Four prolyl endopeptidase inhibitors isolated from actinomycetes, named propeptin, SNA-8073-B, staurosporine, and enduracidin were classified into 3 groups on the basis of their inhibition potency against prolyl endopeptidase from a bacterium (Flavobacterium) and a mammal (human placenta). Staurosporine inhibited the enzyme from Flavobacterium more strongly than that from human placenta. Enduracidin inhibited the enzyme from human placenta more strongly than that from Flavobacterium. Propeptin and SNA-8073-B, both new compounds, inhibited the enzymes from both origins to the same extent.  相似文献

Zusammenfassung Ein Stamm von Streptomyces flaveolus, Tü 1240, bildet neben dem schon länger bekannten Tirandamycin A das Tirandamycin B, das sich durch eine zusätzliche Hydroxylgruppe von A unterscheidet. Beide Antibiotica besitzen ein ähnliches Wirkungsspektrum und offensichtlich gleichen Wirkunsmechanismus. Anhand der Daten aus der Massenspektrometric, der 13C-und 1H-NMR-Spektren läßt sich dem Tirandamycin B die Formel II zuordnen.

Streptomyces flaveolus, strain Tü 1240 produces besides Tirandamycin A, a hitherto unknown antibiotic, which is closely related to Tirandamycin A. The new antibiotic Tirandamycin B contains one additional hydroxylgroup. Both antibiotics exhibit a similar antimicrobial spectrum and they seem to have the same mechanism of action. According to the data obtained from mass spectrometry, 13C-and 1H-NMR spectra formula II could be deduced for Tirandamycin B.

Summary A total of 94 actinomycete strains were isolated from the marine sediments of a shrimp farm, 87.2% belonged to the genus Streptomyces, others were Micromonospora spp. Fifty-one percent of the actinomycete strains showed activity against the pathogenic Vibrio spp. strains. Thirty-eight percent of marine Streptomyces strains produced siderophores on chrome azurol S (CAS) agar plates. Seven strains of Streptomyces were found to produce siderophores and to inhibit the growth of Vibrio spp. in vitro. Two of them belonged to the Cinerogriseus group, the most frequently isolated group of Streptomyces. The results showed that streptomycetes could be a promising source for biocontrol agents in aquaculture.  相似文献

In liquid culture, filamentous organisms often grow in the form of pellets. Growth result in an increase in radius, whereas shear forces result in release of hyphal fragments which act as centers for further pellet growth and development. A previously published model for pellet growth of filamentous microorganisms has been examined and is found to be unstable for certain parameter values. This instability has been identified as being due to inaccuracies in estimating the numbers of fragments which seed the pellet population. A revised model has been formulated, based on similar premises, but adopting a finite element approach. This considers the population of pellets to be distributed in a range of size classes. Growth results in movement to classes of increasing pellet size, while fragments enter the smallest size class, from which they grow to form further pellets. The revised model is stable and predicts changes in the distribution of pellet sizes within a population growing in liquid batch culture. It considers pellet growth and death, with fragmentation providing new centers of growth within the pellet population, and predicts the effects of shear forces on pellet growth and size distribution. Predictions of pellet size distributions are tested using previously published data on the growth of fungal pellets and further predictions are generated which are suitable for experimental testing using cultures of filamentous fungi or actinomycetes. 【目的】研究药用植物南方红豆杉内生及根际土壤放线菌的多样性及其抑菌、抗肿瘤等重要生物活性并获得一些具有强抑制植物病原真菌以及抗肿瘤等重要生物活性的菌株。【方法】选择7种培养基从南方红豆杉及其根际土壤中分离放线菌,对链霉菌进行形态学分类,去重复后对其进行抑制植物病原真菌以及抗肿瘤活性的筛选并对高活性菌株进行初步鉴定。对部分菌株进行16S rRNA基因测序分析研究其多样性。【结果】研究共分离得到277株放线菌,经去重复后剩余111株放线菌,可归类到6个亚目、7个科、8个属。其中链霉菌可分为10个类群。生物活性研究结果显示:30.9%的菌株具有抑制植物病原真菌活性,其中6株放线菌对多种植物病原真菌显示了强的抑菌活性。分别有44.1%和33.3%的菌株对胃癌肿瘤细胞株SGC-7901和肺癌肿瘤细胞株NCI-H460的抑制率在40%以上。【结论】药用植物南方红豆杉及其根际土壤蕴含种类丰富的放线菌资源,具有良好的生物学活性。菌株KLBMP 2170具有显著的抑菌以及抗肿瘤活性,值得我们去进一步研究。  相似文献

