报告迷宫漏斗蛛的染色体数目、形态结构和性染色体组成。结果表明,迷宫漏斗蛛体细胞染色体数目是:雄性2n=42,雌性2n=44,性别决定机制属X_1X_2O型。X染色体是全部染色体中最长的和最短的2个(对)。染色体似乎均为端或亚端着丝粒染色体,这由C-显带标本分析所证实,没有亚中着丝粒染色体的证据。C-带标本分析表明,常染色体中,各条染色体的C-带纹,其大小和染色深浅无明显差异,但X_2染色体上有明显宽的C-带纹。C-显带处理没有得到稳定的有重复性的带纹。  相似文献

During conjugation, the micronucleus of Tetrahymena thermophila undergoes five consecutive nuclear divisions: meiosis, third prezygotic division (pregamic mitosis) and two postzygotic mitoses of the synkaryon. The four products of the synkaryon differentiate into macronuclear anlagen and new micronuclei and the old macronucleus is resorbed. The protein synthesis inhibitor cycloheximide, applied during conjugation, induced several developmental blocks. Pairs shifted to the drug during early meiotic prophase (stages I–III) were arrested at prophase. Cycloheximide applied to cells at pachytene (stages IV-VI) to metaphase arrested the conjugants at the stage of modified prometaphase/metaphase with overcondensed, swollen bivalents. In contrast to other systems, in the presence of cycloheximide, separation of chromatids, decondensation of chromosomes and exit from metaphase I were inhibited in both diploid and haploid cells. Pairs shifted to the drug after metaphase I were arrested at postmeiotic interphase after completing one nuclear cycle. The same rule applied to the subsequent cycle; then cells were arrested at the stage of pronuclei, and those pairs with functional pronuclei and synkarya were arrested at the stage of two products of the first postzygotic division (pronuclei were not arrested in nuclear transfer and karyogamy). Only pairs with two products of the first postzygotic division were arrested at the same stage after the cycloheximide treatment. Pairs shifted to cycloheximide during the second postzygotic division were arrested in development of macronuclear anlagen and resorption of old macronuclei. The postmeiotic conjugants pulse-treated with cycloheximide (2 h) yielded heterokaryons retaining parental macronuclei (i.e. they exhibited macronuclear retention).  相似文献

ES细胞(EmbryonicStemCells)是来源于小鼠早期胚胎的多潜能干细胞,它可以在体外大量培养。并以单细胞的形式注射到早期胚胎里,发育为嵌合体。到目前为止,通常使用的129小鼠品系是来源于近交系(inbred)小鼠的胚胎.与之相比,远交系小鼠应当具有较强的生命力和抗病能力。曾有人报道过建成了远交系小鼠胚胎干细胞系,但是尚没有见到获得嵌合鼠的报道。有人甚至认为:由于不同品系小鼠所具有的遗传背景不同,有的小鼠不能建成ES细胞系。最近,本实验室在这方面做了有益的探索,成功地建成了远交系小鼠胚胎干细胞系,并在这里报导首例用远交系小鼠胚胎干细胞系培育成功嵌合体小鼠。采用源于Swiss小鼠远交群的昆明(KM)品系小鼠囊胚建成了三个小鼠胚胎干细胞系(KE1.KE2.KE5)。核型正常率均达到70%以上。自第八代起分批冻存,复苏后,培养至第12代,消化成单细胞,通过囊胚显微注射,将其注射到615品系小鼠胚胎。在幸存的幼鼠中获得了一只来源于KE1细胞的嵌合鼠(Table1).其毛色表现为受体鼠(615)的白色中嵌合有供体鼠(KM)的黑褐色(PlateI-A).嵌合鼠与受体鼠的杂交后代鼠中仍然出现了受体鼠的毛色类型(  相似文献

Translocation frequencies (as compared to the standard chromosome arrangement typified by that in Chinese Spring) in 9 or more genotypes from each of 15 populations of Triticum dicoccoides in Israel were determined. Data also were obtained from 2 genotypes of the southernmost population (Jaba). A single population from Turkey was also investigated. There were 119 genotypes with translocations in the sample of 171 genotypes investigated (70%). The frequency of translocations in different populations varied from 0.27 to 1.00, and all populations had 1 or more genotypes with one or more translocations. Some populations such as Qazrin appeared to be homogeneous for translocations, but most populations were heterogeneous. A sample of 17 genotypes from 12 of the populations were crossed with the Langdon D-genome disomic substitutions to determine the identity of the chromosomes involved in the translocations. There were nine genotypes with translocations and with the exception of a 2A/2B translocation, none of them involved the same chromosomes. The B-genome chromosomes were involved in translocations more frequently than the A-genome chromosomes. Translocation frequencies (TF) of the various populations were correlated with environmental variables, primarily with water availability and humidity, and possibly also with soil type. In general, TF was higher in peripheral populations in the ecologically heterogeneous frontiers of species distribution than in the central populations located in the catchment area of the upper Jordan valley.  相似文献

