched with CO2 to 300 μmol mol?1 above the ambient concentration. At 2-month intervals over the last 12 months, we have determined the fine-root biomass in the top 0.4 m of the soil profile beneath the trees. Results from both treatments define a single relationship between fine-root biomass and trunk cross-sectional area. The data also show the CO2-enriched trees to have approximately 2.3 times more fine-root biomass in this soil layer than the trees grown in ambient air.  相似文献

用汁液摩擦接种方法对柑桔碎叶病毒(Citrus tatter leaf virus,CTLV)进行了进一步的生物学鉴定.结果表明,该病毒除在豇豆上引起枯斑外,还侵染克里芙兰烟(Nicotianaclevilandii)产生系统斑驳和轻花叶,侵染昆诺藜(Chenopodium quinoa)和苋色藜(C.amaranticolor)引起系统坏死和花叶,侵染鸡冠花(Celosia cristata)引起局部环斑.这些草本寄主均可作为CTLV的指示植物.在发病的柑桔叶汁液中,测得CTLV的稀释限点为10~(-5).经汁液摩擦接种,可以将CTLV从克里芙兰烟传播到木本指示植物柑桔和枳橙上.感病植物材料的组织超微结构观察结果表明:CTLV感染柑桔、枳橙、克里芙兰烟和昆诺藜均使其叶肉薄壁细胞的叶绿体出现淀粉沉积、叶绿体片层结构解体和消失;病毒在受感染的植株叶脉韧皮部细胞中紧密聚集,形成病毒结晶体;这种结构也出现在叶肉薄壁细胞中.这是关于该病毒组织病变的首次报道.用直接负染方法从感染CTLV的柑桔(枳橙)、克里芙兰烟和昆诺藜病叶中均能检查到线状病毒粒子,用改良的Deriick’s免疫电镜方法和琼脂双扩散方法测定均显示该病毒…  相似文献

Abscisic acid is considered an apocarotenoid formed by cleavage of a C-40 precursor and subsequent oxidation of xanthoxin and abscisic aldehyde. Confirmation of this reaction sequence is still awaited, and might best be achieved using a cell-free system capable of both carotenoid and abscisic acid biosynthesis. An abscisic acid biosynthesizing cell-free system, prepared from flavedo of mature orange fruits, was used to demonstrate conversion of farnesyl pyrophosphate, geranylgeranyl pyrophosphate and all-trans-β-carotene into a range of β,β-xanthophylls, xanthoxin, xanthoxin acid, 1′,4′-trans-abscisic acid diol and abscisic acid. Identification of product carotenoids was achieved by high-performance liquid chromatography and on-line spectral analysis of individual components together with co-chromatography. Putative C-15 intermediates and product abscisic acid were identified by combined capillary gas chroma-tography-mass spectrometry. Kinetic studies revealed that β-carotene, formed from either famesyl pyrophosphate or geranylgeranyl pyrophosphate, reached a maximum within 30 min of initiation of the reaction. Thereafter, β-carotene levels declined exponentially. Catabolism of substrate β-carotene into xanthophylls, putative abscisic acid precursors and product abscisic acid was restricted to the all-trans-isomer. However, when a combination of all-trans- and 9-cis-β-carotene in the ratio 1:1 was used as substrate, formation of abscisic acid and related metabolites was enhanced. Biosyn-thetically prepared [14C]-all-trans-violaxanthin, [14C]-all-trans-neoxanthin and [14C]-9′-cis-neoxanthin were used as substrates to confirm the metabolic interrelationship between carotenoids and abscisic acid. The results are consistent with 9′-cis-neoxan-thin being the immediate carotenoid precursor to ABA, which is oxidatively cleaved to produce xanthoxin. Formation of abscisic aldehyde was not observed. Rather, xanthoxin appeared to be converted to abscisic acid via xanthoxin acid and 1′,4′-trans-abscisic acid diol. An alternative pathway for abscisic acid biosynthesis is therefore proposed.  相似文献

Little information is available on the variability of the dynamics of the actual and observed root respiration rate in relation to abiotic factors. In this study, we describe I) interactions between soil CO2 concentration, temperature, soil water content and root respiration, and II) the effect of short-term fluctuations of these three environmental factors on the relation between actual and observed root respiration rates. We designed an automated, open, gas-exchange system that allows continuous measurements on 12 chambers with intact roots in soil. By using three distinct chamber designs with each a different path for the air flow, we were able to measure root respiration over a 50-fold range of soil CO2 concentrations (400 to 25000 ppm) and to separate the effect of irrigation on observed vs. actual root respiration rate. All respiration measurements were made on one-year-old citrus seedlings in sterilized sandy soil with minimal organic material.Root respiration was strongly affected by diurnal fluctuations in temperature (Q10 = 2), which agrees well with the literature. In contrast to earlier findings for Douglas-fir (Qi et al., 1994), root respiration rates of citrus were not affected by soil CO2 concentrations (400 to 25000 ppm CO2; pH around 6). Soil CO2 was strongly affected by soil water content but not by respiration measurements, unless the air flow for root respiration measurements was directed through the soil. The latter method of measuring root respiration reduced soil CO2 concentration to that of incoming air. Irrigation caused a temporary reduction in CO2 diffusion, decreasing the observed respiration rates obtained by techniques that depended on diffusion. This apparent drop in respiration rate did not occur if the air flow was directed through the soil. Our dynamic data are used to indicate the optimal method of measuring root respiration in soil, in relation to the objectives and limitations of the experimental conditions.  相似文献

An endo-polygalacturonase (PG) (EC:3.2.1.15) with a pI of 9.4 and an Mr of 44,500 was purified to electrophoretic homogeneity from the gut of West Indies sugarcane rootstalk borer weevil (Diaprepes abbreviatus L.) larvae. Hydrolytic activity was maximal in 150 mM sodium acetate, pH 5.5, at 30°C. Kinetic determinations yielded an apparent Km of 3.68 mg polygalacturonic acid (PGA)/ml and a Vmax of 283 μmol galacturonic acid/min/mg protein for PGA. Enzymatic activity was inhibited by a polygalacturonase inhibitor protein from “Hamlin” orange flavedo. The purified protein does not appear to be glycosylated, and its N-terminal sequence showed no homology to any PG protein sequences in data banks.  相似文献

对四季桔胚轴直接出芽、愈伤组织诱导及植株再生能力的研究表明:上、下胚轴在含有BA的MT培养基上,均能直接出芽,但上胚轴出芽能力明显高于下胚轴,最高出芽率为96.9%。外植体在培养基上的接种方式,对出芽也有一定影响,上胚轴切段以形态学下端垂直插入,出芽率高,且在培养前期表现尤为突出。胚轴愈伤诱导的适宜培养基为MT+IBA0.5mg-1+BAgmg-1。愈伤组织分化时,上、下胚轴愈伤组织所要求的BA浓度分别为4mgL-1及2mgL-1。在根的诱导中,附加的生长素种类,不仅影响生根率的高低,而且影响上部茎叶的生长,其中以加入lmgL-1IBA的效果最好,生根率达84.4%。  相似文献
