目的:探讨高效液相色谱法(HPLC)测定小牛血去蛋白滴眼液中尼泊金乙酯含量的可行性。方法:利用Agilent 1100 Series高效液相色谱仪,在Diamonsil C18色谱柱(250 mm×4.6 mm,5μm),甲醇-0.05%乙酸水溶液(65:35)作为流动相,1.0 m L·min~(-1)流速,和30℃柱温下,利用254 nm检测波长对小牛血去蛋白滴眼液中尼泊金乙酯进行定量分析。结果:尼泊金乙酯进样量在0.29~1.43μg之间呈现良好的线性关系(r=0.9999),平均加样回收率为99.80%,RSD为0.91%。结论:HPLC方法简单快速,分离效果好,可用于小牛血去蛋白滴眼液中尼泊金乙酯的定量测定。  相似文献

The effectiveness of Zn at moderating the pro-oxidant effects of Cu was evaluated in two rat models that differed in the route and mode of administration. The endpoints investigated included measurement of the concentrations of Cu, Zn, metallothionein and glutathione concentrations, as well as SOD and catalase activity, in liver, kidneys and intestine. In a sub-chronic animal model, the hepatic accumulation of Cu was achieved by administration of dietary Cu (1.8 g/kg solid diet) for 30 days after which oral Zn (6g/kg solid diet) was given. Cu treatment induced an increase in the hepatic and intestinal concentration of Cu of 66 and 455%, respectively, that was not associated with synthesis of metallothionein synthesis, but rather appeared to be related to the higher activity of SOD. Subsequent administration with Zn after dietary Cu induced an increase in the hepatic and intestinal metallothionein content of more twice and reduced the Cu content to control values. Thus, Zn could act as both a competitor for absorption on the luminal side of the intestinal epithelium inducing the synthesis of metallothionein. In the second animal model, we studied the effects of interaction between Cu and Zn administered by i.p. injection at the dose of 3 and 10mg/kg, respectively; Zn was administered subsequent to Cu overload. In this case, when Zn was administered, Cu was already deposited in tissues and thus there is no competition between two metals at the level of membrane transport. In this experimental model treatment with Cu alone induced liver metallothionein synthesis, and the subsequent treatment with Zn did not decrease the hepatic content of Cu. One explanation for these observations is that Zn induces the synthesis of metallothionein, which binds Cu for which it has a higher affinity. Moreover, after treatment with Zn, SOD activity in the liver decreases of almost 30% with respect to treatment with alone Cu, suggesting that Zn has a protective effect.  相似文献

从灰树花(Maitake)子实体中提取分离得到子实体多糖D组分,并分别考察了提取中的几个关键因素对D组分得率的影响,包括加水倍数、浸提时间、醇沉浓度、浓缩倍数。然后,对D组分进行了硫酸化修饰,得到了两种不同取代度的硫酸化多糖。最后,研究了灰树花多糖D组分及硫酸化灰树花多糖D组分对小鼠的免疫器官重量、腹腔巨噬细胞吞噬功能、血清溶血素形成及淋巴细胞转化率的影响。结果表明,在10mgkg剂量下,灰树花多糖D组分及硫酸化灰树花多糖D组分具有免疫增强作用,并且在合适的取代度下硫酸化灰树花多糖D组分比灰树花多糖D组分表现了更强的免疫调节作用。  相似文献

The patatin-like phospholipase domain containing 3 (PNPLA3, also called adiponutrin, ADPN) is a membrane-bound protein highly expressed in the liver. The genetic variant I148M (rs738409) was found to be associated with progression of chronic liver disease. We aimed to establish a protein purification protocol in a yeast system (Pichia pastoris) and to examine the human PNPLA3 enzymatic activity, substrate specificity and the I148M mutation effect. hPNPLA3 148I wild type and 148M mutant cDNA were cloned into P. pastoris expression vectors. Yeast cells were grown in 3 L fermentors. PNPLA3 protein was purified from membrane fractions by Ni-affinity chromatography. Enzymatic activity was assessed using radiolabeled substrates. Both 148I wild type and 148M mutant proteins are localized to the membrane. The wild type protein shows a predominant lipase activity with mild lysophosphatidic acid acyl transferase activity (LPAAT) and the I148M mutation results in a loss of function of both these activities. Our data show that PNPLA3 has a predominant lipase activity and I148M mutation results in a loss of function.  相似文献

