Chinese hamster ovary (CHO) cells are the main platform for production of biotherapeutics in the biopharmaceutical industry. However, relatively little is known about the metabolism of CHO cells in cell culture. In this work, metabolism of CHO cells was studied at the growth phase and early stationary phase using isotopic tracers and mass spectrometry. CHO cells were grown in fed-batch culture over a period of six days. On days 2 and 4, [1,2-13C] glucose was introduced and the labeling of intracellular metabolites was measured by gas chromatography-mass spectrometry (GC–MS) at 6, 12 and 24 h following the introduction of tracer. Intracellular metabolic fluxes were quantified from measured extracellular rates and 13C-labeling dynamics of intracellular metabolites using non-stationary 13C-metabolic flux analysis (13C-MFA). The flux results revealed significant rewiring of intracellular metabolic fluxes in the transition from growth to non-growth, including changes in energy metabolism, redox metabolism, oxidative pentose phosphate pathway and anaplerosis. At the exponential phase, CHO cell metabolism was characterized by a high flux of glycolysis from glucose to lactate, anaplerosis from pyruvate to oxaloacetate and from glutamate to α-ketoglutarate, and cataplerosis though malic enzyme. At the stationary phase, the flux map was characterized by a reduced flux of glycolysis, net lactate uptake, oxidative pentose phosphate pathway flux, and reduced rate of anaplerosis. The fluxes of pyruvate dehydrogenase and TCA cycle were similar at the exponential and stationary phase. The results presented here provide a solid foundation for future studies of CHO cell metabolism for applications such as cell line development and medium optimization for high-titer production of recombinant proteins.  相似文献

Objective: The aims of this study were to investigate the body fat distribution pattern in prepubertal Chinese children and to investigate the relationship between central fat distribution and specific biomarkers of cardiovascular disease. Research Methods and Procedures: The study was conducted in an urban Mainland Chinese (Jinan, Shandong) sample of children using a cross‐sectional design. Pubertal status was determined by Tanner criteria. Measurements included weight, height, waist circumference, DXA measures of total body fat and trunk fat; fasting serum measures of glucose, insulin, triglyceride, cholesterol, high‐density lipoprotein‐cholesterol; and systolic and diastolic blood pressure. Multiple regression models were developed with the biomarkers of cardiovascular risk factor as the dependent variables, and adjustments were made for significant covariates, including sex, age, height, weight, waist circumference, total body fat, trunk fat, and interactions. Results: A total of 247 healthy prepubertal subjects were studied. After co‐varying for age, weight, height, and extremity fat (the sum of arm fat and leg fat), girls had greater trunk fat than boys (p < 0.0001, R2 for model = 0.95). Insulin and triglyceride were positively related to central fat measured by DXA‐trunk fat (p < 0.05) but not related to the waist circumference. In the blood pressure model, waist circumference was a significant predictor of both systolic blood pressure and diastolic blood pressure, while DXA‐trunk fat was associated with diastolic blood pressure only. Significant interactions between sex and trunk fat, and sex and total fat, were found in relation to diastolic blood pressure. Discussion: In prepubertal Chinese children, greater trunk fat was significantly associated with higher insulin and triglyceride in boys and girls and was associated with higher diastolic blood pressure in boys only.  相似文献

The common wheat line three-pistil (TP) is a valuable mutant for wheat breeding. The TP mutation has normal spike morphology; however, it only produces three pistils per floret. Therefore, it has potential to increase the grain number per spike. In order to determine the underlying molecular mechanism, an annealing control primer system was used to identify the different expressed genes in three-pistil mutation. Using 120 arbitrary ACP primers, we identified three differentially expressed genes in young spikes between two near-isogenic lines (i.e., Chuanmai 28 TP and Chinese Spring TP) and their recurrent parents. We tentatively designated the three differentially expressed genes as DETP-1, DETP-2, and DETP-3. DETP-1 showed similar function with maize cytoplasmic membrane protein, which is involved in cell division in bacteria. DETP-3 is homologous to maize endo-1, 4-beta-glucanase (EGases), which is associated with plant development, cell wall loosening, stem flowering, and root expansion. DETP-2 showed no significant hit with any sequence found in the database and translates unknown protein. These genes would likely play an important role in determining the three pistils trait in wheat.  相似文献