水霉拮抗放线菌的分离、筛选与鉴定   总被引:1,自引:0,他引:1  
目的:以珍珠健康养殖水体底泥为材料分离放线菌,筛选对水产动物水霉病病原菌有抗菌活性的放线菌。方法:采用稀释涂布法,选用萘啶酮酸和放线菌酮双抗平板分离获得放线菌;以黄颡鱼和湘云鲫鱼卵水霉病原菌为靶标菌,采用琼脂块法测试所分离菌株抗水霉菌活性及其稳定性;对拮抗活性强的放线菌采用形态观察和16S r DNA序列分析进行分类鉴定。结果:从分离获得的27株放线菌中筛选出3株对水霉病原菌有拮抗活性的菌株QF1、DNC17和QHV2,其中QHV2抗菌活性与稳定性最好;形态学观察与16S r DNA序列分析结果表明QF1、DNC17和QHV2均属于链霉菌属(Streptomycete sp.),分别鉴定为Streptomyces diastatochromogenes、Streptomyces variabilis和Streptomyces collinus。结论:3株放线菌对水霉病原菌具有较好的拮抗活性,具有开发成抗水霉药物的潜在价值。  相似文献

为了从土壤环境中筛选产生抗菌活性物质的放线菌,采用琼脂扩散法对分离自湖南省高望界自然保护区森林土壤的169株放线菌进行抗菌活性检测,采用16S RNA基因序列分析法进行系统发育分析,采用常规方法进行生物学特性研究。结果表明,有47株受试菌株的发酵产物显示抗菌活性阳性(27.8%),其中6株具有较强抗菌活性。形态观察结果表明,这6株菌株均为典型的产丝产孢放线菌,在多数培养基上生长良好,气生菌丝和孢子丝丰富。系统发育分析表明,6株菌株均属于链霉菌属(Streptomyces),归为5个物种。菌株JSM 147612、JSM 147786、JSM 147831和JSM 147846分别与链霉菌属的已知物种S.violascens(序列相似性为99.93%)、S.malachitospinus(99.65%)、S.anulatus(99.72%)和S.aureus(99.38%)系统发育关系最为密切;菌株JSM 147823和JSM 147842之间的序列相似性为99.86%,应该属于同一物种,与它们系统发育关系最为密切的是S.albidoflavus(相似性分别为99.72%和99.58%)。  相似文献

Fifty-four carboxydotrophic actinomycetes isolated from soils and composts were compared through 119 unit characters with representative mesophilic and thermophilic streptomycetes. The data were examined using the Jaccard, pattern and simple matching coefficients and clustering achieved using the unweighted pair group method with arithmetic averages algorithm. Acceptable cophenetic correlation and test error values allowed confidence to be placed in the resultant numerical taxonomies. The carboxydotrophic actinomycetes, which were distinct from cluster-groups corresponding to the mesophilic and thermophilic streptomycetes, formed two major cluster-groups the members of which were examined for the presence of diagnostic chemical markers. All but two of the carboxydotrophic actinomycetes had a profile of chemical properties consistent with their assignment to the genusStreptomyces. Quantitative fatty acid data were examined using the SIMCA package and the two statistically significant groups obtained corresponded with the cluster-groups circumscribed in the numerical phenetic analysis. Members of the two groups were also distinguished on the basis of their phospholipid composition. The two strains that containedmeso- as opposed to LL-diaminopimelic acid in their peptidoglycan also showed a distinct chemotaxonomic profile. It was concluded that the carboxydotrophic actinomycetes form a novel and taxonomically diverse group.  相似文献
