In previous studies on the quaternary structure of Na+,K+-ATPase, cupric-phenanthroline complex (CP) has been used for the cross-linking of the enzyme subunits. Here we show that the same products obtained in the presence of CP (α,α-dimer, α,β-dimer, and products of higher molecular weight) are also obtained when the enzyme is exposed to Cu2+ without o-phenanthroline. The α,β-dimer (but not the α,α-dimer) is dissociated in the presence of EDTA; indicating that this product is not the result of the covalent cross-linking of the subunits through a disulfide bond. The nature of the α,α-dimer remains to be determined. The findings suggest that the results of “cross-linking” experiments with CP should be interpreted with caution until the products are more clearly identified. 相似文献

An inhibitor of adenosylhomocysteine hydrolase, 3-deazaadenosine, caused profound inhibition of phagocytosis of opsonized erythrocytes by mouse resident peritoneal macrophages . The inhibition was evident at concentrations as low as 2×10?7M, and increased with increasing concentration and time of exposure to the analogue. It was not associated with detachment of the macrophage monolayers or with loss of cell viability. Although the inhibition was not reversible, progression of the functional impairment was interrupted by washing out the analogue. The urinary excretion of 3-methylhistidine (3-MH) was used as an index of muscle protein catabolism in primary and secondary infections of the guinea pig with Trichostrongylus colubriformis and in uninfected animals fed quantitatively reduced rations. Catabolism, which was depressed in all three groups, was directly related to a fall in food consumption. Possible explanations for the greater depression of catabolism in the primary infection than in the uninfected guinea pigs and its fall in the secondary infection in spite of little change in consumption are briefly discussed. It was concluded that the faster rate of whole-body protein turnover reported earlier in this series on protein metabolism in intestinal nematode infection was not partly due to a faster rate of muscle protein catabolism. [3H] -Concanavalin A binding to brain particulate preparations measured by a filtration technique was found to show a characteristic regional specificity with the caudate-putamen area exhibiting the greatest density of concanavalin A (con A) binding sites. The synaptic membranes were shown to be the most highly enriched of the subcellular fractions examined in terms of lectin-binding glycoproteins. Con A was also shown to inhibit the basal adenylate cyclase activity of cerebral, cerebellar, and caudate-putamen particulate preparations in a concentration-dependent manner. This lectin sensitivity of the adenylate cyclase is apparently an intrinsic property of the enzyme complex since a detergent dispersed preparation of the cerebral cortical enzyme was equally inhibited by con A. It is proposed that one of the membrane con A binding sites in brain tissue is a component of the adenylate cyclase system. 相似文献

Human fibroblast cell lines were pulse-treated for 1 h with either methylnitrosourea (MNU) or ethylnitrosourea (ENU) at various time intervals before harvesting for chromosome analysis. Cells treated with 1 X 10(-3) M, 5 X 10(-4) M, and 1 X 10(-4) M final concentrations of MNU and ENU during the G2 or M phases of the cell cycle showed a significant increase in chromatid-type abnormalities over controls. Cells exposed to MNU or ENU 23 h before harvest showed some chromosome-type abnormalities, reflecting probable damage induced during the G1 phase of the cell cycle or derived from chromatid damage induced during the previous cell cycle. The mitotic indices and incidences of abnormalities suggested a dose response effect when cells were treated with the two higher concentrations and the three concentrations, respectively, of MNU or ENU. Chromatid abnormalities were observed in MUN and ENU-treated cells from each of four cell lines. From this investigation, it was concluded that MNU and ENU treatment of human diploid cell lines in vitro induced both chromatid and chromosome aberrations. MNU and ENU, both of which had previously been shown to be mutagenic in experimental animals, are, therefore, also considered to be mutagenic at the chromosome level in human fibroblasts grown and treated in cell culture. 相似文献
