People with reduced esterase D (ESD) activity are susceptible to many diseases. However, how to activate ESD is still unknown. To address the question, we identified that 4-chloro-2-(5-phenyl-1-(pyridin-2-yl)-4, 5-dihydro-1H-pyrazol-3-yl) phenol (FPD5) could be a good candidate activator for ESD activity. We found that FPD5 could increase ESD activity in a dose-dependent way. FPD5 bound directly to ESD at Lys180 rather than its ubiquitination site Lys213. Site-directed mutagenesis at the binding site or the ubiquitination site inhibited FPD5 action. FPD5 increased the level of ESD mono-ubiquitination and mutESD K213A completely inhibited this action. Our findings highlighted the activation mechanism of ESD via promoting the mono-ubiquitination of ESD.  相似文献

The present research project details synthesis of new hybrid methanofullerenes based on acetylene and triazole esters of malonic acid containing 5Z,9Z-dienoic acids and fullerene C60 under Bingel-Hirsch conditions, including study of the cytotoxic activity with respect to Jurkat, K562, U937 and HL60 tumor cell lines. Hybrid methanofullerenes containing acetylenic fragments, unlike triazole substituents, were found to exhibit higher cytotoxicity, but are characterized by lower selectivity of action in relation to healthy cells.  相似文献

Galactose-1-phosphate uridylyltransferase (uridine diphosphoglucose: α-d-galactose-1-phosphate uridylyltransferase, EC 2.7.7.12) has been purified 4000-fold from human placenta in four chromatographic steps using DEAE-cellulose, hydrocylapatite, ethyliminohexylagarose, and Sephacryl S-200. The specific activity of the homogeneous enzyme was 56 units/mg protein. The placental enzyme consists of two similar subunits, each of molecular weight about 48,000. The placental enzyme was similar to published results for the red cell enzyme (V. P. Williams, Arch. Biochem. Biophys., 1978, 191, 182–191) with respect to subunit molecular weight, electrophoretic migration, and immunological properties. The more purified fractions of the placental enzyme invariably contained a glycoprotein which was removed in the gel filtration step. After this glycoprotein was removed, the enzyme was very labile and only about 20% of the catalytic activity was recovered.  相似文献

This paper examines the relationship between protected and endangered riverine species (target species) and hydrodynamics in river-floodplain ecosystems, combining ecological and policy-legal aspects of biodiversity conservation in river management. The importance of different hydrodynamic conditions along a lateral gradient was quantified for various taxonomic groups. Our results show that (i) target species require ecotopes along the entire hydrodynamic gradient; (ii) different parts of the hydrodynamic gradient are important to different species, belonging to different taxonomic groups; (iii) in particular low-dynamic parts are important for many species and (iv) species differ in their specificity for hydrodynamic conditions. Many species of higher plants, fish and butterflies have a narrow range for hydrodynamics and many species of birds and mammals use ecotopes along the entire gradient. Even when focussing only on target species, the entire natural hydrodynamic gradient is important. This means that the riverine species assemblage as a whole can benefit from measures focussing on target species only. River reconstruction and management should aim at re-establishing the entire hydrodynamic gradient, increasing the spatial heterogeneity of hydrodynamic conditions.  相似文献

Eco-environmental vulnerability assessment is crucial for environmental and resource management. However, evaluation of eco-environmental vulnerability over large areas is a difficult and complex process because it is affected by many variables including hydro-meteorology, topography, land resources, and human activities. The Thua Thien – Hue Province and its largest river system, the Perfume River, are vital to the social-economic development of the north central coastal region of Vietnam, but there is no zoning system for environmental protection in this region. An assessment framework is proposed to evaluate the vulnerable eco-environment in association with 16 variables with 6 of them constructed from Landsat 8 satellite image products. The remaining variables were extracted from digital maps. Each variable was evaluated and spatially mapped with the aid of an analytical hierarchy process (AHP) and geographical information system (GIS). An eco-environmental vulnerability map is assorted into six vulnerability levels consisting of potential, slight, light, medium, heavy, and very heavy vulnerabilities, representing 14%, 27%, 17%, 26%, 13%, 3% of the study area, respectively. It is found that heavy and very heavy vulnerable areas appear mainly in the low and medium lands where social-economic activities have been developing rapidly. Tiny percentages of medium and heavy vulnerable levels occur in high land areas probably caused by agricultural practices in highlands, slash and burn cultivation and removal of natural forests with new plantation forests. Based on our results, three ecological zones requiring different development and protection solutions are proposed to restore local eco-environment toward sustainable development. The proposed integrated method of remote sensing (RS), GIS, and AHP to evaluate the eco-environmental vulnerability is useful for environmental protection and proper planning for land use and construction in the future.  相似文献

This study evaluated the gait stability, variability, and complexity of healthy young adults on inclined surfaces. A total of 49 individuals walked on a treadmill at their preferred speed for 4 min at inclinations of 6%, 8%, and 10% in upward (UP) and downward (DOWN) conditions, and in horizontal (0%) condition. Gait variability was assessed using average standard deviation trunk acceleration between strides (VAR), gait stability was assessed using margin of stability (MoS) and maximum Lyapunov exponent (λs), and gait complexity was assessed using sample entropy (SEn). Trunk variability (VAR) increased in the medial-lateral (ML), anterior-posterior, and vertical directions for all inclined conditions. The SEn values indicated that movement complexity decreased almost linearly from DOWN to UP conditions, reflecting changes in gait pattern with longer and slower steps as inclination increased. The DOWN conditions were associated with the highest variability and lowest stability in the MoS ML, but not in λs. Stability was lower in UP conditions, which exhibited the largest λs values. The overall results support the hypothesis that inclined surfaces decrease gait stability and alter gait variability, particularly in UP conditions.  相似文献

Three species of tilapia native to Zimbabwe, Oreochromis mossambicus, O. mortimeri and O. macrochir are not naturally sympatric, but because of transplantation they have been brought into sympatry. Allozymes of fish examined from Lakes Kariba, Kyle and Chivero showed evidence of introgressive hybridization. The data showed non-random mating populations in Lakes Kariba and Kyle. All three reservoirs displayed individuals with intermediate principal component scores and hybrid index scores indicating hybridization.  相似文献