Fusarium oxysporum f. sp. vasinfectum penetration hyphae infect living cells in the meristematic zone of cotton (Gossypium barbadense L.) roots. We characterized wall modifications induced by the fungus during infection of the protodermis using antibodies against callose, arabinogalactan-proteins, xyloglucan, pectin, polygalacturonic acid and rhamnogalacturonan I in high-pressure frozen, freeze-substituted root tissue. Using quantitative immunogold labelling we compared the cell walls before and after hyphal contact, cell plates with plasmodesmata during cytokinesis, and wall appositions induced by fungal contact. In the already-existing wall, fungal contact induced only minor modifications such as an increase of xyloglucan epitopes. Wall appositions mostly exhibited epitopes similar to the cell plate except that wall appositions had a much higher callose content. This study shows that wall appositions induced by Fusarium oxysporum hyphae are the result of normal cell wall synthesis and the addition of large amounts of callose. Summary Chromosome endoreduplication is a very common process in higher plants but its function and genetic control are still to be clarified. In our experiments we analyzed, by Feulgen cytophotometry, chromosome endoreduplication in endosperm cells of two maize genotypes, IHP and ILP, having high and low protein content in their seed, respectively. Chromosome endoreduplication occurs in both lines within 24 days after pollination, attaining a maximum ploidy level of 384C (7 DNA replication rounds) in IHP and of 192C (6 replication rounds) in ILP. In the mature seed, endosperms of the two lines show different mean ploidy level. In reciprocal crosses between IHP and ILP the f1 endosperms have mean ploidy levels analogous to that of the maternal parent, showing that the difference in ploidy level between the two genotypes is maintained. After selfing of the f1 plants, the difference in ploidy level between the two F2 populations is reduced. In F2 the mean ploidy level is as variable as in f1, indicating the absence of genetic segregation. From our data, it is apparent that both the genetic constitution (cytoplasmic and nuclear) of the maternal parent and the genotype of the individual endosperms influence the ploidy level. An analysis of the protein content in endosperms carried out on the same seed sample as analyzed cytophotometrically showed that the protein content increases, during seed development, parallel to chromosome endoreduplication and varies, in the two lines, in reciprocal crosses and their progeny, according to the same trend as mean ploidy level, suggesting a correlation between the two parameters.  相似文献

The kinetics of substitution reactions of [η-CpFe(CO)3]PF6 with PPh3 in the presence of R-PyOs have been studied. For all the R-PyOs (R = 4-OMe, 4-Me, 3,4-(CH)4, 4-Ph, 3-Me, 2,3-(CH)4, 2,6-Me2, 2-Me), the reactions yeild the same product [η5-CpFe(CO)2PPh3]PF6, according to a second-order rate law that is first order in concentrations of [η5-CpFe(CO)3]PF6 and of R-PyO but zero order in PPh3 concentration. These results, along with the dependence of the reaction rate on the nature of R-PyO, are consistent with an associative mechanism. Activation parameters further support the bimmolecular nature of the reactions: ΔH = 13.4 ± 0.4 kcal mol−1, ΔS = −19.1 ± 1.3 cal k−1 mol−1 for 4-PhPyO; ΔH = 12.3 ± 0.3 kcal mol−1, ΔS = 24.7 ±1.0 cal K−1 mol−1 for 2-MePyO. For the various substituted pyridine N-oxides studied in this paper, the rates of reaction increase with the increasing electron-donating abilities of the substituents on the pyridine ring or N-oxide basicities, but decrease with increasing 17O chemical shifts of the N-oxides. Electronic and steric factors contributing to the reactivity of pyridine N-oxides have been quantitatively assessed.  相似文献

The nearly neutral theory of molecular evolution predicts larger generation-time effects for synonymous than for nonsynonymous substitutions. This prediction is tested using the sequences of 49 single-copy genes by calculating the average and variance of synonymous and nonsynonymous substitutions in mammalian star phylogenies (rodentia, artiodactyla, and primates). The average pattern of the 49 genes supports the prediction of the nearly neutral theory, with some notable exceptions.The nearly neutral theory also predicts that the variance of the evolutionary rate is larger than the value predicted by the completely neutral theory. This prediction is tested by examining the dispersion index (ratio of the variance to the mean), which is positively correlated with the average substitution number. After weighting by the lineage effects, this correlation almost disappears for nonsynonymous substitutions, but not quite so for synonymous substitutions. After weighting, the dispersion indices of both synonymous and nonsynonymous substitutions still exceed values expected under the simple Poisson process. The results indicate that both the systematic bias in evolutionary rate among the lineages and the episodic type of rate variation are contributing to the large variance. The former is more significant to synonymous substitutions than to nonsynonymous substitutions. Isochore evolution may be similar to synonymous substitutions. The rate and pattern found here are consistent with the nearly neutral theory, such that the relative contributions of drift and selection differ between the two types of substitutions. The results are also consistent with Gillespie's episodic selection theory.  相似文献