Antibacterial and antifungal activity of crude extract, alcoholic extract and extracted phenol from various parts of tropical pteridophyta, Hemionitis arifolia were tested by agar diffusion and tube dilution assay. Both the crude and alcoholic extracts of vegetative and reproductive leaves of H. arifolia showed considerable antibacterial activity against Gram negative test strain of Escherichia coli (MTCC-739). Extract from reproductive leaves also showed moderate antibacterial activity against Bacillus subtilis (MTCC-441) (Gram positive test strain) but didn’t show any antifungal activity against Candida albicans (MTCC-7353). Mycorrhizal and other symbiotic association with the root system of H. arifolia was studied and it is revealed that a number of mycorrhizal strains were present in both vegetative and reproductive form. Presence of Dark Septate Endophytic Fungi (DSF) was also detected.  相似文献

Alternaria represents the most common decay organism of the post-harvest tomato fruit. The prevalent type of decay, black rot lesion, is caused byA. alternata which may invade tomato tissue damaged by sun scald.Aspergillus niger, A. flavus andRhizopus stolonifer come in the second count level and occupy high to moderate occurrence. The mainly natural mycotoxins produced in rotted tomato are alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TA). Altertoxin I & II (AT-I & AT-II), in addition to AOH, AME and TA were produced by localA. alternata in a synthetic medium. The optimum temperature for toxin production byA. alternata IMI 89344 was 28 °C for AOH and AME, 21 °C for TA, and 14 °C for AT-I and AT-II. The growth and toxin were produced in a noticeable amount at 7 °C but drop at 35 °C. Significant inhibition in these toxins was attained at 500 ppm cinnamon oil in YES-Czapeks medium and in a tomato homogenate.  相似文献

The premise of the pharmacology of natural product is to explore benefits of natural resources for the mankind. Medicines extracted from natural resources are considered as primary source for drug discovery. Thus, the current study was designed to evaluate the safety profile and explore the analgesic and anti-inflammatory activity of ethanol extract of Cucurbita maxima (C. maxima) and Cucumis sativus (C. sativus) seeds. These seeds are edible, good in taste and have been used for several therapeutic purposes. Acute toxicity of the seeds was evaluated by Lorke’s method while Eddy’s hot plate and tail immersion methods were used to assess analgesic activity in mice. Anti-inflammatory activity was evaluated by rat hind paw edema method. The seed extracts of C. maxima and C. sativus were found to be safe and showed significant analgesic and anti-inflammatory activity in comparison with the control group. The therapeutic effects of these extracts were almost comparable to aspirin and brufen. Therefore, the seeds can be used as effective analgesic and anti-inflammatory agents.  相似文献

莲心是具有多种保健功能的天然产物,很有研究和开发的价值。本义对利用莲心生产保健饮料进行了研究。采用煎煮后捣碎离心、过滤的方法提取的莲心汁在浓度、色泽及稳定性方面综合效果最佳。在煎煮过程中添加适量的柠檬酸及异抗坏血酸钠后,能较有效地控制所得莲心汁的褐变速度,达到护色目的。加入PVPP或聚酰胺可增加莲心汁的稳定性。用提取的莲心汁可与其它不同的组分配成口感和风味各异的莲心饮料。  相似文献

Diabetes mellitus (DM) is a metabolic condition characterized by high blood sugar levels with serious system complications. Ginger (Zingiber officinale) and Cinnamon (Cinnamomum zeylanicum) have anti-diabetic activities. The goal of this study is to evaluate the possible protective and therapeutic effects of ginger and Cinnamon against histological, Ki67 Immunohistochemistry (IHC) and biochemical studies in testis and coda epididymis of Streptozotocin (STZ) induced diabetic rats. The experimental rats were divided into six groups: G1 was the control, G2 induced diabetic without treatment, G3 was treated with ginger before induction of DM (ginger protective), G4 were given ginger after DM induction (ginger therapeutic), G5 were given cinnamon before induction of DM (cinnamon protective) and G6 were given cinnamon after DM induction (cinnamon therapeutic). In diabetic rats’ significant increases in fasting blood sugar and body weight were observed after three weeks. Ginger and cinnamon effectively decreased serum glucose levels. Histopathological evaluations of seminiferous tubules and coda epididymis sections from diabetic rats showed severe damage to them. Furthermore, the sections of seminiferous tubules and coda epididymis rats administered ginger and cinnamon extract showed normal structure, healthy lining epithelium and sperm contents compared to diabetic rats. The results of the study show that both Ginger and Cinnamon aqueous extracts are effective as both hypoglycemic natural supplements that can protect against diabetic-induced testicular damage as well as share in the reservation of the cauda epididymal structure and sperm contents.  相似文献