20 0 3年 3~ 4月 ,在甘肃省莲花山自然保护区 ,利用无线电遥测和直接观察 ,分析了斑尾榛鸡(Bonasasewerzowi)的栖息地选择特征。结果表明 ,斑尾榛鸡春季栖息地一般在东北坡向 ,并具有高大乔木、下层植被盖度较高、灌丛较丰富的特点 ,这与食物丰富度较高及环境隐蔽性较强有关。栖息地质量对于配对活动的成功与否有一定影响。分析表明 ,栖息地内 0 5~ 2 5m植被水平遮挡度、柳树数量、箭竹数量是影响斑尾榛鸡春季栖息地选择的关键因子。建议在对斑尾榛鸡栖息地采取保护措施时 ,不仅要保护原生乔木 ,还要加强对灌丛生境的保护。  相似文献

目的对中华眼镜蛇咬伤致局部组织损伤的各种治疗方法进行疗效优劣比较,找出最佳治疗方法。方法总结我院569例中华眼镜蛇咬伤致局部组织损伤的各种治疗方法。结果治疗方法从优到劣依次是:抗蛇毒血清局部注射-糜蛋白酶局部注射-蛇伤药酒外敷-坏死组织早期切除-局部烧灼法-局部组织切开冲洗。结论中华眼镜蛇伤致局部组织损伤的治疗方法应首选抗蛇毒血清局部注射和糜蛋白酶局部注射,其次选用蛇伤药酒外敷。  相似文献

应用荧光显微法和石蜡切片解剖法对‘晚大新高’梨授粉受精过程进行了系统观察研究。结果表明:‘晚大新高’梨自花授粉不结实;异花最佳授粉品种为‘黄花’,其次为‘翠冠’和‘丰水’。与选用‘黄花’为异花授粉品种相比,自花和异花的授粉受精过程存在明显差异,自花花粉在授粉后2h开始萌发,8h花粉管生长至离柱头约1/3处停止生长,顶端膨大呈球形,表现出自交不亲和性;异花花粉在授粉后1h开始部分萌发,8h花粉管生长至花柱中部,24h到达花柱基部并进入子房,48h进入胚囊,72h完成双受精过程。  相似文献

The aim of this work was to study the influence of storage in liquid nitrogen on the viability of seeds of the hybrid orchid Bratonia and further development of its protocorms in vitro. Seeds were frozen in ampoules by direct immersion in liquid nitrogen and stored in the cryobank for a month. The germination rates of cryopreserved and control (nonfrozen) seeds did not differ and remained as high as 100%. The protocorms derived were cultured on the agar-solidified Murashige and Skoog nutrient medium (MS), half-strength MS and Knop media and also in Morel liquid medium. During the first 45 days of culturing, protocorms derived from cryopreserved seeds grew faster than control protocorms on the MS and half-strength MS media but, at longer culturing (496 days), the size of control protocorms was significantly larger. After 639 days of culturing, there was no difference in the amount of perished, budding, and newly formed protocorms obtained from cryopreserved and control seeds, except half-strength MS medium where the number of budding protocorms in the case of cryopreserved seeds was a little greater than in the control treatment. After seed cryopreservation, the frequency of budding and newly formed protocorms was greater on the agarized MS and in liquid Morel media. Cryopreservation had little effect on the subsequent growth of protocorms in vitro. The preferable nutrient media for culturing the protocorms have been suggested.  相似文献

中医体质学以人为研究对象,为了研究不同体质构成特点、演变规律、影响因素、分类标准,从而指导疾病的预防、诊治、康复与养生。目前关于生理和病理状态下的中医体质研究日益受到关注,因为对其研究符合中医”治未病”的理念,而关于少数民族的中医体质类型研究开展较少,但对少数民族之间和汉族之间中医体质的比较研究及与生活习惯,生活环境的联系将能更好地说明先天因素和后天环境对中医体质形成的影响。本文就这一领域的研究现状进行总结,对少数民族中医体质的研究一方面可为少数民族人群的疾病防治提供依据,另一方面有助于中医体质形成机理的阐明。  相似文献

External ATP causes passive permeability change in several transformed cells, but not in untransformed cells. We studied the effect of external ATP on the passive permeability of CHO-K1 cells, a transformed clone of Chinese hamster ovary cells. Treatment of the cells with external ATP alone did not produce a permeability change, and this was observed only when a mitochondrial inhibitor, such as rotenone or oligomycin, was present together with ATP. These inhibitors reduced the concentration of intracellular ATP and a permeability change by external ATP was observed when intracellular ATP was decreased more than 70%. This requirement for permeability change of CHO-K1 cells was quite unique, since passive permeability change of other transformed cells so far tested was induced by ATP alone. Treatment of CHO-K1 cells with cyclic AMP analogues increased their sensitivity to external ATP about 2-fold. The roles of external and intracellular ATP in controlling passive permeability are discussed.  相似文献
